| 8 | IDH1 fine-tunes cap-dependent translation initiation显示文摘The metabolic enzyme isocitrate dehydrogenase 1(IDH1)catalyzes the oxidative decarboxylation of isocitrate to a-ketoglutarate(a-KG).Its mutation often leads to aberrant gene expression in cancer.IDH1 was reported to bind thousands of RNA transcripts in a sequence-dependent manner;yet,the functional significance of this RNA-binding activity remains elusive.Here,we report that IDH1 promotes mRNA translation via direct associations with polysome mRNA and translation machinery.Comprehensive proteomic analysis in embryonic stem cells(ESCs)revealed strikingenrichmentof ribosomal proteins and translation regulators in IDH1-bound protein interactomes.We performed ribosomal profiling and analyzed mRNA transcripts that are associated with actively translating polysomes.Interestingly,knockout of IDH1 in ESCs led to significant downregulation of polysome-bound mRNA in IDH1 targets and subtle upregulation of ribosome densities at the start codon,indicating inefficient translation initiation upon loss of IDH1.Tethering IDH1 to a luciferase mRNA via the MS2-MBP system promotes luciferase translation,independently of the catalytic activity of IDH1.Intriguingly,IDH1 fails to enhance luciferase translation driven by an internal ribosome entry site.Together,these results reveal an unforeseen role of IDH1 in fine-tuning cap-dependent translation via the initiation step. | Lichao Liu JYuyang Lu Fajin Li Xudong Xing Tong Li Xuerui Yang Xiaohua Shen | 2019 | Journal of Molecular Cell Biology2019,11,10: | 2 |
| 9 | Oxygen dependent oxidation of trimethoprim by sulfate radical:Kinetic and mechanistic investigations显示文摘Trimethoprim(TMP)is a typical antibiotic to treat infectious disease,which is among the most commonly detected antibacterial agents in natural waters and municipal wastewaters.In the present study,the impacts of dissolved oxygen(DO)on the oxidation efficiency and pathways of TMP by reaction with sulfate radicals(SO_(4)^(-1))were investigated.Our results revealed that the presence of DO was favourable for TMP degradation.Specifically,TMP would react initially with SO_(4)^(-1)via electron-transfer process to form a carbon-centered radical.In the absence of oxygen,the carbon-centered radical could undergo hydrolysis to produceα-hydroxytrimethoprim(TMPàOH),followed by the further oxidation which generatedα-ketotrimethoprim(TMP=O).However,in the presence of oxygen,the carbon-centered radical would alternatively combine with oxygen,leading to a sequential reaction in which peroxyl radical and a tetroxide were formed,and finally generated TMPàOH and TMP=O simultaneously.The proposed pathways were further confirmed by density functional theory(DFT)calculations.The results obtained in this study would emphasize the significance of DO on the oxidation of organic micropollutants by SO_(4)^(-1). | Xuerui Yang Xi Ding Lei Zhou Yuefei Ji Guangli Xiu | 2021 | Chinese Chemical Letters2021,32,10: | 1 |

| 15 | Comparison of the Arctic upper-air temperatures from radiosonde and radio occultation observations显示文摘The air temperature is one of the most important parameters used for monitoring the Arctic climate change. The constellation observing system for meteorology, ionosphere, and climate and Formosa Satellite Mission 3(COSMIC/FORMOSAT-3) radio occultation(RO) 'wet' temperature product(i.e., 'wet Prf') is used to analyze the Arctic air temperature profiles at 925–200 hPa in 2007–2012. The 'wet' temperatures are further compared with radiosonde(RS) observations. The results from the spatially and temporally synchronized RS and COSMIC observations show that their temperatures agree well with each other, especially at 400 hPa. Comparisons of seasonal temperatures and anomalies from the COSMIC and homogenized RS observations suggest that the limited number of COSMIC observations during the spatial matchup may be insufficient to describe the smallscale spatial structure of temperature variations. Furthermore, comparisons of the seasonal temperature anomalies from the RS and 5°×5° gridded COSMIC observations at 400 hPa during the sea ice minimum(SIM) of2007 and 2012 are also made. The results reveal that similar Arctic temperature variation patterns can be obtained from both RS and COSMIC observations over the land area, while extra information can be further provided from the densely distributed COSMIC observations. | 17 | Ribosome Profiling Reveals Genome-wide Cellular Translational Regulation upon Heat Stress in Escherichia coli显示文摘Heat shock response is a classical stress-induced regulatory system in bacteria, characterized by extensive transcriptional reprogramming. To compare the impact of heat stress on the transcriptome and translatome in Escherichia coli, we conducted ribosome profiling in parallel with RNA-Seq to investigate the alterations in transcription and translation efficiency when E. coli cells were exposed to a mild heat stress(from 30 °C to 45 °C). While general changes in ribosome footprints correlate with the changes of mRNA transcripts upon heat stress, a number of genes show differential changes at the transcription and translation levels. Translation efficiency of a few genes that are related to environment stimulus response is up-regulated, and in contrast, some genes functioning in mRNA translation and amino acid biosynthesis are down-regulated at the translation level in response to heat stress. Moreover, our ribosome occupancy data suggest that in general ribosomes accumulate remarkably in the starting regions of ORFs upon heat stress. | 18 | Fluid Analysis and Structure Optimization of Impeller Based on Surrogate Model显示文摘The surrogate model technology has a good performance in solving black-box optimization problems,which is widely used in multi-domain engineering optimization problems.The adaptive surrogate model is the mainstream research direction of surrogate model technology,which can realize model fitting and global optimization of engineering problems by infilling criteria.Based on the idea of the adaptive surrogate model,this paper proposes an efficient global optimization algorithm based on the local remodeling method(EGO-LR),which aims at improving the accuracy and optimization efficiency of the model.The proposed algorithm firstly constructs the expectation improvement(EI)function in the local area and optimizes it to get the update points.Secondly,the obtained update points are added to the global region until the global accuracy of the model meets the requirements.Then the differential evolution algorithm is used for global optimization.Sixteen benchmark functions are used to compare the EGO-LR algorithm with the existing algorithms.The results show that the EGO-LR algorithm can quickly converge to the accuracy requirements of the model and find the optimal value efficiently when facing complex problems with many local extrema and large variable spaces.The proposed algorithm is applied to the optimization design of the structural parameter of the impeller,and the outflow field analysis of the impeller is realized through finite element analysis.The optimization with the maximum fluid pressure(MP value)of the impeller as the objective function is completed,which effectively reduces the pressure value of the impeller under load. | Huanwei Xu Wenzhang Wei Hanjin He Xuerui Yang | 2022 | Computer Modeling in Engineering & Sciences2022,,7: | 0 |
