1Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. 2Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 3A New Species of the Genus Achalinus from Huangshan, Anhui, China(Squamata: Xenodermidae)显示文摘A new species of the genus Achalinus is described based on five specimens collected from the villages of Huangjialing and Fuxi, Huangshan, Anhui, China. It can be morphologically differentia ted from all the other species in Achalinus except for A. spinalis and A. werneri by the presence of a dotted black streak in the middle of the subcaudal. It can be distinguished from A. spinalis in that its two anterior temporals are in contact with eye, and A. werneri by its light brown flanks. The phylogenetic rela tionship of Achalinus was reconstructed using the mitochondrial locus of cytochrome coxidase subunit 1(CO1). The five new specimens form a monophyletic clade with strong support. The uncorrected p-dista nces between the new species and other representatives of Achalinus range from 13.6% to 21.7%. 4A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences.We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://gffzz4a3adbfa122d4a47hbxvx0onbpkpw6ouw.ffgz.tsg.suse.edu.cn),annotated the sequences by sequence-based clustering,and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools,our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas,part of the predicted human target genes were experimentally validated,suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines.We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions.Moreover,the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction,demonstrated potent medical effects.We propose a Bencao(herbal)Index,including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine.The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine,and may provide potential remedies for the theory and practice of one medicine.Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang 2023Science China(Life Sciences)2023,66,10:2

6StRAP2.3, an ERF-Ⅶ transcription factor, directly activates StInvInh2 to enhance cold-induced sweetening resistance in potato显示文摘Potato invertase inhibitor(StInvInh2)positively regulates cold-induced sweetening(CIS)resistance by inhibiting the activity of vacuolar invertase.The distinct expression patterns of StInvInh2 have been thoroughly characterized in different potato genotypes,but the related CIS ability has not been characterized.The understanding of the regulatory mechanisms that control StInvInh2 transcription is unclear.In this study,we identified an ERF‐Ⅶ transcription factor,StRAP2.3,that directly regulates StInvInh2 to positively modulate CIS resistance.Acting as a nuclear-localized transcriptional activator,StRAP2.3 directly binds the ACCGAC cis-element in the promoter region of StInvInh2,enabling promoter activity.Overexpression of StRAP2.3 in CIS-sensitive potato tubers induced StInvInh2 mRNA abundance and increased CIS resistance.In contrast,silencing StRAP2.3 in CIS-resistant potato tubers repressed the expression of StInvInh2 and decreased CIS resistance.We conclude that cold-responsive StInvInh2 is due to the binding of StRAP2.3 to the ACCGAC cis-element in the promoter region of StInvInh2.Overall,these findings indicate that StRAP2.3 directly regulates StInvInh2 to positively modulate CIS resistance,which may provide a strategy to improve the processing quality of potatoes.Weiling Shi Yuhao Song Tiantian Liu Qiuqin Ma Wang Yin Yuchen Shen Tengfei Liu Chunyan Jiang Kai Zhang Dianqiu Lv Botao Song Jichun Wang Xun Liu 2021Horticulture Research2021,8,1:1
7Microbial community coexisting with harmful alga Karenia mikimotoi and microbial control of algal bloom in laboratory显示文摘Algicidal bacteria have been frequently isolated from algal blooming areas.However,knowledge regarding the microbial communities coexisting with microalgae and their potential application in preventing harmful algal blooms(HABs)is limited.In this study,we investigated the composition of the microbial community coexisting with harmful alga Karenia mikimotoi and its responses to algal control via nutrient stimulation or by adding algicidal strain in microcosms.The microorganisms inhabiting the K.mikimotoi culture consisted of 24 identifi ed phyla,including dominant Proteobacteria(relative abundance 76.24%±7.28%)and Bacteroidetes(22.67%±8.32%).Rhodobacteraceae,Phaeodactylibacter,and Maritimibacter predominated during the algal cultivation.Both the added nutrient and fermentation broth of algicidal strain Pseudoalteromonas QF1 caused a massive death of K.mikimotoi and substantial changes in the coexisting microbial community,in which Rhodobacteraceae and Phaeodactylibacter signifi cantly decreased,while Halomonas and Alteromonas increased.Core operational taxonomic units(OTUs)analysis indicated that 13 OTUs belonging to Rhodobacteraceae,Maritimibacter,Marivita,Nisaea,Phaeodactylibacter,Citreicella,Halomonas,Alteromonas,Marinobacter,Muricauda,and Pseudoalteromonas dominated the changes of the microbial communities observed in the K.mikimotoi culture with or without treatments.Collectively,this study indicated that microbial community inhabiting K.mikimotoi culture includes potential algicidal bacteria,and improves our knowledge about microbial community succession during biocontrol of K.mikimotoi via nutrient stimulation or by adding isolated algicidal strains.Li SUN Peike GAO Yu LI Chao WANG Ning DING Junfeng CHEN Yuhao SONG Chunchen LIU Lun SONG Renjun WANG 2022Journal of Oceanology and Limnology2022,40,3:1
8Effects of Combined Stress from Salt and Herbicide on Microbial Remediation of Soil Contaminated by n-Hexadecane and Phenanthrene显示文摘In this study,physicochemical,cluster,and high-throughput sequencing analyses were used to investigate the joint effects of salt and herbicide(glyphosate)stress factors on the microbial remediation of soil contaminated by n-hexadecane and phenanthrene.Based on the soil’s physical and chemical properties,differences in microbial quantity and enzyme activities were analyzed among the samples,and how that influenced distribution of soil community structure was focused upon.After 120 days of indoor simulated remediation,the cumulative degradation rate of n-hexadecane decreased by 94.92%(blank control),96.96%(low concentration salt and glyphosate stress group),65.07%(high concentration salt and glyphosate stress group),while that of phenanthrene decreased by 87.33%(blank control),86.25%(low concentration salt and glyphosate stress group),58.45%(high concentration salt and glyphosate stress group).The combined stress of salt and herbicides was capable of lowering the reduction efficiency of organic matter,total nitrogen,and total phosphorus,and also restricting the growth of microorganisms and enzyme activities.Cluster analysis results indicated that the non-stress group was similar to the low-concentration compound stress group during different remedial period,whereas both of those differed starkly from the high-concentration compound stress group.High-throughput sequencing revealed that the dominant soil bacteria phyla shifted from Firmicutes to Actinobacteria within 120 days of remediation.Liu Bingkun Zuo Shuai Wang Ruirui Dong Kangning Zhang Xiuxia Li Ruyue Hao Boyu Li Jing Yin Linghao Bai Yuhao Song Jiayu Gu Yingying Sun Juan 2022China Petroleum Processing & Petrochemical Technology2022,24,2:0
9Fano resonance-enhanced Si/MoS_(2) photodetector显示文摘In this work,a Si∕MoS_(2) heterojunction photodetector enhanced by hot electron injection through Fano resonance is developed.By preparing Au oligomers using capillary-assisted particle assembly(CAPA)on the silicon substrate with a nanohole array and covering few-layer MoS_(2) with Au electrodes on top of the oligomer structures,the Fano resonance couples with a Si∕MoS_(2) heterojunction.With on-resonance excitation,Fano resonance generated many hot electrons on the surface of oligomers,and the hot electrons were injected into MoS_(2),providing an increased current in the photodetector under a bias voltage.The photodetectors exhibited a broadband photoresponse ranging from 450 to 1064 nm,and a large responsivity up to 52 A/W at a wavelength of 785 nm under a bias voltage of 3 V.The demonstrated Fano resonance-enhanced Si∕MoS_(2) heterojunction photodetector provides a strategy to improve the photoresponsivity of two-dimensional materials-based photodetectors for optoelectronic applications in the field of visible and near-infrared detection.TIANXUN GONG BOYUAN YAN TAIPING ZHANG WEN HUANG YUHAO HE XIAOYU XU SONG SUN XIAOSHENG ZHANG 2023Photonics Research2023,11,12:0
10Effects of Diesel Concentration on the Thermal Conductivity,Specific Heat Capacity and Thermal Diffusivity of Diesel-Contaminated Soil显示文摘High energy consumption is a serious issue associated with in situ thermal desorption(TD)remediation of sites contaminated by petroleum hydrocarbons(PHs).The knowledge on the thermophysical properties of contaminated soil can help predict accurately the transient temperature distribution in a remediation site,for the purpose of energy conservation.However,such data are rarely reported for PH-contaminated soil.In this study,by taking diesel as a representative example for PHs,soil samples with constant dry bulk density but different diesel mass concentrations ranging from 0% to 20% were prepared,and the variations of their thermal conductivity,specific heat capacity and thermal diffusivity were measured and analyzed over a wide temperature range between 0℃ and 120℃.It was found that the effect of diesel concentration on the thermal conductivity of soil is negligible when it is below 1%.When diesel concentration is below 10%,the thermal conductivity of soil increases with raising the temperature.However,when diesel concentration becomes above 10%,the change of the thermal conductivity of soil with temperature exhibits the opposite trend.This is mainly due to the competition between soil minerals and diesel,because the thermal conductivity of minerals increases with temperature,whereas the thermal conductivity of diesel decreases with temperature.The analysis results showed that,compared with temperature,the diesel concentration has more significant effects on soil thermal conductivity.Regardless of the diesel concentration,with the increase of temperature,the specific heat capacity of soil increases,while the thermal diffusivity of soil decreases.In addition,the results of a control experiment exhibited that the relative differences of the thermal conductivity of the soil samples containing the same concentration of both diesel and a pure alkane are all below 10%,indicating that the results obtained with diesel in this study can be extended to the family of PHs.A theoretical prediction model was proposed based on cubic fractal and thermal resistance analysis,which confirmed that diesel concentration does have a significant effect on soil thermal conductivity.For the sake of practical applications,a regression model with the diesel concentration as a primary parameter was also proposed.WU Yuhao WU Yuefei LUO Gubai ZHANG Teng WANG Qing FAN Liwu SONG Xin YU Zitao 2024Journal of Thermal Science2024,33,2:0
11Recent Advances in Cadmium Sulfide-Based Photocatalysts for Photocatalytic Hydrogen Evolution显示文摘High-efficiency photocatalytic hydrogen evolution(PHE)relies on the development of inexpensive,stable,and efficient photocatalysts.Cadmium sulfide(CdS),as a typical binary metal sulfide,has attracted considerable research attention due to its negative conduction band position,narrow band gap for visible-light response,and strong driving force for PHE.However,the construction of CdSbased photocatalysts and the PHE rate still require improvement for practical applications.In this review,recent advances in CdS-based photocatalysts for PHE via water splitting are systematically summarized.First,the semiconductor properties of CdS,including the crystal and band structures,are briefly introduced.Afterward,the fundamental mechanisms of PHE using semiconductor photocatalysts via water splitting are discussed.Subsequently,the photoactivity of bare CdS with different morphologies and structures,CdS with cocatalyst loading,and CdS-based heterojunction photocatalysts are reviewed and discussed in detail.Finally,the challenges and prospects for exploring advanced CdS-based photocatalysts are provided.Xinlong Zheng Yuhao Liu Yuqi Yang Yiming Song Peilin Deng Jing Li Weifeng Liu Yijun Shen Xinlong Tian 2023Renewables2023,1,1:0
12Targeting UBE2C for degradation by bioPROTACs based on bacterial E3 ligase显示文摘UBE2C(Ubiquitin conjugating enzyme E2 C), a key regulator of cell cycle progression, is a promising target for discovery of antitumor agents. However, it is challenging to develop inhibitors of UBE2C owing to its lack of “druggable” pockets. Bio PROTACs(biological proteolysis targeting chimeras) are a kind of protein-based degraders by fusing an adaptor to a subunit of E3 ligase for ubiquitination and subsequent proteasome-dependent degradation of target protein. We report herein the design and biological evaluation of a UBE2C-targeting bio PROTAC based on the NEL(novel E3 ligase) domain of bacterial E3 ligase Ipa H9.8 and the UBE2C-binding WHB(winged-helix B) domain of APC_(2)(anaphase promoting complex subunit 2). The in vitro ubiquitination test and Mass Spectrometry analysis showed that the bio PROTAC could transfer ubiquitin to surface exposed lysines on UBE2C and catalyzed the formation of polyubiquitin chains.
