| 2 | A structural and data-driven approach to engineering a plant cytochrome P450 enzyme显示文摘Functional manipulation of biosynthetic enzymes such as cytochrome P450 s(or P450 s) has attracted great interest in metabolic engineering of plant natural products.Cucurbitacins and mogrosides are plant triterpenoids that share the same backbone but display contrasting bioactivities.This structural and functional diversity of the two metabolites can be manipulated by engineering P450 s.However,the functional redesign of P450 s through directed evolution(DE) or structure-guided protein engineering is time consuming and challenging,often because of a lack of high-throughput screening methods and crystal structures of P450 s.In this study,we used an integrated approach combining computational protein design,evolutionary information,and experimental data-driven optimization to alter the substrate specificity of a multifunctional P450(CYP87 D20)from cucumber.After three rounds of iterative design and evaluation of 96 protein variants,CYP87 D20,which is involved in the cucurbitacin C biosynthetic pathway,was successfully transformed into a P450 mono-oxygenase that performs a single specific hydroxylation at C11 of cucurbitadienol.This integrated P450-engineering approach can be further applied to create a de novo pathway to produce mogrol,the precursor of the natural sweetener mogroside,or to alter the structural diversity of plant triterpenoids by functionally manipulating other P450 s. | Dawei Li Yongshuo Ma Yuan Zhou Junbo Gou Yang Zhong Lingling Zhao Lida Han Sergey Ovchinnikov Ling Ma Sanwen Huang Per Greisen Yi Shang | 2019 | Science China(Life Sciences)2019,62,7: | 5 |
| 3 | Preparation and characterization of nanostructured Al_2O_3-13wt.%TiO_2 ceramic coatings by plasma spraying显示文摘Nanostructured and conventional Al2O3-13wt.%TiO2 ceramic coatings were prepared by plasma spraying with nanostructured agglomerated and conventional powders, respectively. The microstructure and microhardness of the coatings were investigated using scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and microhardness measurement. Meanwhile, the friction and wear behaviors were analyzed and compared using a ball-on-disk tribometer. The results show that the conventional coating has lamellar stacking characteristic and has some pores. However, the nanostructured coating shows a bimodal microstructure, which is composed of both fully melted regions and partially melted regions. According to the microstructural difference, the partially melted regions can be divided into liquid-phase sintered regions (a three-dimensional net or skeleton-like structure: Al2O3-rich submicron particles embedded in the TiO2-rich matrix) and solid-phase sintered regions (remained nanoparticles). The microstructural characteristics of the liquid-phase sintered region are formed due to the selective melting of TiO2 nanoparticles during plasma spraying. On the other hand, the TiO2 and Al2O3 nanoparticles of the solid-phase sintered regions are all unmelted during plasma spraying. | 5 | Enhanced chemoselectivity of a plant cytochrome P450 through protein engineering of surface and catalytic residues显示文摘Cytochrome P450s(P450s)are the most versatile catalysts utilized by plants to produce structurally and functionally diverse metabolites.Given the high degree of gene redundancy and challenge to functionally characterize plant P450s,protein engineering is used as a complementarystrategy to study the mechanisms of P450-mediated reactions,or to alter their functions.We previously proposed an approach of engineering plant P450s based on combining high accuracy homology models generated by Rosetta combined with data-driven design using evoluti onary information of these enzymes.With this strategy,we repurposed a multi-functional P450(CYP87D20)into a monooxygenase after red esigning its active site.Since most plant P450s are membrane-anchored proteins that are adapted to the micro-environments of plant cells,expressing them in heterologous hosts usually results in problems of expression or activity.Here,we applied compu-tational design to tackle these issues by simultaneous optimization of the protein surface and active site.After screening 17 variants,effective su bstitutions of surface residues were observed to improve both expression and activity of CYP87D20.In addition,the identified substitutions were additive and by com-bining them a highly eficient C11 hydroxylase of cucurbitadienol was created to participate in the mogrol biosynthesis.This study shows the importance of considering the interplay between surface and active site residues for P450 engineering.Our integrated strategy also opens an avenue to create more tai loring enzymes with desired functions for the metabolic engineering of high-valued compounds like mogrol,the precursor of natural sweetener mogrosides. | Xiaopeng Zhang Wei Luo Yinying Yao Xuming Luo Chao Han Yang Zhong Bo Zhang Dawei Li Lida Han Sanwen Huang Per Greisen Yi Shang | 2021 | aBIOTECH2021,2,3: | 1 |

| 13 | Reasoning Disaster Chains with Bayesian Network Estimated Under Expert Prior Knowledge显示文摘With the acceleration of global climate change and urbanization,disaster chains are always connected to artificial systems like critical infrastructure.The complexity and uncertainty of the disaster chain development process and the severity of the consequences have brought great challenges to emergency decision makers.The Bayesian network(BN)was applied in this study to reason about disaster chain scenarios to support the choice of appropriate response strategies.To capture the interacting relationships among different factors,a scenario representation model of disaster chains was developed,followed by the determination of the BN structure.In deriving the conditional probability tables of the BN model,we found that,due to the lack of data and the significant uncertainty of disaster chains,parameter learning methodologies based on data or expert knowledge alone are insufficient.By integrating both sample data and expert knowledge with the maximum entropy principle,we proposed a parameter estimation algorithm under expert prior knowledge(PEUK).Taking the rainstorm disaster chain as an example,we demonstrated the superiority of the PEUK-built BN model over the traditional maximum a posterior(MAP)algorithm and the direct expert opinion elicitation method.The results also demonstrate the potential of our BN scenario reasoning paradigm to assist real-world disaster decisions. | Lida Huang Tao Chen Qing Deng Yuli Zhou | 2023 | International Journal of Disaster Risk Science2023,14,6: | 0 |
| 14 | Research on the security of China's oil resources supply based on the objective weight method显示文摘With the rapid development of China's economy,external dependence on petroleum resources continues to increase,and their security has become an important part of national security.To evaluate the security of China's petroleum resource supply in a scientific and objective manner,this study establishes a corresponding petroleum life-cycle evaluation index system,based on the theory and method of the whole life-cycle security evaluation of mineral resources,and conducts further independence and grey correlation analysis on the indexes for the purpose of evaluating the petroleum risk situation in China,based on relevant public data from the past 10 years.The results show that the overall trend of China's oil risk has a“U”-shaped characteristic of first decreasing and then increasing.Furthermore,the analysis finds that China's mineral resources have been greatly influenced by the domestic production situation and international trade.These results suggest that the security of petroleum supply can be improved by safeguarding international trade in petroleum resources,strengthening the strategic reserves of domestic petroleum resources,and developing new alternative clean energy sources to improve the resilience of petroleum supply security.This study's research methodology is more logical and systematic than traditional methods,and the analysis of the factors is comprehensive and of high application value,providing implications for the establishment of a big data analysis and evaluation index system for oil resource security. | Yuli Zhou Ming Wang Hongyong Yuan Lida Huang | 2023 | Journal of Safety Science and Resilience2023,4,3: | 0 |
