1Data-Driven Discovery in Mineralogy: Recent Advances in Data Resources, Analysis, and Visualization显示文摘Large and growing data resources on the diversity, distribution, and properties of minerals are ushering in a new era of data-driven discovery in mineralogy. The most comprehensive international mineral database is the IMA database, which includes information on more than 5400 approved mineral species and their properties, and the mindat.org data source, which contains more than 1 million species/locality data on minerals found at more than 300 000 localities. Analysis and visualization of these data with diverse techniques—including chord diagrams, cluster diagrams, Klee diagrams, skyline diagrams, and varied methods of network analysis—are leading to a greater understanding of the co-evolving geosphere and biosphere. New data-driven approaches include mineral evolution, mineral ecology, and mineral network analysis—methods that collectively consider the distribution and diversity of minerals through space and time. These strategies are fostering a deeper understanding of mineral co-occurrences and, for the first time, facilitating predictions of mineral species that occur on Earth but have yet to be discovered and described.Robert M. 4The WNT1^(G177C) mutation specifically affects skeletal integrity in a mouse model of osteogenesis imperfecta type XV显示文摘The recent identification of homozygous WNT1 mutations in individuals with osteogenesis imperfecta type XV(OI-XV)has suggested that WNT1 is a key ligand promoting the differentiation and function of bone-forming osteoblasts.Although such aninfluence was supported by subsequent studies,a mouse model of OI-XV remained to be established.Therefore,we introduced a previously identified disease-causing mutation(G177C)into the murine Wnt1 gene.Homozygous Wnt1^(G177C/G177C)mice were viable and did not display defects in brain development,but the majority of 24-week-old Wnt1^(G177C/G177C)mice had skeletal fractures.This increased bone fragility was not fully explained by reduced bone mass but also by impaired bone matrix quality.Importantly,the homozygous presence of the G177C mutation did not interfere with the osteoanabolic influence of either parathyroid hormone injection or activating mutation of LRP5,the latter mimicking the effect of sclerostin neutralization.Finally,transcriptomic analyses revealed that short-term administration of WNT1 to osteogenic cells induced not only the expression of canonical WNT signaling targets but also the expression of genes encoding extracellular matrix modifiers.Taken together,our data demonstrate that regulating bone matrix quality is a primary function of WNT1.They further suggest that individuals with WNT1 mutations should profit from existing osteoanabolic therapies.Nele Vollersen Wenbo Zhao Tim Rolvien Fabiola Lange Felix Nikolai Schmidt Stephan Sonntag Doron Shmerling Simon von Kroge Kilian Elia Stockhausen Ahmed Sharaf Michaela Schweizer Meliha Karsak Bjorn Busse Ernesto Bockamp Oliver Semler Michael Amling Ralf Oheim Thorsten Schinke Timur Alexander Yorgan 2021Bone Research2021,9,4:1

15LINGO-1 and AMIGO3, potential therapeutic targets for neurological and dysmyelinating disorders?显示文摘Leucine rich repeat proteins have gained considerable interest as therapeutic targets due to their expression and biological activity within the central nervous system. LINGO-1 has received particular attention since it inhibits axonal regeneration after spinal cord injury in a Rho A dependent manner while inhibiting leucine rich repeat and immunoglobulin-like domain-containing protein 1(LINGO-1) disinhibits neuron outgrowth. Furthermore, LINGO-1 suppresses oligodendrocyte precursor cell maturation and myelin production. Inhibiting the action of LINGO-1 encourages remyelination both in vitro and in vivo. Accordingly, LINGO-1 antagonists show promise as therapies for demyelinating diseases. An analogous protein to LINGO-1, amphoterin-induced gene and open reading frame-3(AMIGO3), exerts the same inhibitory effect on the axonal outgrowth of central nervous system neurons, as well as interacting with the same receptors as LINGO-1. However, AMIGO3 is upregulated more rapidly after spinal cord injury than LINGO-1. We speculate that AMIGO3 has a similar inhibitory effect on oligodendrocyte precursor cell maturation and myelin production as with axogenesis.
