| 1 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 2 | Autumn Photoproduction of Carbon Monoxide in Jiaozhou Bay, China显示文摘Carbon monoxide(CO) plays a significant role in global warming and atmospheric chemistry. Global oceans are net natural sources of atmospheric CO. CO at surface ocean is primarily produced from the photochemical degradation of chromophoric dissolved organic matter(CDOM). In this study, the effects of photobleaching, temperature and the origin(terrestrial or marine) of CDOM on the apparent quantum yields(AQY) of CO were studied for seawater samples collected from Jiaozhou Bay. Our results demonstrat that photobleaching, temperature and the origin of CDOM strongly affected the efficiency of CO photoproduction. The concentration, absorbance and fluorescence of CDOM exponentially decreased with increasing light dose. Terrestrial riverine organic matter could be more prone to photodegradation than the marine algae-derived one. The relationships between CO AQY and the dissolved organic carbon-specific absorption coefficient at 254 nm for the photobleaching study were nonlinear, whereas those of the original samples were strongly linear. This suggests that: 1) terrestrial riverine CDOM was more efficient than marine algae-derived CDOM for CO photoproduction; 2) aromatic and olefinic moieties of the CDOM pool were affected more strongly by degradation processes than by aliphatic ones. Water temperature and the origin of CDOM strongly affected the efficiency of CO photoproduction. The photoproduction rate of CO in autumn was estimated to be 31.98 μmol m-2 d-1 and the total DOC photomineralization was equivalent to 3.25%- 6.35% of primary production in Jiaozhou Bay. | 3 | Impact of land use/land cover changes on ecosystem services in the Nenjiang River Basin, Northeast China显示文摘Introduction:The Nenjiang River Basin is an important foodstuff base and eco-environmental fragile area in Northeast China.With the rapid rise in human population,human-induced changes in land use/land cover form an important component of regional environment and ecosystem service change.At the local and regional level,the ecosystem service concept can act as a decision support tool for a stakeholder to reach sustainable land use management.However,the prevailing ecosystem service evaluation would produce a biggish warp when it is applied to concrete area.So,it is essential to evaluate ecosystem service change according to the local actuality.Method:According to 1:250,000 land use/land cover maps of China and the adjusted equivalent value per unit area of ecosystem services in the Nenjiang River Basin,we evaluated the ecosystem service change of the river basin from 1980 to 2005.Results:The forest and wetland,which are mainly located in the upstream mountainous area of the Nenjiang River Basin,were the two valuable land cover types,accounting for more than three quarters of the total ecosystem service value of the river basin.As for individual ecosystem service,besides the food production,all of the ecosystem service values declined from 1980 to 2005.The total decline of 2.43 billion USD was mainly due to the cultivation of grassland(14.34%of the area in 1980)and wetland(4.62%of the area in 1980)in the downstream plain.Conclusions:Due to the increase in population and the concomitant requirement of grain,the inconsistency between decision-making at the macro-level,and the objective of agricultural production at the micro-level,cultivated land was increased through zealous reclamation of grassland,marginal woodland,and even fallow land.Tremendous land use/land cover changes had caused great damages to the ecological environment such as land degradation and ecosystem service recession.So,the policies of the Grain for Green and Construction of Ecological Province projects should be well-implemented to optimize land use/land cover. | Zhiliang Wang Zongming Wang Bai Zhang Chunyan Lu Chunying Ren | 2015 | Ecological Processes2015,4,1: | 5 |
| 4 | Outcomes of Coaxial Micro-incision Phacoemulsification in Nanophthalmic Eyes: Report of Retrospective Case Series显示文摘Purpose: The surgical risk and complication rate after cataract surgery are extremely high in patients with nanophthalmos.This study is designed to compare the visual and refractive outcomes before and after coaxial micro-incision phacoemulsification and evaluate postoperative complications.Methods:.Fifty nine patients(89 eyes) with axial length(AL)< 21 mm diagnosed with nanophthalmos were enrolled in this retrospective study. All patients underwent coaxial micro-incision phacoemulsification and IOL implantation..The main outcome measures included anterior chamber depth(ACD),anterior chamber volume(ACV), anterior chamber angle(A CA), intraocular pressure(IOP) and best corrected visual acuity(BCVA). Wilcoxon signed rank test or Mann-Whitney test,and Chi-square test and logistic regression analysis were performed for statistical tests as appropriate.Results:.The median AL was 19.63 mm...Sixty-six eyes(74.16%) had a history of surgical intervention. Postoperative ACD, ACV and ACA were increased significantly(all P<0.001),whereas postoperative IOP was decreased significantly.(P <0.001) after surgery. Previous surgical intervention was related to a reduction in the postoperative ACD and ACA(P<0.01),and both preoperative and postoperative IOP.(P <0.001).Postoperative BCVA was improved in 94.38% of the cases.Intraoperative complications mainly included iridoschisis(6eyes, 6.74%). Early postoperative complications included temporary corneal edema(TCE)(23 eyes, 25.84%), anterior inflammatory response(AIR)(19 eyes, 21.35%), cystoid macular edema(CME)(14 eyes, 15.73%), and uveal effusion(4 eyes, 4.49%). Late postoperative complications included CME(8 eyes, 8.99%), uveal effusion(8 eyes, 8.99%),malignant glaucoma(2 eyes, 2.25%) and posterior capsular opacification(PCO)(10 eyes, 11.24%). The majority of complications(80%) were successfully resolved by pharmacotherapy or operation. | 5 | The efficacy of NP11-4-derived immunotoxin scFv-artesunate in reducing hepatic fibrosis inducedby Schistosoma japonicum in mice显示文摘Schistosomiasis is one of the most prevalent parasitic diseases in China,and hepatic fibrosis caused by schistosome infection is the principal cause of death.The aim of this study was to evaluate the efficacy of NP11-4derived immunotoxin scFv-artesunate on Schistosoma japonicum-induced hepatic fibrosis.A single-chain variable fragment (scFv) was generated from the murine anti-Schistosoma japonicum (S.japanicum) monoclonal antibody NP11-4.The scFv was expressed as a soluble protein and purified by Ni-affinity chromatography.After conjugation with artesunate,the binding ability with soluble egg antigens (SEA) was determined by an enzyme-linked immunosorbent assay (ELISA).The biological activity of purified scFv,scFv-artesunate (immunotoxin),and artesunate was detected in vivo.Image-Pro Plus software was used to analyze the size of egg granuloma and the extent of liver fibrosis.The recombinant scFv expession vector was constructed and expressed successfully.After purification by a His-trap Ni-affinity column,the scFv yield was approximately 0.8 mg/L of culture medium.ELISA results showed that chemical conjugation did not affect the binding activity of the immunotoxin.Our animal experiments indicated that the immunotoxin could significantly reduce the size of egg granuloma in the liver and inhibit hepatic fibrosis.The immunotoxin could be used as a promising candidate in the targeted therapy of S.japonicum-induced hepatic fibrosis. | Hong Li Chunyan Gu Yongya Ren Yang Dai Xiaojuan Zhu Jing Xu Yuhua Li Zhenning Qiu Jin Zhu Yinchang Zhu Xiaohong Guan Zhenqing Feng | 2011 | The Journal of Biomedical Research2011,25,2: | 3 |
| 6 | Identifying EGFR-Expressed Cells and Detecting EGFR Multi-Mutations at Single-Cell Level by Microfluidic Chip显示文摘EGFR mutations companion diagnostics have been proved to be crucial for the efficacy of tyrosine kinase inhibitor targeted cancer therapies. To uncover multiple mutations occurred in minority of EGFR-mutated cells,which may be covered by the noises from majority of unmutated cells, is currently becoming an urgent clinical requirement. Here we present the validation of a microfluidic-chip-based method for detecting EGFR multimutations at single-cell level. By trapping and immunofluorescently imaging single cells in specifically designed silicon microwells, the EGFR-expressed cellswere easily identified. By in situ lysing single cells, the cell lysates of EGFR-expressed cells were retrieved without cross-contamination. Benefited from excluding the noise from cells without EGFR expression, the simple and cost-effective Sanger's sequencing, but not the expensive deep sequencing of the whole cell population, was used to discover multi-mutations. We verified the new method with precisely discovering three most important EGFR drugrelated mutations from a sample in which EGFR-mutated cells only account for a small percentage of whole cell population. The microfluidic chip is capable of discovering not only the existence of specific EGFR multi-mutations,but also other valuable single-cell-level information: on which specific cells the mutations occurred, or whether different mutations coexist on the same cells. | 8 | Eastern Ancient Terrane of the North China Craton显示文摘Based on the spatial distribution of ancient rocks and zircons, three ancient terranes older than ca. 2.6 Ga have recently been identified in the North China Craton, namely the Eastern, Southern, and Central Ancient Terranes. The Eastern Ancient Terrane is the best studied and understood of the three ancient terranes. It has a long geological history back to ca. 3.8 Ga ago and includes the areas of Anshan-Benxi, eastern Hebei, eastern Shandong and western Shandong. In Anshan-Benxi, several different types of 3.8 Ga rocks were discovered together with 3.1-3.7 Ga rocks, whereas 2.9-3.0 Ga K-rich granites and 2.5 Ga syenogranite occur on larger scales. In eastern Hebei, 3.0-3.4 Ga rocks and older detrital and xenocrystic zircons were identified. In eastern Shandong, there are a large volumes of 2.7 Ga and 2.9 Ga rocks. In western Shandong, early Neoarchean(2.6-2.7 Ga) intrusive and supracrustal rocks are widely distributed. Whole-rock Nd and zircon Hf isotope data suggest that both mantle additions and crustal recycling played important roles within the Eastern Ancient Terrane during almost every tectono-magmatic event.
