1Bifunctional chimeric SuperCD suicide gene -YCD: YUPRT fusion is highly effective in a rat hepatoma model显示文摘AIM: To investigate the effects of catalytically superior gene-directed enzyme prodrug therapy systems on a rat hepatoma model.METHODS: To increase hepatoma cell chemosensitivity for the prodrug 5-fluorocytosine (5-FC), we generated a chimeric bifunctional SuperCD suicide gene, a fusion of the yeast cytosine deaminase (YCD) and the yeast uracil phosphoribosyltransferase (YUPRT) gene.RESULTS: In vitro stably transduced Morris rat hepatoma cells (MH) expressing the bifunctional SuperCD suicide gene (MH SuperCD) showed a clearly marked enhancement in cell killing when incubated with 5-FC as compared with MH ceils stably expressing YCD solely (MH YCD) or the cytosine deaminase gene of bacterial origin(MH BCD), respectively. In vivo, MH SuperCD tumors implanted both subcutaneously as well as orthotopically into the livers of syngeneic ACI rats demonstrated significant tumor regressions (P<0.01) under both high dose as well as low dose systemic 5-FC application,whereas MH tumors without transgene expression (MH naive) showed rapid progression. 5Porcine model characterizing various parameters assessing the outcome after acetaminophen intoxication induced acute liver failure显示文摘AIM To investigate the changes of hemodynamic and laboratory parameters during the course of acute liver failure following acetaminophen overdose.METHODS Eight pigs underwent a midline laparotomy following jejunal catheter placement for further acetaminophen intoxication and positioning of a portal vein Doppler flow-probe. Acute liver failure was realized by intrajejunal acetaminophen administration in six animals, two animals were sham operated. All animals were invasively monitored and received standardized intensive care support throughout the study. Portal blood flow, hemodynamic and ventilation parameters were continuously recorded. Laboratory parameters were analysed every eight hours. Liver biopsies were sampled every 24 h following intoxication and upon autopsy.RESULTS Acute liver failure (ALF) occurred after 28 ± 5 h resulted in multiple organ failure and death despite maximal support after further 21 ± 1 h (study end). Portal blood flow (baseline 1100 ± 156 m L/min) increased to a maximum flow of 1873 ± 175 m L/min at manifestation of ALF, which was significantly elevated(P < 0.01). Immediately after peaking, portal flow declined rapidly to 283 ± 135 m L/min at study end. Thrombocyte values (baseline 307 × 103/μL± 34 × 103/μL) of intoxicated animals declined slowly to values of 145 × 103/μL± 46 × 103/μL when liver failure occurred. 8FITC-linked Fibrin-Binding Peptide and real-time live confocal microscopy as a novel tool to visualize fibrin(ogen)in coagulation显示文摘Background and Aim:Although fibrinogen has been established as a key player in the process of coagulation,many questions about the role of fibrinogen under specific conditions remain.Confocal microscopic assessment of clot formation,and in particular the role that fibrinogen plays in this process,is commonly investigated using pre-labeled fibrinogen.This has a number of drawbacks,primarily that it is impossible to stain fibrin networks after their formation.The aim of the present study is to present an alternative for conveniently post-staining fibrin in a wide range of models/situations,in real time and with high resolution.Methods:We combined a peptide known to bind fibrin and linked it to fluorescein isothiocyanate(FITC),creating the FITC-Fibrin-Binding Peptide(FFBP).We subsequently tested its fibrin-binding capability in vitro under static conditions,as well as under simulated flow,using real-time live confocal microscopy.Results:Fibrin stained with FFBP overlaps with fibrin stained with fibrinogen pre-labeled with Alexa Fluor 647 following coagulation induction.In contrast to pre-labeled fibrinogen,FFBP also stains already formed fibrin networks.By combining FFBP with real-time live confocal microscopy even the visualization of single fibrin fibers is possible.Conclusions:These data indicate that FFBP is a valid and valuable tool for real-time live confocal assessment of clot formation.Relevance for patients:Our findings present a valuable alternative for the visualization of fibrin even after its formation,and we believe this approach will be particularly valuable for future investigations of important,but previously overlooked,aspects of clot formation.Nikolaj Weiss Bettina Schenk Mirjam Bachler Cristina Solomon Dietmar Fries Martin Hermann 2017Journal of Clinical & Translational Research2017,3,2:1
9Algorithm-based arterial blood sampling recognition increasing safety in point-of-care diagnostics显示文摘AIM To detect blood withdrawal for patients with arterial blood pressure monitoring to increase patient safety and provide better sample dating.METHODS Blood pressure information obtained from a patient monitor was fed as a real-time data stream to an experimental medical framework. This framework was connected to an analytical application which observes changes in systolic, diastolic and mean pressure to determine anomalies in the continuous data stream. Detection was based on an increased mean blood pressure caused by the closing of the withdrawal three-way tap and an absence of systolic and diastolic measurements during this manipulation. For evaluation of the proposed algorithm, measured data from animal studies in healthy pigs were used.RESULTS Using this novel approach for processing real-time measurement data of arterial pressure monitoring, the exact time of blood withdrawal could be successfully detected retrospectively and in real-time. The algorithm was able to detect 422 of 434(97%) blood withdrawals for blood gas analysis in the retrospective analysis of 7 study trials. Additionally, 64 sampling events for other procedures like laboratory and activated clotting time analyses were detected.
