2Generation of new glutinous rice by CRISPR/Cas9-targeted mutagenesis of the Waxy gene in elite rice varieties显示文摘In rice, amylose content(AC) is controlled by a single dominant Waxy gene. We used Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPR-associated 9(Cas_9) to introduce a loss-of-function mutation into the Waxy gene in two widely cultivated elite japonica varieties. Our results show that mutations in the Waxy gene reduce AC and convert the rice into glutinous ones without affecting other desirable agronomic traits, offering an effective and easy strategy to improve glutinosity in elite varieties. Importantly, we successfully removed the transgenes from the progeny. 4Artificial root exudates and soil organic carbon mineralization in a degraded sandy grassland in northern China显示文摘Plant root exudates contain various organic and inorganic components that include glucose, citric and oxalic acid. These components affect rhizosphere microbial and microfaunal activities, but the mechanisms are not fully known. Studies concerned from degraded grassland ecosystems with low soil carbon(C) contents are rare, in spite of the global distribution of grasslands in need of restoration. All these have a high potential for carbon sequestration, with a reduced carbon content due to overutilization. An exudate component that rapidly decomposes will increase soil respiration and CO2 emission, while a component that reduces decomposition of native soil carbon can reduce CO2 emission and actually help sequestering carbon in soil. Therefore, to investigate root exudate effects on rhizosphere activity, citric acid, glucose and oxalic acid(0.6 g C/kg dry soil) were added to soils from three biotopes(grassland, fixed dune and mobile dune) located in Naiman, Horqin Sandy Land, Inner Mongolia, China) and subjected to a 24-day incubation experiment together with a control. The soils were also analyzed for general soil properties. The results show that total respiration without exudate addition was highest in grassland soil, intermediate in fixed dune and lowest in mobile dune soil. However, the proportion of native soil carbon mineralized was highest in mobile dune soil, reflecting the low C/N ratio found there. The exudate effects on CO2-C emissions and other variables differed somewhat between biotopes, but total respiration(including that from the added substrates) was significantly increased in all combinations compared with the control, except for oxalic acid addition to mobile dune soil, which reduced CO2-C emissions from native soil carbon. A small but statistically significant increase in pH by the exudate additions in grassland and fixed dune soil was observed, but there was a major decrease from acid additions to mobile dune soil. In contrast, electrical conductivity decreased in grassland and fixed dune soil and increased in mobile dune. Thus, discrete components of root exudates affected soil environmental conditions differently, and responses to root exudates in soils with low carbon contents can differ from those in normal soils. 7Micronuclei in Nasal Mucosa, Oral Mucosa and Lymphocytes in Students Exposed to Formaldehyde Vapor in Anatomy Class显示文摘The frequency of micronuclei (MN) in cells of the nasal mucosa, oral mucosa and in lym-phocytes was evaluated for 25 students in anatomy classes exposed to formaldehyde (FA) overan 8-week period. Each student served as his or her own control. The time-weighted averageconcentration (TWA) of fOrmaldehyde in anatomical laboratories and in students’ dormitorieswas 0. 508 ± 0. 299 mg/m3 and 0. 012 ± 0. 0025mg/m3, respectively. A higher frequency ofmicronuclei was observed in nasal and oral exfoliative cells after formaldehyde exposure (3. 85± 1. 48 vs 1 .20± 0. 676 and 0. 857 ± 0. 558 vs 0. 568 ± 0. 317, paired-t test: P< 0. 001and P < 0. 01, respectively). 8肝的周期性的巨大的 hemangiomas : 有文学评论的二个稀罕案例的报告显示文摘 Most hepatic hemangiomas(HHs) are small,asymptomatic and do not require clinical intervention.Surgical resection is only indicated for symptomatic hemangiomas.We report here cases of recurrent HHs in 2 women of 37 and 40 years old,who initially presented with abdominal pain and mass.Radiological examination of each tumor revealed a solitary tumor of 14 and 20 cm in diameter,respectively.Surgical liver segmental resections were performed in both,and the diagnosis of cavernous hemangioma was confirmed.Both patients had recurrent tumor on subsequent radiological examination 4-5 years after the initial surgery.In the first patient,a 15 cm recurrent hemangioma was resected,but multiple hemangiomas were again detected 8 years later occupying the other hepatic lobe,which was not amendable for resection.In the second patient,a 16 cm hemangioma was seen on radiogram,and because the lesion was not symptomatic,conservative observation was offered.Recurrence after liver resection of giant hemangioma is extremely rare.The pathogenesis of tumor progression and recurrence is unknown,as is the management of these patients with recurrent hemangioma,particularly when it is extensive and unresectable.Hongfa Zhu Khaled Obeidat Jie Ouyang Sasan Roayaie Myron E Schwartz Swan N Thung 2012World Journal of Gastrointestinal Surgery2012,4,11:5

13Dissecting anxiety-related QTLs in mice by univariate and multivariate mapping显示文摘To detect genes underlying anxiety-related traits in mice,we performed univariate and multivariate QTL mapping analyses of phenotypes obtained from 71 mice of the BXD recombinant inbred (RI) strains (n=528 mice) and their parental strains (C57BL/6J and DBA/2J).Separate and joint mapping analyses were carried out using a linkage map composed of 506 simple sequence repeats (SSRs).The main QTL effects,interactions between pairs of QTLs (epistasis),and their environmental interactions were estimated.The results showed that anxiety-related traits were influenced by multiple QTLs (five main effect QTLs and three epistatic QTLs).Ten potential anxiety-related candidate genes within the QTL intervals on chromosomes 5,13 and 15 were identified.Some of these genes have been reported previously to be associated with the anxiety response.Based on our results,it is suggested that the multivariate QTL mapping approach improves the statistical power for detecting QTL and the precision of parameter estimation.Moreover,multivariate mapping can also detect pleiotropic QTL effects.ZHU ZhiHong ZHANG ChenHao WANG XuSheng COOK Melloni N WILLIAMS Robert LU Lu ZHU Jun 2012Chinese Science Bulletin2012,57,21:2
