| 2 | The HDAC inhibitor GCJ-490A suppresses c-Met expression through IKKα and overcomes gefitinib resistance in non-small cell lung cancer显示文摘Objective:The novel compound GCJ-490A has been discovered as a pan-histone deacetylase(HDAC)inhibitor that exerts potent inhibitory activity against HDAC1,HDAC3,and HDAC6.Because of the important roles of HDACs in lung cancer development and the high distribution of GCJ-490A in lung tissue,we explored the anti-tumor potency of GCJ-490A against non-small cell lung cancer(NSCLC)in vitro and in vivo in this study.Methods:The in vitro effects of GCJ-490A alone or combined with the EGFR inhibitor gefitinib against NSCLC were measured with proliferation,apoptosis,and colony formation assays.NSCLC xenograft models were used to investigate the efficacy of GCJ-490A combined with gefitinib for the treatment of NSCLC in vivo.Western blot assays,luciferase reporter assays,chromatin immunoprecipitation assays,quantitative real time-PCR,immunohistochemistry,and transcription factor activity assays were used to elucidate possible mechanisms.Results:GCJ-490A effectively inhibited NSCLC cell proliferation and induced apoptosis in vitro and in vivo.Interestingly,inhibition of HDAC1 and HDAC6 by GCJ-490A increased histone acetylation at the IKKαpromoter and enhanced IKKαtranscription,thus decreasing c-Met.Moreover,this c-Met downregulation was found to be essential for the synergistic anti-tumor activity of GCJ-490A and gefitinib.Conclusions:These findings highlight the promising potential of HDAC inhibitors in NSCLC treatment and provide a rational basis for the application of HDAC inhibitors in combination with EGFR inhibitors in clinical trials. | Ting He Yinglei Gao Yanfen Fang Yangming Zhang Shuwei Zhang Fajun Nan Jian Ding Yi Chen | 2022 | Cancer Biology & Medicine2022,19,8: | 3 |
| 3 | Review of ultraviolet ageing mechanisms and anti-ageing methods for asphalt binders显示文摘Asphalt binder is inevitably aged by ultraviolet(UV)light during its service period.UV ageing can significantly decrease the technical properties of asphalt binder.The sensitivity of asphalt to UV ageing and thermal-oxidative ageing differs,such that the UV ageing performance cannot be determined based on the thermal-oxidative ageing performance.Previous researches mainly focused on the chemical composite and technical performance changes of asphalt binder during UV ageing,and the UV light parameters effect on the ageing rate of asphalt binder.However,the theory for characterizing and explaining the development of UV ageing depth does not get too much attentions,and the UV ageing mechanism of asphalt binder is not very clear.Therefore,it cannot guide to develop or select the good methods or anti-UV ageing additives for asphalt binders.This paper focuses on the latest researches of the mechanisms and anti-ageing methods of asphalt binders.With the increase of UV ageing time,the UV ageing of asphalt binder develops gradually from the surface to inner part.There are various methods,such as low-penetration grade asphalt,less air void ratio,UV stabilizers and UV light absorbers,that can improve the UV ageing resistance of asphalt binders.A new theory of sensitive wavelengths of asphalt UV ageing is proposed,which can enrich the basic theory of asphalt UV ageing.Depending on this theory,different wavelengths of UV light have different ageing effects on asphalt binder.The composite anti-UV ageing additives with barrier and specific absorption effects on UV light is proposed,and may have better improvement effect on the anti-UV ageing performance of asphalt binder. | Yuanyuan Li Jianlin Feng Shaopeng Wu Anqi Chen Dongliang Kuang Tao Bai Yangming Gao Jizhe Zhang Linglin Li Liyao Wan Quantao Liu Zongwu Chen Dengjun Gu | 2022 | Journal of Road Engineering2022,2,2: | 1 |

| 6 | A pan-coronavirus peptide inhibitor prevents SARS-CoV-2 infection in mice by intranasal delivery显示文摘Coronaviruses(Co Vs)have brought serious threats to humans,particularly severe acute respiratory syndrome coronavirus 2(SARS-Co V-2),which continually evolves into multiple variants.These variants,especially Omicron,reportedly escape therapeutic antibodies and vaccines,indicating an urgent need for new antivirals with pan-SARS-Co V-2 inhibitory activity.We previously reported that a peptide fusion inhibitor,P3,targeting heptad repeated-1(HR1)of SARS-Co V-2 spike(S)protein,could inhibit viral infections.Here,we further designed multiple derivatives of the P3 based on structural analysis and found that one derivative,the P315V3,showed the most efficient antiviral activity against SARS-Co V-2 variants and several other sarbecoviruses,as well as other human-Co Vs(HCo Vs).P315V3 also exhibited effective prophylactic efficacy against the SARS-Co V-2 Delta and Omicron variants in mice via intranasal administration.These results suggest that P315V3,which is in PhaseⅡclinical trial,is promising for further development as a nasal pan-SARS-Co V-2 or pan-Co Vs inhibitor to prevent or treat CoV diseases. | Lili Wu Anqi Zheng Yangming Tang Yan Chai Jiantao Chen Lin Cheng Yu Hu Jing Qu Wenwen Lei William Jun Liu Guizhen Wu Shaogui Zeng Hang Yang Qihui Wang George Fu Gao | 2023 | Science China(Life Sciences)2023,66,10: | 0 |
