| 1 | Three-dimensional multimodal sub-diffraction imaging with spinning-disk confocal microscopy using blinking/ fluctuating probes显示文摘Three-dimensional imaging cannot be achieved easily using previously developed localization super-resolution techniques. Here, we present a three-dimensional multimodal sub-diffraction imaging technique with spinning-disk (SD) confocal microscopy called 3D-MUSIC, which not only has all the advantages of SD confocal microscopy such as fast imaging speed, high signal-to-noise ratio, and optical-sectioning capability, but also extends its spatial resolution limit along all three dimensions. Both axial and lateral resolution can be improved simul- taneously by virtue of the blinking/fluctuating nature of modified fluorescent probes, exemplified with the quantum dots. Further, super-resolution images with dual modality can be obtained through super-resolution optical fluctuation imaging (SOFI) and bleaching/blinking-assisted localization microscopy (BALM). | 5 | Factors affecting directional migration of bone marrow mesenchymal stem cells to the injured spinal cord显示文摘Microtubule-associated protein 1B plays an important role in axon guidance and neuronal migration. In the present study, we sought to discover the mechanisms underlying microtubule-associated protein 1B mediation of axon guidance and neuronal migration. We exposed bone marrow mesenchymal stem cells to okadaic acid or N-acetyl-D-erythro-sphingosine(an inhibitor and stimulator, respectively, of protein phosphatase 2A) for 24 hours. The expression of the phosphorylated form of type I microtubule-associated protein 1B in the cells was greater after exposure to okadaic acid and lower after N-acetyl-D-erythro-sphingosine. We then injected the bone marrow mesenchymal stem cells through the ear vein into rabbit models of spinal cord contusion. The migration of bone marrow mesenchymal stem cells towards the injured spinal cord was poorer in cells exposed to okadaic acid- and N-acetyl-D-erythro-sphingosine than in non-treated bone marrow mesenchymal stem cells. Finally, we blocked phosphatidylinositol 3-kinase(PI3K) and extracellular signal-regulated kinase 1/2(ERK1/2) pathways in rabbit bone marrow mesenchymal stem cells using the inhibitors LY294002 and U0126, respectively. LY294002 resulted in an elevated expression of phosphorylated type I microtubule-associated protein 1B, whereas U0126 caused a reduction in expression. | 6 | Hysteresis behaviors of compressor rotating stall with cusp catastrophic model显示文摘Rotating stall is a complex nonlinear dynamic phenomenon which is always characterized by catastrophe and hysteresis in high aerodynamic-loading compressor. Exploring the key contributing factors and characteristic rules of hysteresis is very important for compressor design and flow instability control. In this paper, a novel model method is proposed to analyze the hysteresis behaviors to extend the understanding of compressor rotating stall. The equilibrium states of compressor system under different conditions are first described based on Moore-Greitzer model. Then,through assessing the stability of the equilibrium points by Liapunov's theorem, the ratio of shutoff head to compressor characteristic semi-height is found to affect the stall hysteresis: the size of hysteresis loop will gradually decrease, even disappear with the increase of the ratio. Combing the effects of both the ratio and throttle coefficient, the hysteresis behaviors of compressor stall under multi-parameters can be found to be consistent with the topological properties of cusp catastrophic model by Thom's catastrophe theory. | 9 | Intercellular adhesion molecule-5 protects PAJU cells during in vitro ischemia following cerebral infarction显示文摘BACKGROUND: Intercellular adhesion molecule-5 (ICAM-5) relieves the damage of beta-amyloid protein to PAJU cells. However, little is known about how ICAM-5 works as a neurotrophic factor, or whether ICAM-5 lessens neuronal damage under ischemic conditions following cerebral infarction. OBJECTIVE: To investigate the effects of ICAM-5 on PAJU cells growth in serum-free medium under ischemic conditions following cerebral infarction. DESIGN, TIME AND SETTING: The cytological in vitro study was performed at the Central Laboratory, Second Xiangya Hospital, Central South University, China, in June 2009. MATERIALS: Human ICAM-5 gene transfected into PAJU-TLN cells was supplied by the Life Science College, Helsinki University, Finland. Empty vector transfected PAJU-NEO cells were established by the Gene Center, Second Xiangya Hospital, Central South University, China. METHODS: PAJU-TLN cells transfected with human ICAM-5 or empty vector were incubated in serum-free medium. MAIN OUTCOME MEASURES: Phase contrast microscopy was used to observe changes in PAJU cell morphology. 3-(4, 5-dimethylthiazolzyl)-2, 5-diphenyltetrazolium bromide was used to determine cell viability. Hoechst 33258 was used to stain cell nuclei. Flow cytometry was utilized to measure the apoptosis rate of both PAJU-TLN and PAJU-NEO cells. RESULTS: Both PAJU-TLN and PAJU-NEO cells were injured by cultivating in serum-free medium, but the survival rate of PAJU-TLN cells was significantly higher. | 10 | Alleviating the crosstalk effect via a fine-moulded light-blocking matrix for colour-converted micro-LED display with a 122% NTSC gamut显示文摘One of the major challenges when fabricating high gamut colour-converted micro-light-emitting diodes(LEDs)displays is severe crosstalk effect among adjacent pixels because of the wide view-angle feature of micro-LED chips.In this study,potential factors that contribute to the crosstalk effect were systematically simulated.We observed that precisely filling the space between each micro-LED chip with a light blocking matrix(LBM)can be a promising solution to alleviate this risk.After careful investigations,a press-assisted moulding technique was demonstrated to be an effective approach of fabricating the LBM.Nevertheless,experimental observations further revealed that residual black LBM on the surface of micro-LEDs severely reduces the brightness,thereby compromising the display performance.This problem was successfully addressed by employing a plasma etching technique to efficiently extract the trapped light.Eventually,a top-emitting blue micro-LED-based backlight fine-moulded with a black LBM was developed and combined with red and green quantum dot colour-conversion layers for full-colour display.The colour gamut of our manufactured display prototype can cover as high as 122%that of the National Television Standards Committee. | Yongming Yin Zhiping Hu Muhammad Umair Ali Miao Duan Yongwei Wu Ming Liu Wenxiang Peng Jun Hou Dongze Li Xin Zhang Hong Meng | 2022 | Light(Advanced Manufacturing)2022,3,3: | 2 |
