1Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. 2Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE.Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu 2019Cellular & Molecular Immunology2019,16,3:15
3Science and Prediction of Heavy Rainfall over China:Research Progress since the Reform and Opening-Up of New China显示文摘This paper reviews the major progress on development of the science and prediction of heavy rainfall over China since the beginning of the reform and opening-up of new China(roughly between 1980 and 2019).The progress of research on the physical mechanisms of heavy rainfall over China is summarized from three perspectives:1)the relevant synoptic weather systems,2)heavy rainfall in major sub-regions of China,and 3)heavy rainfall induced by typhoons.The development and application of forecasting techniques for heavy rainfall are summarized in terms of numerical weather prediction techniques and objective forecasting methods.Greatly aided by the rapid progress in meteorological observing technology and substantial improvement in electronic computing,studies of heavy rainfall in China have advanced to investigating the evolution of heavy-rain-producing storms and observational analysis of the cloud microphysical features.A deeper and more systematic understanding of the synoptic systems of importance to the production of heavy rainfall has also been developed.Operational forecast of heavy rainfall in China has changed from subjective weather event forecasts to a combination of both subjective and objective quantitative precipitation forecasts,and is now advancing toward probabilistic quantitative precipitation forecasts with the provision of forecast uncertainty information.Yali LUO Jisong SUN Ying LI Rudi XIA Yu DU Shuai YANG Yuanchun ZHANG Jing CHEN Kan DAI Xueshun SHEN Haoming CHEN Feifan ZHOU Yimin LIU Shenming FU Mengwen WU Tiangui XIAO Yangruixue CHEN Huiqi LI Mingxin LI 2020Journal of Meteorological Research2020,34,3:10
4Pravastatin alleviates lipopolysaccharide-induced placental TLR4 over-activation and promotes uterine arteriole remodeling without impairing rat fetal development显示文摘Preeclampsia is associated with over-activation of the innate immune system in the placenta,in which toll-like receptor 4(TLR4) plays an essential part.With their potent anti-inflammatory effects,statins have been suggested as potential prevention or treatment of preeclampsia,although evidence remains inadequate.Herewith,we investigated whether pravastatin could ameliorate preeclampsia-like phenotypes in a previously established lipopolysaccharide(LPS)-induced rat preeclampsia model,through targeting the TLR4/NF-κB pathway.The results showed that pravastatin reduced the blood pressure [maximum decline on gestational day(GD) 12,(101.33±2.49) mmHg vs.(118.3±1.37) mmHg,P<0.05] and urine protein level [maximum decline on GD9,(3,726.23± 1,572.86) μg vs.(1,991.03 ±609.37)μg,P< 0.05],which were elevated following LPS administration.Pravastatin also significantly reduced the rate of fetal growth restriction in LPS-treated rats(34.10% vs.8.99%,P<0.05).Further pathological analyses suggested a restoration of normal spiral artery remodeling in preeclampsia rats by pravastatin treatment.These effects of pravastatin were associated with decreased TLR4/NF-κB protein levels in the placenta and IL-6/MCP-1 levels in serum.Additionally,no obvious abnormalities in fetal liver,brain,and kidney were found after administration of pravastatin.These results provide supportive evidence for use of pravastatin in preventing preeclampsia.Muyi Yang Zhenyu Diao Zhiyin Wang Guijun Yan Guangfeng Zhao Mingming Zheng Anyi Dai Yimin Dai Yali Hu 2018The Journal of Biomedical Research2018,32,4:5
5Preparation of core-shell magnetic Fe_3O_4@SiO_2-dithiocarbamate nanoparticle and its application for the Ni^(2+), Cu^(2+) removal显示文摘A novel magnetic nanoparticles-based dithiocarbamate absorbent(Fe_3O_4@SiO_2-DTC) with core-shell structure was synthesized under mild conditions and used in aqueous solution Ni^(2+) and Cu^(2+) ions treatment. The structure, morphology and magnetic properties of the adsorbent were characterized by Xray diffraction(XRD), fourier transformed infrared spectroscopy(FTIR), scanning electron microscopy(SEM), transmission electron microscopy(TEM), and vibrating sample magnetometer(VSM).Fe_3O_4@SiO_2-DTC exhibited a typical superparamagnetic with a saturation magnetization value of52.7 emu/g, which could be rapidly separated from aqueous solution under external magnetic field. We investigated the effects of solution p H, adsorption time, and the initial concentration of heavy metal ions on the adsorption of Ni^(2+) and Cu^(2+). The adsorption equilibrium times of Ni^(2+)and Cu^(2+) on Fe3 O_4@SiO_2-DTC were reached at 15 min and 90 min, respectively. The adsorption kinetic data were fitted to the pseudosecond-order model, and the adsorption data were consistent with the Frenudlich isotherm model. When the initial concentration of heavy metal ions was 250 mg/L, the maximum adsorption capacity of Ni^(2+) and Cu^(2+) at room temperature was 235.23 mg/g and 230.49 mg/g, respectively. In addition, we discussed the plausible adsorption mechanism. 7Changes in wheat starch grains using different cooking methods: Insights into ancient food processing techniques显示文摘We investigate the morphological changes in starch grains from wheat(Triticum aestivum) using different cooking methods(boiling, steaming, frying, and baking). We compare the cooked starch grains with starch grains from ancient wheat flour cakes(Astana Cemetery, Turpan Basin, Xinjiang, China) to determine the cooking techniques used by people in Xinjiang 1200 years ago. Heat and water content affect starch grains when different cooking methods are used. Boiling and steaming results in the collapse of wheat starch grains accompanied by extreme swelling, curved granules, pasting, almost full gelatinization, a distinct extinction cross and vague granule outlines. Frying and baking cause less swelling, fewer curved granules, less pasting and only partial gelatinization of wheat starch grains, but the extinction lines are still distinct and the outlines of granules relatively clear. 8Placenta-derived IL-32β activates neutrophils to promote preeclampsia development显示文摘Immune activation at the maternal-fetal interface is a main pathogenic factor of preeclampsia(PE).Neutrophils(PMNs)are activated in PE patients,but the mechanism and consequences of PMN activation need to be further explored.Here,we demonstrated that interleukin-32(IL-32)expression was significantly upregulated in syncytiotrophoblasts(STBs)and that IL-32β was the major isoform with increased expression in the placenta of severe PE(sPE)patients.Furthermore,the level of IL-32 expression in the placenta was correlated with its level in the serum of sPE patients,indicating that IL-32 in the serum is derived mainly from the placenta.Then,in vitro experiments showed that IL-32β could highly activate PMNs and that these IL-32β-activated PMNs were better able to adhere to endothelial cells(HUVECs)and enhance the expression of vascular cell adhesion molecule-1(VCAM-1)and intercellular cell adhesion molecule-1(ICAM-1)in HUVECs,which could be reversed by preincubation with the NADPH oxidase inhibitor VAS 2870.In addition,we showed that IL-32β mainly activated PMNs by binding to proteinase 3.Finally,IL-32β administration induced a PE-like phenotype in a pregnant mouse model.This study provides evidence of the involvement of IL-32β in the pathogenesis of PE.Dan Liu Qiang Li Hailin Ding Guangfeng Zhao Zhiyin Wang Chenrui Cao Yimin Dai Mingming Zheng Xiangyu Zhu Qianwen Wu Ya Wang Honglei Duan Huirong Tang Xianyan Lu Yayi Hou Yali Hu 2021Cellular & Molecular Immunology2021,18,4:2

12Coextraction of vanadium and manganese from highmanganese containing vanadium wastewater by a solvent extraction-precipitation process显示文摘High-manganese containing vanadium waste-water(HMVW)is commonly produced during the vanadium extraction process from vanadium titano-magnetite.HMVW cannot be reused and discharged directly,and is harmful to the environment and affect product quality due to heavy metals in the wastewater.The wastewater is usually treated by lime neutralization,but valuable metals(especially V and Mn)cannot be recovered.In this study,an efficient and environmentally friendly method was developed to recover valuable metals by using a solvent extraction-precipitation process.In the solvent extraction process,98.15%of vanadium was recovered,and the V2Os product,with a purity of 98.60%,was obtained under optimal conditions.For the precipitation process,91.05%of manganese was recovered as MnCO3 which meets the III grade standard of HG/T 2836-2011.Thermodynamic simulation analysis indicated that MnCO3 was selectively precipitated at pH 6.5 while Mg and Ca could hardly be precipitated.The results of X-ray diffraction and scanning electron microscopy demonstrated that the obtained V2Os and MnCO3 displayed a good degree of crystallinity.The treated wastewater can be returned for leaching,and resources(V and Mn)in the wastewater were utilized efficiently in an environmentally friendly way.Therefore,this study provides a novel method for the coextraction of V and Mn from HMVW.Zishuai Liu Yimin Zhang Zilin Dai Jing Huang Cong Liu 2020Frontiers of Chemical Science and Engineering2020,14,5:1

15Conservative Evolution of Hepatitis B Virus Precore and Core Gene During Immune Tolerant Phase in Intrafamilial Transmission显示文摘Hepatitis B virus(HBV)is characterized with high mutations,which is attributed to the lack of proof-reading of the viral reverse transcriptase and host immune pressure.In this study,31 HBV chronic carriers from 14 families were enrolled to investigate the evolution of the same original HBV sources in different hosts.Sequences of pre-C and C(pre-C/C)genes were analyzed in eight pairs of HBV-infected mothers with longitudinal sera(at an interval of 6.0–7.2 years)and their children(5.5–6.7 years old),and in 15 adults(21–78 years old)from six families with known intrafamilial HBV infection.The pre-C/C sequences had almost no change in eight mothers during 6.0–7.2 years and their children who were in immune tolerant phase.The pre-C/C sequences from the 15 adults of six families,mostly in the immune-clearance phase or the low replicative phase,showed various diversified mutations between individuals from each family.Compared to a reference stain(GQ205441)isolated nearby,the pre-C/C in individuals in immune tolerant phase showed 98.56%–99.52%homology at nucleotide level and 99.5%–100%homology at amino acid level.In contrast,multiple mutations were developed in the immune-clearance phase or the low replicative phase,affecting immune epitopes in core gene and G1896 in pre-C gene.The results indicate that the evolution of new HBV variants is not mainly resulted from the spontaneous error rate of viral reverse transcription,but from the host immune pressure.Yuqian Luo Le Zhang Yimin Dai Yali Hu Biyun Xu Yi-Hua Zhou 2020Virologica Sinica2020,35,4:0
16Canonical Correlation between the Leaf Quality Indicators of 'Moderate Aroma' Flue-cured Tobacco显示文摘In order to find out the correlation between tobacco quality evaluation indicators in China's traditional ' moderate aroma' tobaccoproducing areas and simplify the tobacco quality evaluation indicators,we evaluate the appearance quality and smoking quality of 143 flue-cured tobacco leaf samples in China's ' moderate aroma' tobacco-producing areas,test the physical traits and chemical component,and analyze the canonical correlation between four quality evaluation indicators. The results show that there is significant or extremely significant canonical correlation between four evaluation indicators( tobacco smoking quality,chemical component,appearance quality and physical trait quality); the cumulative variance contribution rate of evaluation indicators is in the order of chemical component( 69. 17%) > appearance quality( 68. 76%) > physical traits( 64. 13%); appearance quality is most closely related to physical traits( 93. 84%). The individual indicators for tobacco quality evaluation make different contribution to the correlation between quality evaluation indicators. The chemical component evaluation indicators mainly include total sugar and ratio of total sugar to betaine; sensory taste indicators mainly include aroma volume,smoke concentration,irritation and softness degree; physical trait evaluation indicators mainly include leaf weight,leaf length and leaf density; appearance quality indicators mainly include leaf organizational structure,color,maturity and identity. 18Simulating Aerosol Optical Depth and Direct Radiative Effects over the Tibetan Plateau with a High-Resolution CAS FGOALS-f3 Model显示文摘Current global climate models cannot resolve the complex topography over the Tibetan Plateau(TP)due to their coarse resolution.This study investigates the impacts of horizontal resolution on simulating aerosol and its direct radiative effect(DRE)over the TP by applying two horizontal resolutions of about 100 km and 25 km to the Chinese Academy of Sciences Flexible Global Ocean-Atmosphere Land System(CAS FGOALS-f3)over a 10-year period.Compared to the AErosol RObotic NETwork observations,a high-resolution model(HRM)can better reproduce the spatial distribution and seasonal cycles of aerosol optical depth(AOD)compared to a low-resolution model(LRM).The HRM bias and RMSE of AOD decreased by 0.08 and 0.12,and the correlation coefficient increased by 0.22 compared to the LRM.An LRM is not sufficient to reproduce the aerosol variations associated with fine-scale topographic forcing,such as in the eastern marginal region of the TP.The difference between hydrophilic aerosols in an HRM and LRM is caused by the divergence of the simulated relative humidity(RH).More reasonable distributions and variations of RH are conducive to simulating hydrophilic aerosols.An increase of the 10-m wind speed in winter by an HRM leads to increased dust emissions.The simulated aerosol DREs at the top of the atmosphere(TOA)and at the surface by the HRM are–0.76 W m^(–2)and–8.72 W m^(–2)over the TP,respectively.Both resolution models can capture the key feature that dust TOA DRE transitions from positive in spring to negative in the other seasons.Min ZHAO Tie DAI Hao WANG Qing BAO Yimin LIU Hua ZHANG Guangyu SHI 2022Advances in Atmospheric Sciences2022,39,12:0
19Na_(2)CO_(3)-responsive Photosynthetic and ROS Scavenging Mechanisms in Chloroplasts of Alkaligrass Revealed by Phosphoproteomics显示文摘Alkali-salinity exerts severe osmotic,ionic,and high-p H stresses to plants.To understand the alkali-salinity responsive mechanisms underlying photosynthetic modulation and reactive oxygen species(ROS)homeostasis,physiological and diverse quantitative proteomics analyses of alkaligrass(Puccinellia tenuiflora)under Na_(2)CO_(3)stress were conducted.In addition,Western blot,real-time PCR,and transgenic techniques were applied to validate the proteomic results and test the functions of the Na_(2)CO_(3)-responsive proteins.A total of 104 and 102 Na_(2)CO_(3)-responsive proteins were identified in leaves and chloroplasts,respectively.In addition,84 Na_(2)CO_(3)-responsive phosphoproteins were identified,including 56 new phosphorylation sites in 56 phosphoproteins from chloroplasts,which are crucial for the regulation of photosynthesis,ion transport,signal transduction,and energy homeostasis.A full-length Pt FBA encoding an alkaligrass chloroplastic fructosebisphosphate aldolase(FBA)was overexpressed in wild-type cells of cyanobacterium Synechocystis sp.Strain PCC 6803,leading to enhanced Na_(2)CO_(3)tolerance.All these results indicate that thermal dissipation,state transition,cyclic electron transport,photorespiration,repair of photosystem(PS)Ⅱ,PSI activity,and ROS homeostasis were altered in response to Na_(2)CO_(3)stress,which help to improve our understanding of the Na_(2)CO_(3)-responsive mechanisms in halophytes.Jinwei Suo Heng Zhang Qi Zhao Nan Zhang Yongxue Zhang Ying Li Baohua Song Juanjuan Yu Jianguo Cao Tai Wang Ji Luo Lihai Guo Jun Ma Xumin Zhang Yimin She Lianwei Peng Weimin Ma Siyi Guo Yuchen Miao Sixue Chen Zhi Qin Shaojun Dai 2020Genomics, Proteomics & Bioinformatics2020,18,3:0
