2Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9

4Cenozoic Evolution of Sediments and Climate Change and Response to Tectonic Uplift of the Northeastern Tibetan Plateau显示文摘Through a comprehensive study of magnetostratigraphy and sedimentology of several basins in the northeastern Tibetan Plateau, we reveal that the study area mainly experienced six tectonic uplift stages at approximately 52 Ma, 34–30 Ma, 24–20 Ma, 16–12 Ma, 8–6 Ma, and 3.6–2.6 Ma. Comprehensive analyses of pollen assemblages from the Qaidam, Linxia, Xining, and West Jiuquan Basins show that the northeastern Tibetan Plateau has undergone six major changes in vegetation types and climate: 50–40 Ma for the warm-humid forest vegetation, 40–23 Ma for the warm-arid and temperate-arid forest steppe vegetation, 23–18.6 Ma for the warm-humid and temperatehumid forest vegetation, 18.6–8.5 Ma for the warm-humid and cool-humid forest steppe vegetation, 8.6–5 Ma for the temperate sub-humid savanna steppe vegetation, and 5–1.8 Ma for the cold-arid steppe vegetation. 5Hepatitis C Virus NS2 Protein Suppresses RNA Interference in Cells显示文摘RNAi interference(RNAi)is an evolutionarily conserved post-transcriptional gene silencing mechanism and has been well recognized as an important antiviral immunity in eukaryotes.Numerous viruses have been shown to encode viral suppressors of RNAi(VSRs)to antagonize antiviral RNAi.Hepatitis C virus(HCV)is a medically important human pathogen that causes acute and chronic hepatitis.In this study,we screened all the nonstructural proteins of HCV and found that HCV NS2 could suppress RNAi induced either by small hairpin RNAs(shRNAs)or small interfering RNAs(siRNAs)in mammalian cells.Moreover,we demonstrated that NS2 could suppress RNAi via its direct interaction with doublestranded RNAs(dsRNAs)and siRNAs,and further identified that the cysteine 184 of NS2 is required for the RNAi suppression activity through a serial of point mutation analyses.Together,our findings uncovered that HCV NS2 can act as a VSR in vitro,thereby providing novel insights into the life cycle and virus-host interactions of HCV.Hui Zhou Qi Qian Ting Shu Jiuyue Xu Jing Kong Jingfang Mu Yang Qiu Xi Zhou 2020Virologica Sinica2020,35,4:3
6Effect of Structural Parameters on the Performance of NS High-efficiency Composite TraysⅠ.The Effect of Aperture and Opening Ratio on NS High-efficiency Composite Trays显示文摘In this paper,the structure and characteristics of the NS high-efficiency composite trays based on the doublelayer aperture jet sieve plate and compositely structured packing were investigated.The effect of aperture and opening ratio of plate on the fluid dynamics of the NS high-efficiency composite trays,such as the dry tray pressure drop,the wet tray pressure drop,the entrainment,the froth height,the leakage and mass transfer characteristics,were investigated.As a result, the low pressure drop,the high efficiency and the high capacity are the main advantages of the NS high-efficiency composite trays compared to other types of trays.According to this study,small aperture is useful for reducing the pressure drop and entrainment with a high mass transfer efficiency;while large aperture can achieve high capacity and efficiency in a broader operating range at the same pressure drop and entrainment.Qiao Yingyun Tian Yuanyu Xu Jingfang Xie Kechang 2009China Petroleum Processing & Petrochemical Technology2009,11,3:2
7OsbZIP72 Is Involved in Transcriptional Gene-Regulation Pathway of Abscisic Acid Signal Transduction by Activating Rice High-Affinity Potassium Transporter OsHKT1;1显示文摘We created CRISPR-Cas9 knock-out and overexpressing OsbZIP72 transgenic rice plants to gain a better understanding of the role and molecular mechanism of OsbZIP72 gene in stress tolerance,which has remained largely elusive.OsbZIP72 was expressed and integrated into rice transgenic plant genomes,and the OsbZIP72 transcript in overexpression lines was elicited by salinity,abscisic acid(ABA)and drought stresses.OsbZIP72 overexpressing plants showed higher tolerance to drought and salinity stresses,while knock-out transgenic lines showed higher sensitivity to these stresses.The differentially expressed genes(DEGs)from RNA-sequencing data encompassed several abiotic stress genes,and the functional classification of these DEGs demonstrated the robust transcriptome diversity in OsbZIP72.Yeast one-hybrid,along with luciferase assay,indicated that OsbZIP72 acted as a transcriptional initiator.Remarkably,electrophoresis mobility assay revealed that OsbZIP72 bound directly to the ABAresponsive element in the OsHKT1;1 promoter region and activated its transcription.Overall,our findings revealed that OsbZIP72 can act as a transcriptional modulator with the ability to induce the expression of OsHKT1;1 in response to environmental stress through an ABA-dependent regulatory pathway,indicating that OsbZIP72 can play a crucial role in the ABA-mediated salt and drought tolerance pathway in rice.WANG Baoxiang LIU Yan WANG Yifeng LI Jingfang SUN Zhiguang CHI Ming XING Yungao XU Bo YANG Bo LI Jian LIU Jinbo CHEN Tingmu FANG Zhaowei LU Baiguan XU Dayong Babatunde Kazeem BELLO 2021Rice science2021,28,3:2

14Knowledge Graph Construction and Applications for Web Search and Beyond显示文摘Knowledge graph(KG)has played an important role in enhancing the performance of many intelligent systems.In this paper,we introduce the solution of building a large-scale multi-source knowledge graph from scratch in Sogou Inc.,including its architecture,technical implementation and applications.Unlike previous works that build knowledge graph with graph databases,we build the knowledge graph on top of SogouQdb,a distributed search engine developed by Sogou Web Search Department,which can be easily scaled to support petabytes of data.As a supplement to the search engine,we also introduce a series of models to support inference and graph based querying.Currently,the data of Sogou knowledge graph that are collected from 136 different websites and constantly updated consist of 54 million entities and over 600 million entity links.We also introduce three applications of knowledge graph in Sogou Inc.:entity detection and linking,knowledge based question answering and knowledge based dialog system.These applications have been used in Web search products to help user acquire information more efficiently.Peilu Wang Hao Jiang Jingfang Xu Qi Zhang 2019Data Intelligence2019,1,4:1

16Transcriptomic analysis reveals the effect of the exopolysaccharide of Psychrobacter sp.B-3 on gene expression in RAW264.7 macrophage cells显示文摘B-3 exopolysaccharide is extracted from the Antarctic psychrophilic bacterium Psychrobacter sp. B-3. We have previously shown that it activates macrophages and affects their immunoregulatory activities. To determine what genes are affected during this process, we detected the genes differentially expressed in cells of RAW264.7 macrophages treated with B-3 exopolysaccharide by transcriptomic analysis. B-3 exopolysaccharide treatment caused differential expression of 420 genes, of which 178 were up-regulated and 242 were down-regulated. These genes were shown to be involved in many aspects of cell function, mainly metabolism and immunity. Genes were enriched in multiple immune-related pathways, and the most significantly enriched genes were involved in antigen processing and presentation pathways. The pathway in which differentially expressed genes were the most significantly enriched was the metabolic pathway; specifically, the expression of many metabolic enzyme genes was altered by B-3 exopolysaccharide treatment. Additionally, the genes involved in metabolisms of amino acids, carbohydrates, lipids and nucleotides, varied to certain degrees. B-3 exopolysaccharide, therefore, appears to directly affect the immune function of RAW264.7 macrophages as an immunostimulant, or to indirectly change intracellular metabolism. This is the first study to determine the effect of an Antarctic psychrophilic bacterial exopolysaccharide on RAW264.7 macrophages. 18Monteggia equivalent lesion in children:a narrative review显示文摘Background Monteggia equivalent lesion represents a group of injury or combined injury patterns that resemble the Monteggia lesion in its presentations and mechanisms.Unlike Monteggia lesions,the equivalent ones,which share vague definitions and mostly occur as sporadic single case reports in the literature,have not been thoroughly reviewed since Bado first proposed the term,especially in the pediatric population.The objective of this review was to elucidate the definition by elaborating on its clinical styles and thus analyzing the mechanism,diagnosis,and management through related literature.Data sources Based on the terms of‘Monteggia equivalent’,‘radial neck fracture’and‘pediatric’,all of the related literature was searched on the PubMed and Google Scholar search engine.Results The advance of the definitions for pediatric Monteggia equivalent lesion(PMEL)was reviewed.The functional roles of the ulnar and the related mechanism theories in this injury were analyzed.The status of the radiocapitellar joint in this injury was emphasized.According to the previous statements,a new classification model was proposed and proper diagnosis and treatment approaches were suggested.Conclusions PMEL should be defined as an ulnar fracture at any level combined with a proximal radial fracture.According to the status of the radiocapitellar joint,it could be divided into three groups.The occult ulnar bowing and delayed radial head dislocation should be a serious concern of orthopedists.Surgical need is usually warranted.Maintaining the ulnar length and securing the radiocapitellar joint are highly recommended.Lujie Xu Wensong Ye Haibing Li Jingfang Xu Weiwei Zhu Zhefeng Zhen Yi Yang 2021World Journal of Pediatric Surgery2021,4,3:0
19Construction of a 980 nm laser-activated Pt(Ⅱ)metallacycle nanosystem for efficient and safe photo-induced bacteria sterilization显示文摘Although metal-based chemical agents have demonstrated promising bacteriostatic effects in phototherapy,their short excitation/emission wavelengths and inadequate phototherapy efficiencies make their application in vivo difficult.We therefore synthesized a novel Pt(Ⅱ)metallacycle(Pt1110)that can be activated with a 980 nm laser for photodiagnosis/treatment in deep tissue.We found that Pt1110 significantly improved photothermal conversion(95%improvement)and ^(1)O_(2) generation(ΦΔ75%increase)compared to the ligand itself 1 and was well capable of light-induced sterilization under safe laser irradiation(0.72 W/cm^(2)).In addition,Pt1110 has little to no toxicity to cells.After incorporated into liposome,Pt1110 NPs was effective in wound healing in infection and keratitis models upon laser irradiation,which was accurately observed by NIR-Ⅱfluorescence imaging.This novel metal-coordinated supramolecular material has a potential to become a universal platform for phototherapy in deep tissue.Yuling Xu Chonglu Li Jusung An Xin Ma Jingfang Yang Lishi Luo Yun Deng Jong Seung Kim Yao Sun 2023Science China Chemistry2023,66,1:0
