2Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5
3Bibliometric analysis and system review of vehicle routing optimization for emergency material distribution显示文摘Determining the optimal vehicle routing of emergency material distribution(VREMD)is one of the core issues of emergency management,which is strategically important to improve the effectiveness of emergency response and thus reduce the negative impact of large-scale emergency events.To summarize the latest research progress,we collected 511VREMD-related articles published from 2010 to the present from the Scopus database and conducted a bibliometric analysis using VOSviewer software.Subsequently,we cautiously selected 49 articles from these publications for system review;sorted out the latest research progress in model construction and solution algorithms;and summarized the evolution trend of keywords,research gaps,and future works.The results show that domestic scholars and research organizations held an unqualified advantage regarding the number of published papers.However,these organizations with the most publications performed poorly regarding the number of literature citations.China and the US have contributed the vast majority of the literature,and there are close collaborations between researchers from both countries.The optimization model of VREMD can be divided into single-,multi-,and joint-objective models.The shortest travel time is the most common optimization objective in the single-objective optimization model.Several scholars focus on multiobjective optimization models to consider conflicting objectives simultaneously.In recent literature,scholars have focused on the impact of uncertainty and special events(e.g.,COVID-19)on VREMD.Moreover,some scholars focus on joint optimization models to optimize vehicle routes and central locations(or material allocation)simultaneously.Solution algorithms can be divided into two primary categories,i.e.,mathematical planning methods and intelligent evolutionary algorithms.The branch and bound algorithm is the most dominant mathematical planning algorithm,while genetic algorithms and their enhancements are the most commonly used intelligent evolutionary algorithms.It is shown that the nondominated sorting genetic algorithmⅡ(NSGA-Ⅱ)can effectively solve the multiobjective model of VREMD.To further improve the algorithm’s performance,researchers have proposed improved hybrid intelligent algorithms that combine the advantages of NSGA-Ⅱand certain other algorithms.Scholars have also proposed a series of optimization algorithms for specific scenarios.With the development of new technologies and computation methods,it will be exciting to construct optimization models that consider uncertainty,heterogeneity,and temporality for large-scale real-world issues and develop generalized solution approaches rather than those applicable to specific scenarios.Jinxing Shen Kun Liu Changxi Ma Yongpeng Zhao Chuwei Shi 2022Journal of Traffic and Transportation Engineering(English Edition)2022,9,6:5
4Admissibility analysis and control synthesis for switched linear singular systems显示文摘The problem of admissibility analysis and control synthesis of discrete-time switched linear singular (SLS) systems for arbitrary switching laws is solved.By using the switched Lyapunov function approach,some new sufficient conditions under which the SLS system is admissible for arbitrary switching laws are derived in terms of linear matrix inequalities(LMIs).Based on the admissibility results,control synthesis is then to design switched state feedback and static output feedback controllers,guaranteeing that the resulting closed-loop system is admissible.The presented results can be viewed as the extensions of previous works on switched Lyapunov function approach from the regular switched systems to singular switched cases.Examples are provided to demonstrate the reduced conservatism and effectiveness of the proposed conditions.Lin Jinxing Fei Shumin Shen Jiong 2009Journal of Systems Engineering and Electronics2009,20,5:4
5Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. 7Robust stability and stabilization for uncertain discrete-time switched singular systems with time-varying delays显示文摘The problems of robust stability and stabilization via memoryless state feedback for a class of discrete-time switched singular systems with time-varying delays and linear fractional uncertainties are investigated.By constructing a novel switched Lyapunov-Krasovskii functional,a delay-dependent criterion for the unforced system to be regular,causal and uniformly asymptotically stable is established in terms of linear matrix inequalities(LMIs).An explicit expression for the desired memoryless state feedback stabilization controller is also given.The merits of the proposed criteria lie in their less conservativeness and relative simplicity,which are achieved by considering additionally useful terms(ignored in previous methods) when estimating the upper bound of the forward difference of the Lyapunov-Krasovskii functional and by avoiding utilizing any model augmentation transformation.Some numerical examples are provided to illustrate the validity of the proposed methods.Jinxing Lin Shumin Fei Jiong Shen 2010Journal of Systems Engineering and Electronics2010,21,4:2
8The Brassica napus GATA transcription factor BnA5.ZML1 is a stigma compatibility factor显示文摘Self-incompatibility(SI)is a genetic mechanism that rejects self-pollen and thus prevents inbreeding in some hermaphroditic angiosperms.In the Brassicaceae,SI involves a pollen-stigma recognition system controlled by a single locus known as the S locus,which consists of two highly polymorphic genes that encode S-locus cysteine-rich protein(SCR)and S-receptor kinase(SRK).When self-pollen lands on the stigma,the S-haplotype-specific interaction between SCR and SRK triggers SI.Here,we show that the GATA transcription factor BnA5.ZML1 suppresses SI responses in Brassica napus and is induced after compatible pollination.The loss-of-function mutant bna5.zml1 displays reduced self-compatibility.In contrast,overexpression of BnA5.ZML1 in self-incompatible stigmas leads to a partial breakdown of SI responses,suggesting that BnA5.ZML1 is a stigmatic compatibility factor.Furthermore,the expression levels of SRK and ARC1 are up-regulated in bna5.zml1 mutants,and they are down-regulated in BnA5.ZML1 overexpressing lines.SRK affects the cellular localization of BnA5.ZML1 through direct protein–protein interaction.Overall,our findings highlight the fundamental role of BnA5.ZML1 in SI responses in B.napus,establishing a direct interaction between BnA5.ZML1 and SRK in this process.Zhiqiang Duan Yatao Zhang Jinxing Tu Jinxiong Shen Bin Yi Tingdong Fu Cheng Dai Chaozhi Ma 2020Journal of Integrative Plant Biology2020,62,8:2

10Global COVID-19 Pandemic Waves: Limited Lessons Learned Worldwide over the Past Year显示文摘The occurrence of coronavirus disease 2019(COVID-19)was followed by a small burst of cases around the world;afterward,due to a series of emergency non-pharmaceutical interventions(NPIs),the increas-ing number of confirmed cases slowed down in many countries.However,the lifting of control measures by the government and the public’s loosening of precautionary behaviors led to a sudden increase in cases,arousing deep concern across the globe.arousing deep concern across the globe.This study evaluates the situation of the COVID-19 pandemic in countries and territories worldwide from January 2020 to February 2021.According to the time-varying reproduction number(R(t))of each country or territory,the results show that almost half of the countries and territories in the world have never controlled the epidemic.Among the countries and territories that had once contained the occurrence,nearly half failed to maintain their prevention and control,causing the COVID-19 pandemic to rebound across the world—resulting in even higher waves in half of the rebounding countries or territories.This work also proposes and uses a time-varying country-level transmission risk score(CTRS),which takes into account both R(t)and daily new cases,to demonstrate country-level or territory-level transmission potential and trends.Time-varying hierarchical clustering of time-varying CTRS values was used to successfully reveal the countries and territories that contributed to the recent aggravation of the global pandemic in the last quarter of 2020 and the beginning of 2021,and to identify countries and territories with an increasing risk of COVID-19 transmission in the near future.Furthermore,a regression analysis indicated that the introduction and relaxation of NPIs,including workplace closure policies and stay-at-home requirements,appear to be associated with recent global transmission changes.In conclu-sion,a systematic evaluation of the global COVID-19 pandemic over the past year indicates that the world is now in an unexpected situation,with limited lessons learned.Summarizing the lessons learned could help in designing effective public responses for constraining future waves of COVID-19 worldwide.Yongyue Wei Jinxing Guan Xiao Ning Yuelin Li Liangmin Wei Sipeng Shen Ruyang Zhang Yang Zhao Hongbing Shen Feng Chen 2022Engineering2022,8,6:1
11Bn.YCO affects chloroplast development in Brassica napus L.显示文摘Mature chloroplasts,as the main sites of photosynthesis,are essential for seedling growth in higher plants.Loss of function of genes involved in chloroplast development changes plant phenotype.We obtained a YELLOW COTYLEDON (YCO) mutant in rapeseed (Brassica napus L.) using CRISPR-Cas9.Bn.YCO,a gene of unknown function,has two homologous copies (Bna A01.YCO and Bna C01.YCO) in B.napus.Homozygous mutation of these two homologs resulted in yellow cotyledons and chlorotic true leaves.Transmission electron microscopy revealed that the formation of thylakoid membranes was inhibited in yellow cotyledons.Sequence similarity search revealed that YCO was conserved in different species,and a subcellular location assay verified that Bn.YCO was located in the chloroplast.Bn.YCO was expressed in multiple tissues,most highly in cotyledons.Knockout of Bn.YCO blocked the transcription of plastid genes,especially those of photosystem genes transcribed by plastid-encoded polymerase.Transcriptome sequencing showed that the majority of genes involved in ribosome assembly and photosynthesis were down-regulated in Bn.yco mutants.These results suggested that loss of function of Bn.YCO affected plastid gene transcription,which influenced chloroplast biogenesis in rapeseed seedlings.Tingting Liu Baolong Tao Hanfei Wu Jing Wen Bin Yi Chaozhi Ma Jinxing Tu Tingdong Fu Lixia Zhu Jinxiong Shen 2021The Crop Journal2021,9,5:1

19Autonomous bus services:current research status and future recommendations显示文摘Implementing autonomous bus services in several cities has garnered substantial research attention worldwide.However,the benefits and challenges of this emerging mode remain insufficiently understood.Consequently,VOSviewer was employed for a bibliometric analysis involving 300 publications,investigating the associations among authors,journals,and keywords.Subsequently,we comprehensively reviewed the current state of research on two topics and proposed future recommendations.Results indicate that the first document related to autonomous bus services was published in 2009.Most user attitude-related research data are obtained via questionnaires and analyzed using statistical techniques.Autonomous bus services are expected to benefit passengers regarding travel time,cost,safety,etc.,while passenger preferences are inconsistent.However,integrating the service into existing bus systems requires careful consideration of the schedule sequences.Notably,modular autonomous bus services present a new opportunity for the further optimization of bus services.In future studies,standardized data acquisition procedures should be developed to achieve comparable results.Regarding traveler choice behavior,the effect of specific autonomous bus service policies over time and the heterogeneity due to cultural or social contexts across regions should be assessed.To further promote autonomous bus services,based on fluctuating travel demands,the effects of vehicle capacity,speed,and cost of fleet composition should be evaluated comprehensively to optimize the bus network and schedule sequence.Owing to the protracted nature of the transition from conventional to fully autonomous buses,one should prioritize semi-autonomous bus services.Another essential future research direction is to integrate modular autonomous bus assembly or disassembly strategies with different fine-grained operation optimization techniques in various scenarios.Jinxing Shen Qinxin Liu Zi Ye Wenfeng Jiang Changxi Ma 2023Digital Transportation and Safety2023,2,3:1
