| 1 | Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities. | Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu | 2011 | Cellular & Molecular Immunology2011,8,1: | 16 |

| 4 | Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection. | Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng | 2018 | Cellular & Molecular Immunology2018,15,3: | 13 |

Using a novel active region design, a diagonal bound-to-bound lasing transition is guaranteed by efficient electron injection into the upper laser level and fast nonresonant electron extraction through a miniband from the lower laser level. For a 4 mm long and 40 μm wide double channel ridge waveguide laser with 55 stages of the active region, the threshold current density is only 3.13 k A∕cm^2 at room temperature. At 293 K, the maximum singlefacet peak power and average power are up to 830 m W and 75 m W, respectively. | 17 | Junctional and somatic hypermutation-induced CX4C motif is critical for the recognition of a highly conserved epitope on HCV E2 by a human broadly neutralizing antibody显示文摘Induction of broadly neutralizing monoclonal antibodies(bNAbs)that bind to the viral envelope glycoproteins is a major goal of hepatitis C virus(HCV)vaccine research.The study of bNAbs arising in natural infection is essential in this endeavor.We generated a human antibody,8D6,recognizing the E2 protein of HCV isolated from a chronic hepatitis C patient.This antibody shows broadly neutralizing activity,which covers a pan-genotypic panel of cell culture-derived HCV virions(HCVcc).Functional and epitope analyses demonstrated that 8D6 can block the interaction between E2 and CD81 by targeting a highly conserved epitope on E2.We describe how the 8D6 lineage evolved via somatic hypermutation to achieve broad neutralization.We found that the V(D)J recombination-generated junctional and somatic hypermutation-induced disulfide bridge(C-C)motif in the CDRH3 is critical for the broad neutralization and binding activity of 8D6.This motif is conserved among a series of broadly neutralizing HCV antibodies,indicating a common binding model.Next,the 8D6 inferred germline(iGL)was reconstructed and tested for its binding affinity and neutralization activity.Interestingly,8D6 iGL-mediated relatively strong inhibition of the 1b genotype PR79L9 strain,suggesting that PR79L9 may serve as a potential natural viral strain that provides E2 sequences that induce bNAbs.Overall,our detailed epitope mapping and genetic studies of the HCV E2-specific mAb 8D6 have allowed for further refinement of antigenic sites on E2 and reveal a new mechanism to generate a functional CDRH3,while its iGL can serve as a probe to identify potential HCV vaccine strains. | Chunyan Yi Jing Xia Lan He Zhiyang Ling Xuesong Wang Yu Yan Jiangjun Wang Xinhao Zhao Weiguo Fan Xiaoyu Sun Ronghua Zhang Sheng Ye Rongguang Zhang Yongfen Xu Liyan Ma Yaguang Zhang Honglin Zhou Zhong Huang Junqi Niu Gang Long Junxia Lu Jin Zhong Bing Sun | 2021 | Cellular & Molecular Immunology2021,18,3: | 0 |

| 20 | Thermal-annealing-regulated plasmonic enhanced fluorescence platform enables accurate detection of antigen/antibody against infectious diseases显示文摘Plasmonic enhanced fluorescence(PEF)technology is a powerful strategy to improve the sensitivity of immunofluorescence microarrays(IFMA),however,current approaches to constructing PEF platforms are either expensive/time-consuming or reliant on specialized instruments.Here,we develop a completely alternative approach relying on a two-step protocol that includes the self-assembly of gold nanoparticles(GNPs)at the water–oil interface and subsequent annealing-assisted regulation of gold nanogap.Our optimized thermal-annealing GNPs(TA-GNP)platform generates adequate hot spots,and thus produces high-density electromagnetic coupling,eventually enabling 240-fold fluorescence enhancement of probed dyes in the near-infrared region.For clinical detection of human samples,TA-GNP provides super-high sensitivity and low detection limits for both hepatitis B surface antigen and SARS-CoV-2 binding antibody,coupled with a much-improved detection dynamic range up to six orders of magnitude.With fast detection,high sensitivity,and low detection limit,TA-GNP could not only substantially improve the outcomes of IFMA-based precision medicine but also find applications in fields of proteomic research and clinical pathology. | Ying Yue Jingjie Nan Yuanyuan Che Hongqin Xu Weihong Sun Feiran Zhang Lei Wang Wei Xu Junqi Niu Shoujun Zhu Junhu Zhang Bai Yang | 2023 | Nano Research2023,16,2: | 0 |
