Lichong decoction reduces Matrix Metalloproteinases-2 expression but increases Tissue Inhibitors of Matrix Metalloproteinases-2 ex-pression in a rat model of uterine leiomyoma显示文摘OBJECTIVE:To study the effect of Lichong decoction(LD) on expression of matrix metalloproteinase-2(MMP-2) and metalloproteinase-2(TIMP-2) in a rat model of uterine leiomyoma(UL).METHODS:UL was induced in rats using exogenous estrogen and progesterone.LD was administered(p.o.) for 4 weeks, and mifepristone(RU-486)used as a control. To observe the effect of LD on the uterine coefficient and uterine transverse diameter,a radioimmunoassay method was used to detect serum levels of sex hormones.Light microscopic analyses of pathologic changes in the tissues of UL rats were evaluated.Expression of the proteins of matrix metalloproteinases(MMPs) and tissue inhibitors of metalloproteinases(TIMPs) in uterine tissues was assessed by immunohistochemical staining and western blotting.RESULTS:A UL model in rats was established successfully.LD reduced uterine weight, uterine coefficient, and uterine transverse diameter compared with untreated controls.LD reduced levels of estradiol,progesterone,follicle-stimulating hormone, and luteinizing hormone in our UL models.LD improved the pathologic condition of uterine muscle.Expression of MMP-2 protein decreased to varying extents in LD-treated groups, but TIMP-2 levels were enhanced.LD appears to reduce MMP-2 expression and increase TIMP-2 expression in UL tissue.CONCLUSION:These data suggest that the mechanism of action of LD on ULs may involve reduction of MMP-2 expression and increase in TIMP-2 expression in rats. | Wang Yasong Li Donghua Xu Xin Qian Ruiya Zhang Yalan Huang Yuhua Geng Jianguo Zou Xiaoli Han Hon-gjuan Zhang Wufang | 2016 | Journal of Traditional Chinese Medicine2016,36,4: | 12

A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 5 | Development and Evaluation of a Loop-mediated Isothermal Amplification(LAMP) Assay for Rapid Detection of Chinese Giant Salamander Ranavirus显示文摘Loop-mediated isothermal amplification(LAMP) is a novel nucleic acid diagnostic method that can amplify rapidly a target template under isothermal conditions. In this study, a LAMP assay for rapid detection of Chinese giant salamander ranavirus(CGSRV) was developed from culture isolates and clinical samples. The LAMP assay was developed by designing one set of four specific primers, targeting the major capsid protein(MCP) gene of CGSRV. Reaction time and temperature were optimal for 40 min at 62°C. The developed LAMP assay is specific and highly sensitive for CGSRV detection, the detection limit could reach about 5 copies of cloned viral genomic fragments. The sensitivity of the LAMP assay was about 1000 and 10-fold higher than that of both conventional and nested PCR, respectively. The LAMP amplification produces a typical ladder-like pattern of products on an agarose gel that can be visually inspected after addition of ethidium bromide. The LAMP assay was evaluated further with clinical samples, and the results indicated the suitability and simplicity of the test as a rapid diagnostic tool for the detection of CGSRV. | Yi GENG Xingxing LIU Yan ZHOU Kaiyu WANG Xi PENG Zhijun ZHONG Xiaoli HUANG Defang CHEN | 2015 | Asian Herpetological Research2015,6,1: | 3
Expression patterns and functional divergence of homologous genes accompanied by polyploidization in cotton(Gossypium hirsutum L.)显示文摘Naturally allotetraploid cotton has been widely used as an ideal model to investigate gene expression remodeling as a consequence of polyploidization. However, the global gene pattern variation during early fiber development was unknown. In this study, through RNA-seq technology,we comprehensively investigated the expression patterns of homologous genes between allotetraploid cotton(G.hirsutum)and its diploid progenitors(G.arboreum and G.raimondii)at the fiber early development stage. In tetraploid cotton, genes showed expression level dominance(ELD)bias toward the A genome. This phenomenon was explained by the up-/downregulation of the homologs from the nondominant progenitor(D genome).Gene ontology(GO)enrichment results indicated that the ELD-A genes might be a prominent cause responsible for fiber property change through regulating the fatty acid biosynthesis/metabolism and microtubule procession, and the ELD-D genes might be involved in transcription regulation and stress inducement. In addition, the number and proportion of completely A-and D-subfunctionalized gene were similar at different fiber development stages. However, for neofunctionalization, the number and proportion of reactivated D-derived genes were greater than those of A at 3 and 5 DPA.Eventually,we found that some homologous genes belonging to several specific pathways might create novel asymmetric transcripts between two subgenomes during polyploidization and domestication process, further making the fiber property meet the human demands. Our study identified determinate pathways and their involved genes between allotetraploid cotton and their progenitors at early fiber development stages, providing new insights into the mechanism of cotton fiber evolution. | Zhen Peng Hua Cheng Gaofei Sun Zhaoe Pan Xiao Wang Xiaoli Geng Shoupu He Xiongming Du | 2020 | Science China(Life Sciences)2020,63,10: | 2

QTL mapping of agronomic and economic traits for four F_(2)populations of upland cotton显示文摘Background:Upland cotton(Gossypium hirsutum)accounts for more than 90%of the annual world cotton output because of its high yield potential. However, yield and fiber quality traits often show negative correlations. We constructed four F_(2)populations of upland cotton, using two normal lines(4133B and SGK9708)with high yield potential but moderate fiber quality and two introgression lines(Suyuan04–3 and J02–247)with superior fiber quality, and used them to investigate the genetic basis underlying complex traits such as yield and fiber quality in upland cotton. We also phenotyped eight agronomic and economic traits and mapped quantitative trait loci(QTLs).Results:Extensive phenotype variations and transgressive segregation were found across the segregation populations. We constructed four genetic maps of 585.97 centiMorgan(cM),752.45 cM,752.45 cM,and 1163.66 cM,one for each of the four F_(2)populations.Fifty QTLs were identified across the four populations(7 for plant height,27 for fiber quality and 16 for yield). The same QTLs were identified in different populations, including qBW4 and qBW2,which were linked to a common simple sequence repeat(SSR)marker,NAU1255.A QTL cluster containing eight QTLs for six different traits was characterized on linkage group 9 of the 4133B×Suyuan04–3 population. Conclusions:These findings will provide insights into the genetic basis of simultaneous improvement of yield and fiber quality in upland cotton breeding. | LI Hongge PAN Zhaoe HE Shoupu JIA Yinhua GENG Xiaoli CHEN Baojun WANG Liru PANG Baoyin DU Xiongming | 2021 | Journal of Cotton Research2021,4,1: | 2
Comparative analysis of membranous and other nephropathy subtypes and establishment of a diagnostic model显示文摘This study aimed to compare clinical features between membranous nephropathy (MN) and nonmembranous nephropathy (non-MN), to explore the clinically difTerential diagnosis of these two types, and to establish a diagnostic model of MN. After renal biopsy was obtained, 798 patients were divided into two groups based on their examination results: primary MN group (n = 248) and non-MN group (n = 550). Their data were statistically analyzed. Logistic regression analysis indicated that anti-PLA2R antibodies, IgG, and Cr were independently correlated with MN, and these three parameters were then used to establish the MN diagnostic model. A receiver operating characteristic (ROC) curve confirmed that our diagnostic model could distinguish between patients with and without MN, and their corresponding sensitivity, specificity, and AUC were 79.9%, 89.4%, and 0.917, respectively. The cutoff value for this combination in MN diagnosis was 0.34. The established diagnostic model that combined multiple factors shows a potential for broad clinical applications in differentiating primary MN from other kidney diseases and provides reliable evidence supporting the feasibility of noninvasive diagnosis of kidney diseases. | Hanyu Zhu Bo Fu Yong Wang Jing Gao Qiuxia Han Wenjia Geng Xiaoli Yang Guangyan Cai Xiangmei Chen Dong Zhang | 2019 | Frontiers of Medicine2019,13,5: | 2
Pathological Changes in Andrias davidianus Infected with Chinese Giant Salamander Ranavirus显示文摘Chinese giant salamander ranavirus(CGSRV) is an emerging pathogen in captive populations of the Chinese giant salamander(Andrias davidianus). We processed 140 morbid Chinese giant salamanders from seven captive breeding populations over five years, and describe the disease associated with CGSRV infection. The most common gross signs were significant swelling of the legs and coelomic cavity, erythema of the legs and ventrum in juveniles; cutaneous erosions and ulcerations in adults, particularly the limbs and the head; and marked petechial or ecchymotic hemorrhages of the internal organs, particularly the liver, spleen and kidney. Histological examination showed degeneration, necrosis, and inflammation in many organs, particularly in the organs where hemorrhage was observed. There was evidence of eosinophilic inclusion bodies in degenerated and necrotic cells. We identified virus particles and empty capsids without viral nucleoid in the inclusion bodies using electron microscopy. Virus particles were hexagonal or round shape, and appeared in paracrystalline arrays, aggregates, or singly. All enveloped viral particles were 140–160 nm. Polymerase chain reaction followed by sequencing verified that the virus particles were CGSRV. These results collectively support that CGSRV was the etiologic agent responsible for these mass die-offs of the Chinese giant salamander. The pathology described herein will be useful in diagnosing cases of ranaviral disease caused by CGSRV, and provide evidence that this pathogen is a significant threat to the Chinese giant salamander. | Yi GENG Matthew James GRAY Kaiyu WANG Defang CHEN Ping OUYANG Xiaoli HUANG Changliang HE Zhijun ZHONG Zexiao YANG | 2016 | Asian Herpetological Research2016,7,4: | 2

The most common toxicities included rash (53.3%) and diarrhea (33.3%). Dehydration and pruritus of the skin developed in 26.7% and 22.2% of the patients, respectively. Hepatic toxicity occurred in 6.7% of patients and oral ulceration occurred in 4.4% of patients. Conclusion: Single agent treatment with gefitinib is effective against advanced NSCLC, and is well tolerated in Chinese patients. | Yongmei Yin Yiting Geng Xiaodong Li Xiaoli Hu Xiaofeng Chen Wei Li Yongqian Shu | 2009 | Journal of Nanjing Medical University2009,23,6: | 1 |
A Study of the Structural Evolution and Strength Damage Mechanisms of PishaSandstone Cement Soil Modified with Fly Ash显示文摘In the present study,in order to investigate the effects of fly ash on the structural evolution and strength damage mechanism of Pisha-sandstone cement soil, unconfined compressive strength tests of Pisha-sandstone cement soil with different fly ash content levels and various ages were carried out.The apparent morphology, microstructures, and chemical compositions of the samples were observed and analyzed using ultra-depth three-dimensional microscopy, scanning electron microscopy, and XRD methods. The results revealed that the unconfined compressive strength levels of Pisha-sandstone cement soil samples displayed increasing trends with the increases in fly ash content and age.For example, when the fly ash content levels were increased from 12%to 15%,the strength of Pisha-sandstone cement soil had only slightly increased under the curing ages of 7,28,and 60 days.In addition, the unconfined compressive strength levels of the samples with 15%fly ash content only increased 0.02%,0.51%,and 0.54%,respectively,when compared with the samples containing 12%fly ash.It was observed that with the increases in the fly ash content, the number of pores on the outer surfaces of the samples were significantly reduced. Also, the height differences of cross-sectional gullies were reduced, and the apparent morphology was observed to be flatter.Since cement hydration creates a strong alkaline environment for reaction systems, the active degrees of the pozzolanic reactions of the fly ash were stimulated in this study. Moreover,a significant amount of the C-S-H gel phase and the stable five-membered ring structure of the mordenite and ettringite were generated, which connected the loose Pisha-sandstone particles to form a skeleton. The internal microstructures were then observed to be denser and more uniform. At the same time, the micro-pores were filled and refined by the unreacted micro-bead fly ash.Consequently, the defects in the internal microstructures were improved. Also, based on the Weibull distribution,a damage evolution model of the Pisha-sandstone cement soil was established. The analysis results of the damage variable D values during the initial damage stage, damage evolution stage, and residual damage stage of the damage process showed that the damage variables during all three stages displayed decreasing trends with the increases in the fly ash content levels and age. Therefore, based on this study’s findings,it was considered that the incorporation of fly ash could effectively improve the damage degrees of Pisha-sandstone cement soil under external force conditions. | Jian Yang Xiaoli Li Hui Wang Kaiqiang Geng | 2021 | Journal of Renewable Materials2021,9,12: | 1
Genomic variation, origin tracing, and vaccine development of SARS-CoV-2:A systematic review显示文摘COVID-19 has spread globally to over 200 countries with more than 40 million confirmed cases and one million deaths as of November 1,2020.The SARS-CoV-2 virus, leading to COVID-19,shows extremely high rates of infectivity and replication, and can result in pneumonia, acute respiratory distress,or even mortality.SARS-CoV-2 has been found to continue to rapidly evolve, with several genomic variants emerging in different regions throughout the world. In addition, despite intensive study of the spike protein, its origin, and molecular mechanisms in mediating host invasion are still only partially resolved. Finally, the repertoire of drugs for COVID-19 treatment is still limited, with several candidates still under clinical trial and no effective therapeutic yet reported.Although vaccines based on either DNA/mRNA or protein have been deployed, their efficacy against emerging variants requires ongoing study, with multivalent vaccines supplanting the first-generation vaccines due to their low efficacy against new strains. Here,we provide a systematic review of studies on the epidemiology, immunological pathogenesis, molecular mechanisms, and structural biology,as well as approaches for drug or vaccine development for SARSCoV-2. | Tianbao Li Tao Huang Cheng Guo Ailan Wang Xiaoli Shi Xiaofei Mo Qingqing Lu Jing Sun Tingting Hui Geng Tian Leyi Wang Jialiang Yang | 2021 | The Innovation2021,2,2: | 1
IRE1α regulates the PTHrP-IHH feedback loop to orchestrate chondrocyte hypertrophy and cartilage mineralization显示文摘Cartilage development is controlled by the highly synergistic proliferation and differentiation of growth plate chondrocytes,in which the Indian hedgehog(IHH)and parathyroid hormone-related protein-parathyroid hormone-1 receptor(PTHrP-PTH1R)feedback loop is crucial. The inositol-requiring enzyme 1a/X-box-binding protein-1 spliced(IRE1α/XBP1s)branch of the unfolded protein response(UPR)is essential for normal cartilage development. However, the precise role of ER stress effector IRE1α,encoded by endoplasmic reticulum to nucleus signaling 1(ERN1),in skeletal development remains unknown.Herein,we reported that loss of IRE1α accelerates chondrocyte hypertrophy and promotes endochondral bone growth.ERN1 acts as a negative regulator of chondrocyte proliferation and differentiation in postnatal growth plates. Its deficiency interrupted PTHrP/PTH1R and IHH homeostasis leading to impaired chondrocyte hypertrophy and differentiation.XBP1s,produced by p-IRE1α-mediated splicing, binds and up-regulates PTH1R and IHH,which coordinate cartilage development.Meanwhile,ER stress cannot be activated normally in ERN1-deficient chondrocytes. In conclusion,ERN1 deficiency accelerates chondrocyte hypertrophy and cartilage mineralization by impairing the homeostasis of the IHH and PTHrP/PTH1R feedback loop and ER stress.ERN1 may have a potential role as a new target for cartilage growth and maturation. | Mengtian Fan Nana Geng Xingyue Li Danyang Yin Yuyou Yang Rong Jiang Cheng Chen Naibo Feng Li Liang Xiaoli Li Fengtao Luo Huabing Qi Qiaoyan Tan Yangli Xie Fengjin Guo | 2024 | Genes & Diseases2024,11,1: | 0

Progranulin regulation of autophagy contributes to its chondroprotective effect in osteoarthritis显示文摘Progranulin(PGRN)is a multifunctional growth factor involved in many physiolog-ical processes and disease states. The apparent protective role of PGRN and the importance of chondrocyte autophagic function in the progression of osteoarthritis(OA)led us to investi-gate the role of PGRN in the regulation of chondrocyte autophagy.PGRN knockout chondro-cytes exhibited a deficient autophagic response with limited induction following rapamycin, serum starvation, and IL-1b-induced autophagy.PGRN-mediated anabolism and suppression of IL-1b-induced catabolism were largely abrogated in the presence of the BafA1 autophagy inhibitor.Mechanistically, during the process of OA,PGRN and the ATG5eATG12 conjugate form a protein complex;PGRN regulates autophagy in chondrocytes and OA through,at least partially, the interactions between PGRN and the ATG5eATG12 conjugate.Furthermore, the ATG5eATG12 conjugate is critical for cell proliferation and apoptosis.Knockdown or knockout of ATG5 reduces the expression of ATG5eATG12 conjugate and inhibits the chondroprotective effect of PGRN on anabolism and catabolism.Overexpression of PGRN partially reversed this effect. In brief, the PGRN-mediated regulation of chondrocyte autophagy plays a key role in the chondroprotective role of PGRN in OA.Such studies provide new insights into the pathogen-esis of OA and PGRN-associated autophagy in chondrocyte homeostasis. | Yiming Pan Yuyou Yang Mengtian Fan Cheng Chen Rong Jiang Li Liang Menglin Xian Biao Kuang Nana Geng Naibo Feng Lin Deng Wei Zheng Fengmei Zhang Xiaoli Li Fengjin Guo | 2023 | Genes & Diseases2023,10,4: | 0
Gut microbiota dysbiosis contributes toα-synuclein-related pathology associated with C/EBPβ/AEP signaling activation in a mouse model of Parkinson’s disease显示文摘Parkinson’s disease is a neurodegenerative disease characterized by motor and gastrointestinal dysfunction.Gastrointestinal dysfunction can precede the onset of motor symptoms by several years.Gut microbiota dysbiosis is involved in the pathogenesis of Parkinson’s disease, whether it plays a causal role in motor dysfunction, and the mechanism underlying this potential effect, remain unknown.CCAAT/enhancer binding proteinβ/asparagine endopeptidase(C/EBPβ/AEP)signaling,activated by bacterial endotoxin, can promoteα-synuclein transcription, thereby contributing to Parkinson’s disease pathology. In this study,we aimed to investigate the role of the gut microbiota in C/EBPβ/AEP signaling,α-synuclein-related pathology, and motor symptoms using a rotenone-induced mouse model of Parkinson’s disease combined with antibiotic-induced microbiome depletion and fecal microbiota transplantation. We found that rotenone administration resulted in gut microbiota dysbiosis and perturbation of the intestinal barrier,as well as activation of the C/EBP/AEP pathway,α-synuclein aggregation, and tyrosine hydroxylase-positive neuron loss in the substantia nigra in mice with motor deficits. However, treatment with rotenone did not have any of these adverse effects in mice whose gut microbiota was depleted by pretreatment with antibiotics.Importantly,we found that transplanting gut microbiota derived from mice treated with rotenone induced motor deficits, intestinal inflammation, and endotoxemia.Transplantation of fecal microbiota from healthy control mice alleviated rotenone-induced motor deficits, intestinal inflammation, endotoxemia, and intestinal barrier impairment. These results highlight the vital role that gut microbiota dysbiosis plays in inducing motor deficits,C/EBPβ/AEP signaling activation,andα-synuclein-related pathology in a rotenone-induced mouse model of Parkinson’s disease.Additionally, our findings suggest that supplementing with healthy microbiota may be a safe and effective treatment that could help ameliorate the progression of motor deficits in patients with Parkinson’s disease. | Xiaoli Fang Sha Liu Bilal Muhammad Mingxuan Zheng Xing Ge Yan Xu Shu Kan Yang Zhang Yinghua Yu Kuiyang Zheng Deqin Geng Chun-Feng Liu | 2024 | Neural Regeneration Research2024,19,9: | 0

Genetic diversity and population structure of Gossypium arboreum L. collected in China显示文摘Background: Gossypium arboreum is a diploid species cultivated in the Old World. It possesses favorable characters that are valuable for developing superior cotton cultivars.Method: A set of 197 Gossypium arboreum accessions were genotyped using 80 genome-wide SSR markers to establish patterns of the genetic diversity and population structure. These accessions were collected from three major G. arboreum growing areas in China. A total of 255 alleles across 80 markers were identified in the genetic diversity analysis. Results: Three subgroups were found using the population structure analysis, corresponding to the Yangtze River Valley, North China, and Southwest China zones of G.arboreum growing areas in China. Average genetic distance and Polymorphic information content value of G. arboreum population were 0.34 and 0.47, respectively, indicating high genetic diversity in the G. arboreum germplasm pool. The Phylogenetic analysis results concurred with the subgroups identified by Structure analysis with a few exceptions. Variations among and within three groups were observed to be 13.61% and 86.39%, respectively. Conclusion: The information regarding genetic diversity and population structure from this study is useful for genetic and genomic analysis and systematic utilization of economically important traits in G. arboreum. | JIA Yinhua PAN Zhaoe HE Shoupu GONG Wenfang GENG Xiaoli PANG Baoyin WANG Liru DU Xiongming | 2018 | Journal of Cotton Research2018,1,3: | 0
