| 9 | Efficient zonal diagnosis with maximum satisfiability显示文摘Model-based diagnosis(MBD) has been widely acknowledged as an effective diagnosis paradigm.However, for large scale circuits, it is difficult to find all cardinality-minimal diagnoses within a reasonable time. This paper proposes a novel method that takes a significant step in this direction. The idea is to divide a circuit into zones and compute the cardinality-minimal diagnoses by finding subset-minimal diagnoses with cardinality-minimal via a maximum satisfiability(MaxSAT) solver on an abstracted circuit that is composed of these zones instead of all components. We also propose a new propagate-extend method for extending the seed-TLDs to obtain all cardinality-minimal diagnoses efficiently. We implement our method with a state-of-the-art core-guided MaxSAT solver, and present evidence that it significantly improves the diagnosis efficiency on ISCAS-85 circuits. | 15 | Prediction of a common neutralizing epitope of H5N1 avian influenza virus by in silico molecular docking显示文摘The H5N1 avian influenza virus (AIV) has widely spread in Asia, Europe and Africa, making a large amount of economic loss. Recently, our research group has screened a common neutralizing mono- clonal antibody named 8H5, which can neutralize almost all H5 subtype AIV ever isolated so far. Obvi- ously, this monoclonal antibody would benefit for research and development of the universal AIV vac- cine and design of the drug against H5N1 AIV in high mutation rate. In this study, the homology mod- eling was applied to generate the 3D structure of 8H5 Fab fragment, and 'canonical structure' method was used to define the specified loop conformation of CDR regions. The model was subjected to en- ergy minimization in cvff force field with Discovery module in Insight II program. The resulting model has correct stereochemistry as gauged from the Ramachandran plot calculation and good 3D-structure compatibility as assessed by interaction energy analysis, solvent accessible surface (SAS) analysis, and Profiles-3D approach. Furthermore, the 8H5 Fab model was subjected to docking with three H5 subtype hemagglutinin (HA) structures deposited in PDB (ID No: 1jsm, 2ibx and 2fk0) respectively. The result indicates that the three docked complexes share a common binding interface, but differ in bind- ing angle related with HA structure similarity between viral subtypes. In the light of the three HA inter- faces with structural homology analysis, the common neutralizing epitope on HA recognized by 8H5 consists of 9 incontinuous amino acid residues: Asp68, Asn72, Glu112, Lys113, Ile114, Pro118, Ser120, Tyr137, Tyr252 (numbered as for 1jsm sequence). | 18 | Cardiorespiratory Effects of Indoor Ozone Exposure Associated with Changes in Metabolic Profiles among Children:A Repeated-Measure Panel Study显示文摘Ozone is one of the major gaseous pollutants associated with short-term adverse cardiopulmonary effects,even at concentrations below the current indoor air quality limits.However,the underlying biological mechanisms of cardiorespiratory changes with exposure to ozone remain unclear.To further explore molecular linkages between indoor ozone exposure and relevant cardiorespiratory effects,a repeated-measure panel study including 46 schoolchildren was conducted and real-time exposure measurements including ozone were performed inside classrooms every weekday during the study period.Repeated health measurements and urine sample collection were conducted in each participant.Ultra-high-performance liquid chromatography/tandem mass spectrometry and meet-in-metabolite approach were used in metabolomics analysis.Methods including mixed-effect models were adopted to identify metabolites associated with ozone exposure or health indices.Nine metabolites were found to be associated with ozone after mixed-effect model analysis,which are mainly involved in amino acid and bile acid metabolism.Boys may have a greater decrease in bile acid and RNA related metabolites.Four of the nine ozone-related metabolites were also associated with cardiorespiratory function indices.Furthermore,26.67%of the positive association between ozone and heart rate was mediated by cholestane-3,7,12,25-tetrol-3-glucuronide.Exposure to ozone below the current indoor standards was associated with the deteriorated cardiovascular function by disturbing bile acid and endogenous nitric oxide-related oxidation and inflammation,and associated with the exacerbated airway inflammation by reducing GPx-related anti-oxidation.The results provide metabolic evidence of the cardiorespiratory effects of indoor ozone exposure.Indoor ozone pollution should be controlled further,and more attention should be paid to preventing its adverse health effects,especially in children. | Shan Liu Qingyu Huang Xi Zhang Wei Dong Wenlou Zhang Shaowei Wu Di Yang Bingru Nan Jie Zhang Heqing Shen Xinbiao Guo Furong Deng | 2021 | The Innovation2021,2,1: | 1 |
