| 1 | Injectable dual glucose-responsive hydrogel-micelle composite for mimicking physiological basal and prandial insulin delivery显示文摘For type 1 and advanced type 2 diabetic patients, insulin replacement therapy with simulating on-demand prandial and basal insulin secretion is the best option for optimal glycemic control. However, there is no insulin delivery system yet could mimic both controlled basal insulin release and rapid prandial insulin release in response to real-time blood glucose changes. Here we reported an artificial insulin delivery system, mimicking physiological basal and prandial insulin secretion, to achieve real-time glycemic control and reduce risk of hypoglycemia. A phenylboronic acid(PBA)/galactosyl-based glucose-responsive insulin delivery system was prepared with insulin-loaded micelles embedded in hydrogel matrix. At the hyperglycemic state, both the hydrogel and micelles could swell and achieve rapid glucose-responsive release of insulin, mimicking prandial insulin secretion.When the glucose level returned to the normal state, only the micelles partially responded to glucose and still released insulin gradually. The hydrogel with increased crosslinking density could slow down the diffusion speed of insulin inside, resulting in controlled release of insulin and simulating physiological basal insulin secretion. This hydrogel-micelle composite insulin delivery system could quickly reduce the blood glucose level in a mouse model of type 1 diabetes, and maintain normal blood glucose level without hypoglycemia for about 24 h. | 2 | Features of the Early Palaeozoic Mantle beneath Langao County and Its Formation Mechanism显示文摘Phlogopite-amphibole pyroxenite xenoliths contained in an Early Palaeozoic alkali subvolcanic lam-prophyre complex in Langao County, Shaanxi Province, are metasomatized mantle xenoliths, composed mainly of clinopyroxene, amphibole, phlogopite, apatite, pervoskite, ilmenite and sphene with well-developed subsolidus metamorphism-deformation textures, such as 'triple points' and 'cataclastic boundaries' . Minerological studies indicate that clinopyroxene is rich in SiO2 and MgO and poor in TiO2 and Al2O3, which is notably different from magmatogenic deep-seated megacrysts and phenocrysts formed in the range of mantle pressure. Amphibole and phlogopite have the compositional feature of mantle-derived amphibole and phlogopite. Sm-Nd isotope studies suggest that the metasomatized mantle beneath Langao County is the product of metasomatism of primitive mantle by melt (fluid) derived from the mantle plume, and the mantle metasomatism occurred 650 Ma ago. | 3 | In-biofilm generation of nitric oxide using a magnetically-targetable cascade-reaction container for eradication of infectious biofilms显示文摘Cascade-reaction chemistry can generate reactive-oxygen-species that can be used for the eradication of infectious biofilms.However,suitable and sufficient oxygen sources are not always available near an infection site,while the reactive-oxygen-species generated are short-lived.Therefore,we developed a magnetic cascade-reaction container composed of mesoporous Fe_(3)O_(4)@SiO_(2) nanoparticles containing glucose-oxidase and L-arginine for generation of reactive-oxygen-species.Glucose-oxidase was conjugated with APTES facilitating coupling to Fe_(3)O_(4)@SiO_(2) nanoparticles and generation of H_(2)O_(2) from glucose.L-arginine was loaded into the nanoparticles to generate NO from the H_(2)O_(2) generated.Using an externally-applied magnetic field,cascade-reaction containers could be homogeneously distributed across the depth of an infectious biofilm.Cascade-reaction containers with coupled glucose-oxidase were effective in killing planktonic,Gram-positive and Gram-negative bacteria.Additional efficacy of the L-arginine based second cascade-reaction was only observed when H_(2)O_(2) as well as NO were generated in-biofilm.In vivo accumulation of cascade-reaction containers inside abdominal Staphylococcus aureus biofilms upon magnetic targeting was observed real-time in living mice through an implanted,intra-vital window.Moreover,vancomycin-resistant,abdominal S.aureus biofilms could be eradicated consuming solely endogenous glucose,without any glucose addition.Herewith,a new,non-antibiotic-based infection-control strategy has been provided,constituting a welcome addendum to the shrinking clinical armamentarium to control antibiotic-resistant bacterial infections. | Guang Yang Da-Yuan Wang Yong Liu Fan Huang Shuang Tian Yijin Ren Jianfeng Liu Yingli An Henny C.van der Mei Henk J.Busscher Linqi Shi | 2022 | Bioactive Materials2022,7,8: | 0 |

| 7 | Influence of maximum principal stress direction on the failure process and characteristics of D-shaped tunnels显示文摘To investigate the failure process and characteristics of D-shaped tunnels under different maximum principal stress directions θ, true-triaxial tests were conducted on cubic sandstone samples with a through D-shaped hole. The test results show that the failure process can be divided into 4 periods:calm, buckling deformation, gradual buckling and exfoliation of rock fragment, and formation of a Vshaped notch. With an increase in θ from 0° to 90°, the size of the rock fragments first decreases and then increases, whereas the fractal dimension of the rock fragments first increases and then decreases. Meanwhile, the failure position at the left side shifts from the sidewall to the corner and finally to the floor, whereas the failure position at the right side moves from the sidewall to the spandrel and finally to the roof, which is consistent with the failure position in underground engineering. In addition, the initial vertical failure stress first decreases and then increases. | 8 | HLA-mismatched allogeneic adoptive immune therapy in severely immunosuppressed AIDS patients显示文摘Severely immunosuppressed AIDS patients with recurrent opportunistic infections(Ols)represent an unmet medical need even in the era of antiretroviral therapy(ART).Here we report the development of a human leukocyte antigen(HLA)-mismatched allogeneic adaptive immune therapy(AAIT)for severely immunosuppressed AIDS patients.Twelve severely immunosuppressed AIDS patients with severe Ols were enrolled in this single-arm study.Qualified donors received subcutaneous recombinant granulocyte-colony-stimulating factor twice daily for 4-5 days to stimulate hematopoiesis.Peripheral blood mononuclear cells were collected from these donors via leukapheresis and transfused into the coupled patients.Clinical,immunological,and virological parameters were monitored during a 12-month follow-up period.We found AAIT combined with ART was safe and well-tolerated at the examined doses and transfusion regimen in all 12 patients.Improvements in clinical symptoms were evident throughout the study period.All patients exhibited a steady increase of peripheral CD4^(+)T cells from a median 10.5 to 207.5 cells/μl.Rapid increase in peripheral CD8^(+)T-cell count from a median 416.5 to 1206.5 cells/μl was found in the first 90 days since initiation of AAIT.In addition,their inflammatory cytokine levels and HIV RNA viral load decreased.A short-term microchimerism with donor cells was found.There were no adverse events associated with graft-versus-host disease throughout the study period.Overall,AAIT treatment was safe,and might help severely immunosuppressed AIDS patients to achieve a better immune restoration.A further clinical trial with control is necessary to confirm the efficacy of AAIT medication. | Ruonan Xu Ji-Yuan Zhang Bo Tu Zhe Xu Hui-Huang Huang Lei Huang Yan-Mei Jiao Tao Yang Chao Zhang En-Qiang Qin Tian-Jun Jiang Yun-Bo Xie Yuan-Yuan Li Lei Jin Chun-Bao Zhou Ming Shi Mei Guo Hui-Sheng Ai Linqi Zhang Fu-Sheng Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 0 |
| 9 | A forkhead transcription factor contributes to the regulatory differences of pathogenicity in closely related fungal pathogens显示文摘Cryptococcus neoformans and its sister species Cryptococcus deuterogattii are important human fungal pathogens.Despite their phylogenetically close relationship,these two Cryptococcus pathogens are greatly different in their clinical characteristics.However,the determinants underlying the regulatory differences of their pathogenicity remain largely unknown.Here,we show that the forkhead transcription factor Hcm1 promotes infection in C.neoformans but not in C.deuterogattii.Monitoring in vitro and in vivo fitness outcomes of multiple clinical isolates from the two pathogens indicates that Hcm1 mediates pathogenicity in C.neoformans through its key involvement in oxidative stress defense.By comparison,Hcm1 is not critical for antioxidation in C.deuterogattii.Furthermore,we identified SRX1,which encodes the antioxidant sulfiredoxin,as a conserved target of Hcm1 in two Cryptococcus pathogens.Like HCM1,SRX1 had a greater role in antioxidation in C.neoformans than in C.deuterogattii.Significantly,overexpression of SRX1 can largely rescue the defective pathogenicity caused by the absence of Hcm1 in C.neoformans.Conversely,Srx1 is dispensable for virulence in C.deuterogattii.Overall,our findings demonstrate that the difference in the contribution of the antioxidant sulfiredoxin to oxidative stress defense underlies the Hcm1-mediated regulatory differences of pathogenicity in two closely related pathogens. | Weixin Ke Yuyan Xie Yue Hu Hao Ding Xin Fan Jingjing Huang Xiuyun Tian Baokun Zhang Yingchun Xu Xiao Liu Ying Yang Linqi Wang | 2022 | mLife2022,1,1: | 0 |
