| 1 | Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis. | TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang | 2016 | Biomedical and Environmental Sciences2016，29，12： | 21 |
| 2 | Wilson disease: Identification of two novel mutations and clinical correlation in Eastern Chinese patients显示文摘AIM: To study mutations in the P-type ATPase (ATP7B) gene responsible for Wilson disease (WD) in the Eastern Chinese population, and the possible correlation of specific mutations with clinical characteristics. METHODS: Mutations of the ATP7B gene were sought by means of direct sequencing in 50 Eastern Chinese WD patients of Han ethnic origin. RESULTS: Two novel mutations, Asp96Gly and Asp196Glu, were first identified. We also compared the characterization of mutations in ATP7B with the clinical findings, and a significant correlation with hepatic manifestations between patients carrying the Arg778Leu mutation and those without was found. CONCLUSION: Gene sequencing analysis was shown to have a high detection rate and accuracy. | 3 | Interactions and Biological Activity of Rare Earth Perchlorate Complexes with Alanine and Imidazole显示文摘The interactions of nine rare earth perchlorate complexes with alanine and imidazole were studied in methanol. The general formulae of these compounds are RE(Ala) 2Im(ClO 4) 3· n H 2O(RE=Ce, Pr, Nd, Sm, Eu, Gd, Tb, Yb) and La(Ala) 3Im(ClO 4) 3·H 2O, respectively. The composition and structure of the prepared compounds were determined by the chemical analysis, elemental analysis, IR spectroscopy, UV, FS spectroscopy, X ray powder diffraction, 1H NMR and solubility. The thermal decomposition processes of these complexes were studied on the basis of TG DTG. Syngenetic brine sedimentation, hot submarine spring eruption—accumulation, syngenetic sedimentation—vadose brine replacement had been supposed as the genetic models before. Under these models, syngenetic mineralization should be dominant in ore formation. However, a series of geological and geochemical properties investigated by authors of this paper are contrary to these models.
