| 1 | A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. | 3 | The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. | 7 | Association between socioeconomic status and chronic obstructive pulmonary disease in Jiangsu province, China: a population-based study显示文摘Background:Chronic obstructive pulmonary disease(COPD)is a common public health problem worldwide.Recent studies have reported that socioeconomic status(SES)is related to the incidence of COPD.This study aimed to investigate the association between SES and COPD among adults in Jiangsu province,China,and to determine the possible direct and indirect effects of SES on the morbidity of COPD.Methods:A cross-sectional study was conducted among adults aged 40 years and above between May and December of 2015 in Jiangsu province,China.Participants were selected using a multistage sampling approach.COPD,the outcome variable,was diagnosed by physicians based on spirometry,respiratory symptoms,and risk factors.Education,occupation,and monthly family average income(FAI)were used to separately indicate SES as the explanatory variable.Mixed-effects logistic regression models were introduced to calculate odds ratios(ORs)and 95%confidence intervals(CIs)for examining the SES-COPD relationship.A pathway analysis was conducted to further explore the pulmonary function impairment of patients with different SES.Results:The mean age of the 2421 participants was 56.63±9.62 years.The prevalence of COPD was 11.8%(95%CI:10.5%–13.1%)among the overall sample population.After adjustment for age,gender,residence,outdoor and indoor air pollution,body weight status,cigarette smoking,and potential study area-level clustering effects,educational attainment was negatively associated with COPD prevalence in men;white collars were at lower risk(OR:0.60,95%CI:0.43–0.83)of experiencing COPD than blue collars;compared with those within the lower FAI subgroup,participants in the upper(OR:0.68,95%CI:0.49–0.97)tertiles were less likely to experience COPD.Such negative associations between all these three SES indicators and COPD were significant among men only.Education,FAI,and occupation had direct or indirect effects on pulmonary function including post-bronchodilator forced expiratory volume in 1 s/forced vital capacity(FEV1/FVC),FEV1,FVC,and FEV1 percentage of predicted.Education,FAI,and occupation had indirect effects on pulmonary function indices of all participants mainly through smoking status,indoor air pollution,and outdoor air pollution.We also found that occupation could affect post-bronchodilator FEV1/FVC through body mass index.Conclusions:Education,occupation,and FAI had an adverse relationship with COPD prevalence in Jiangsu province,China.SES has both direct and indirect associations with pulmonary function impairment.SES is of great significance for COPD morbidity.It is important that population-based COPD prevention strategies should be tailored for people with different SES. | Dan-Dan Zhang Jian-Nan Liu Qing Ye Zi Chen Ling Wu Xue-Qing Peng Gan Lu Jin-Yi Zhou Ran Tao Zhen Ding Fei Xu Linfu Zhou | 2021 | Chinese Medical Journal2021,,13: | 5 |
| 8 | Outcomes of Coaxial Micro-incision Phacoemulsification in Nanophthalmic Eyes: Report of Retrospective Case Series显示文摘Purpose: The surgical risk and complication rate after cataract surgery are extremely high in patients with nanophthalmos.This study is designed to compare the visual and refractive outcomes before and after coaxial micro-incision phacoemulsification and evaluate postoperative complications.Methods:.Fifty nine patients(89 eyes) with axial length(AL)< 21 mm diagnosed with nanophthalmos were enrolled in this retrospective study. All patients underwent coaxial micro-incision phacoemulsification and IOL implantation..The main outcome measures included anterior chamber depth(ACD),anterior chamber volume(ACV), anterior chamber angle(A CA), intraocular pressure(IOP) and best corrected visual acuity(BCVA). Wilcoxon signed rank test or Mann-Whitney test,and Chi-square test and logistic regression analysis were performed for statistical tests as appropriate.Results:.The median AL was 19.63 mm...Sixty-six eyes(74.16%) had a history of surgical intervention. Postoperative ACD, ACV and ACA were increased significantly(all P<0.001),whereas postoperative IOP was decreased significantly.(P <0.001) after surgery. Previous surgical intervention was related to a reduction in the postoperative ACD and ACA(P<0.01),and both preoperative and postoperative IOP.(P <0.001).Postoperative BCVA was improved in 94.38% of the cases.Intraoperative complications mainly included iridoschisis(6eyes, 6.74%). Early postoperative complications included temporary corneal edema(TCE)(23 eyes, 25.84%), anterior inflammatory response(AIR)(19 eyes, 21.35%), cystoid macular edema(CME)(14 eyes, 15.73%), and uveal effusion(4 eyes, 4.49%). Late postoperative complications included CME(8 eyes, 8.99%), uveal effusion(8 eyes, 8.99%),malignant glaucoma(2 eyes, 2.25%) and posterior capsular opacification(PCO)(10 eyes, 11.24%). The majority of complications(80%) were successfully resolved by pharmacotherapy or operation. | 12 | Effect of Aβ protein on inhibiting proliferation and promoting apoptosis of retinal pigment epithelial cells显示文摘AIM: To identify the effect and regulatory mechanism of amyloid β(Aβ) protein on retinal pigment epithelial(RPE)cells in cell proliferation and apoptosis, and clarify Aβ role in the pathogenesis of age-related macular degeneration(AMD).METHODS: The model of Aβ25-35 protein cytotoxicity in RPE cell was successfully established to investigate the effect of Aβ protein on RPE cells in vitro. Based on Aβprotein, the specific inhibitors(HY-50682 or BAY11-7082) or activating agent(lipopolysaccharide) was used to analyze the regulatory mechanism of Aβ protein to RPE cells on cell proliferation and apoptosis by flow cytometry, realtime polymerase chain reaction, Western blotting, enzymelinked immunosorbent assay and dual-luciferase reporter gene assay.RESULTS: The number of RPE cells, treated with Aβ25-35 from 0.3 to 60 μmol/L, significantly reduce(P<0.01), and had the dose-dependent effect. Aβ protein 60 μmol/L inhibits the G1/S phase transition(P<0.01) and down-regulated cyclin E mRNA level(P<0.01). Similarly, Aβ25-35 induced a significant increase of cell apoptosis, accompanied by the significantly higher level of activated caspase3 protein. Furthermore, nuclear factor-kappaB(NF-κB)activity and phosphorylated Iκ-Ba level would significantly lower in treated RPE cells. Using specific inhibitors or activating agent based on the Aβ, the cell numbers, NF-κB activity, phosphorylated Iκ-Ba level, receptor for advanced glycation endproducts(RAGE) gene expression levels,cyclin E mRNA level and activated caspase 3 level had accordingly changed by different methods, confirming that RAGE/NF-κB signaling pathway involved in the regulation of Aβ protein on RPE cell apoptosis and proliferation.CONCLUSION: Aβ protein inhibits cell proliferation and activates apoptosis via inactivation of the RAGE/NF-κB signaling pathway in RPE cell. | Zi Ye Shou-Zhi He Zhao-Hui Li | 2018 | International Journal of Ophthalmology(English edition)2018,11,6: | 3 |
| 13 | Molecular Characterization and Expression Analysis of S1 self-incompatibility Locus-linked Pollen 3.15 Gene in Citrus reticulata显示文摘Gametophytic self-incompatibility (GSI) is controlled by a highly polymorphic locus called the S-locus, which is an important factor that can result in seedless fruit in Citrus. The S 1 self-incompatibility locus-linked pollen 3.15 gene (S1-3.15) belongs to a type of S locus gene. The role of S1-3.15 in the SI reaction of Citrus has not yet been reported. In this study, full-length sequences of cDNA and DNA encoding the S1-3.15 gene, referred to as CrS1-3.15 , were isolated from 'Wuzishatangju' (Self-incompatibility, SI) and 'Shatangju' (Self-compatibility, SC) . The predicted amino acid sequences of CrS1-3.15 between 'Wuzishatangju' and 'Shatangju' differ by only three amino acids. Compared to 'Wuzishatangju', three bases were substituted in the genomic DNA of CrS1-3.15 from 'Shatangju'. Southern blot results showed that one copy of CrS1-3.15 existed in the genomic DNA of both 'Wuzishatangju' and 'Shatangju'. The expression level of the CrS1-3.15 gene in the ovaries of 'Shatangju' was approximately 60-fold higher than that in the ovaries of 'Wuzishatangju'. When 'Wuzishatangju' was cross-pollinated, the expression of CrS1-3.15 was upregulated in the ovaries at 3d, and the highest expression levels were detected in the ovaries at 6d after cross-pollination of 'Wuzishatangju' × 'Shatangju'. To obtain the CrS1-3.15 protein, the full-length cDNA of CrS1-3.15 genes from 'Wuzishatangju' and 'Shatangju' was successfully expressed in Pichia pastoris. Here one simple point spread function (PSF) based method with only the standard deviation method used to evaluate the PSF stability over the depth of focus is taken to optimize for the best coefficients of pupil phase mask in practical optical systems. Results of imaging simulations for optical systems with and without pupil phase mask are presented, and the sharpness of image is calculated for comparison. | 15 | Diagnosis,treatment,and prevention of severe acute hepatitis of unknown etiology in children显示文摘Background Severe acute hepatitis of unknown etiology in children has recently exhibited a global trend of concentrated occurrence.This review aimed to summarize the current available information regarding the outbreak of severe acute hepatitis and introduce our hospital’s previous experiences with the diagnosis and treatment of severe acute hepatitis for reference.Data sources Websites including the UK Health Security Agency,European Centre for Disease Prevention and Control,CDC,WHO,and databases including PubMed/Medline,Cochrane Library,Embase and Web of Science were searched for articles on severe acute hepatitis in children.Results As of May 26,2022,a total of 650 cases have been reported in 33 countries;at least 38(6%)children required liver transplantation,and nine(1%)died.Cases are predominantly aged between 3 and 5 years old,and there are no epidemiological links among them.The common manifestations are jaundice,vomiting and pale stools.Adenovirus tested positive in most cases,and SARS-CoV-2 and other viruses were detected in a few cases,but virus particles were not found in liver tissue.Adenovirus immunohistochemistry showed immunoreactivity in the intrasinusoidal lumen from some liver samples.The hierarchical treatment includes symptomatic and supportive therapy,management of coagulation disorders and hepatic encephalopathy,artifcial liver support,and liver transplantation(approximately 6%–10%of cases require liver transplant).Conclusions The etiology of this severe acute hepatitis in children is not clear.The clinical features are severe acute hepatitis with signifcantly elevated liver enzymes.Clinicians need to be alert to children with hepatitis. | Ying‑Hu Chen Jin‑Gan Lou Zi‑Hao Yang Qing‑Jiang Chen Chun‑Zhen Hua Sheng Ye Chen‑Mei Zhang Jie Chen Zong‑Wei Huang Jin‑Dan Yu Zhi‑Gang Gao Qiang Shu | 2022 | World Journal of Pediatrics2022,18,8: | 2 |

| 17 | PVT1 affects ENIT and cell proliferation and migration via regulating p21 in triple-negative breast cancer cells cultured with mature adipogenic medium显示文摘Excessive adiposity has long been proved to be associated with greater incidence and mortality of breast cancer in post-menopausal women.However,the effects and underlying mechanisms of human adipocytes on breast cancer cells remain largely unknown.In recent years,several reports have revealed the oncogenic role of long non-coding RNA PVT1 in breast cancer.Here,we aimed to investigate the role and underlying mechanisms of PVT1 in triple-negative breast cancer (TNBC)cells cultured with mature adipogenic medium.At first,we successfully induced adipogenic differentiation from human adipose-derived mesenchymal stern cells and collected the mature adipogenic medium to mimic excessive adiposity.Our results demonstrated that the mature adipogenic medium promoted the epithelial-mesenchymal transition,enhanced the cell viability and migration potential of TNBC cells.In addition,we proved that mature adipogenic medium affected the PVT1 expression and inhibition of the PVT1 disturbed the role of mature adipogenic medium in TNBC cells.Finally,we illustrated that repression of p21 restored the phenotype caused by PVT1 knockdown in TNBC cells treated with mature adipogenic medium.Taken together,our results demonstrated that PVT1 affected the role of mature adipogenic medium in TNBC cells via modulating p21 expression. | Lingli Wang Ruirui Wang Zi Ye Yanyan Wang Xiao Li Weizhen Chen Mengna Zhang Cheguo Cai | 2018 | Acta Biochimica et Biophysica Sinica2018,50,12: | 2 |
