3Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 4OsGRF4 controls grain shape, panicle length and seed shattering in rice显示文摘Traits such as grain shape, panicle length and seed shattering, play important roles in grain yield and harvest. In this study, the cloning and functional analysis of PANICLE TRAITS 2(PT2), a novel gene from the Indica rice Chuandali(CDL), is reported. PT2 is synonymous with Growth-Regulating Factor 4(OsGRF4), which encodes a growth-regulating factor that positively regulates grain shape and panicle length and negatively regulates seed shattering. Higher expression of OsGRF4 is correlated with larger grain, longer panicle and lower seed shattering. A unique OsGRF4 mutation, which occurs at the Osmi RNA396 target site of OsGRF4, seems to be associated with high levels of OsGRF4 expression, and results in phenotypic difference. Further research showed that OsGRF4 regulated two cytokinin dehydrogenase precursor genes(CKX5 and CKX1) resulting in increased cytokinin levels,which might affect the panicle traits. High storage capacity and moderate seed shattering of OsGRF4 may be useful in high-yield breeding and mechanized harvesting of rice. 6Surgical approaches for stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis:feasibility of complete video-assisted thoracoscopic surgery (VATS) thymectomy in comparison with trans-sternal resection显示文摘Complete resection could be achieved in virtually all myasthenic patients with Masaoka stage Ⅰ and Ⅱ thymoma using the trans-sternal technique.Whether this is appropriate for minimally invasive approach is not yet clear.We evaluated the feasibility of complete video-assisted thoracoscopic surgery (VATS) thymectomy for the treatment of Masaoka stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis,compared to conventional trans-sternal thymectomy.We summarized 33 patients with Masaoka stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis between April 2006 and September 2011.Of these,15 patients underwent right-sided complete VATS (the VATS group) by using adjuvant pneuomomediastinum,comparing with 18 patients using the trans-sternal approach (the T3b group).No intraoperative death was found and no VATS case required conversion to median sternotomy.Significant differences between the two groups regarding duration of surgery and volume of intraoperative blood loss (P=0.001 and P < 0.001,respectively) were observed.Postoperative morbidities were 26.7% and 33.3% for the VATS and T3b groups,respectively.All 33 patients were followed up for 12 to 61 months in the study.The cumulative probabilities of reaching complete stable remission and effective rate were 26.7% (4/15) and 93.3% (14/15) in the VATS group,which had a significantly higher complete stable remission and effective rate than those in the T3b group (P=0.026 and P=0.000,respectively).We conclude that VATS thymectomy utilizing adjuvant pneuomomediastinum for the treatment of stage Ⅰ and Ⅱ thymoma-associated myasthenia gravis is technically feasible but deserves further investigation in a large series with long-term follow-up.Zhicheng He Quan Zhu Wei Wen Liang Chen Hai Xu Hai Li 2013The Journal of Biomedical Research2013,27,1:15
7One-stitch anastomosis through the skin with bicanalicular intubation:a modified approach for repair of bicanalicular laceration显示文摘AIM:To evaluate the efficacy and safety of one-stitch anastomosis through the skin with bicanalicular silicone tube intubation in repairing of bicanalicular laceration.METHODS:The clinical data of 15 consecutive patients with both superior and inferior canalicular laceration in one eye who underwent surgical repair using one-stitch anastomosis through the skin and bicanalicular stent were retrospective studied.All the operations were performed under surgical microscope,5-0 silk sutures were used and were with bicanalicular silicone tube(diameter was 8mm) intubation,for one lacerated canaliculi one-stitch anastomosis through the skin.The stents were left in place for 3 months postoperatively and then removed.The follow-up period was 3-36 months(average 14 months). RESULTS:In 15 patients,13 patients were cured entirely,1 patient was meliorated,1 patient with no effects.All patients had got good recovery of eyelid laceration with no traumatic deformity in eyelid and canthus.Complication was seen in one case,for not followed the doctor's guidance to come back to hospital to had the suture removed on the 7thday after operation,when he came at the 15thday,the inferior canalicular wall and eyelid skin were corroded by the suture caused 2mm wound,and the inside silicone tube was exposed,a promptly repair with 10-0 nylon suture was done,the wound healed in a week.There were no early tube protrusions and punctal slits in the patients.CONCLUSION:One-stitch anastomosis through the skin with bicanalicular silicone tube intubation is a good method in repair of bicanalicular laceration in one eye,the cut ends can be anastomosed directly,and with excellent cosmetic results,it is acceptable for the patients.For there is no suture remained in the wound permanently,so there is no suture-related granuloma which may cause obstruction or stenosis of canaliculi.It is simple,economical,effective and safe.Hai Tao Peng Wang Cui Han Jian Zhang Fang Bai Zhao-Yan He 2013International Journal of Ophthalmology(English edition)2013,6,5:14
8Tomato SlAN11 regulates flavonoid biosynthesis and seed dormancy by interaction with bHLH proteins but not with MYB proteins显示文摘The flavonoid compounds are important secondary metabolites with versatile human nutritive benefits and fulfill a multitude of functions during plant growth and development.The abundance of different flavonoid compounds are finely tuned with species-specific pattern by a ternary MBW complex,which consists of a MYB,a bHLH,and a WD40 protein,but the essential role of SlAN11,which is a WD40 protein,is not fully understood in tomato until now.In this study,a tomato WD40 protein named as SlAN11 was characterized as an effective transcription regulator to promote plant anthocyanin and seed proanthocyanidin(PA)contents,with late flavonoid biosynthetic genes activated in 35S::SlAN11 transgenic lines,while the dihydroflavonol flow to the accumulation of flavonols or their glycosylated derivatives was reduced by repressing the expression of SlFLS in this SlAN11-overexpressed lines.The above changes were reversed in 35S::SlAN11-RNAi transgenic lines except remained levels of flavonol compounds and SlFLS expression.Interestingly,our data revealed that SlAN11 gene could affect seed dormancy by regulating the expressions of abscisic acid(ABA)signaling-related genes SlABI3 and SlABI5,and the sensitivity to ABA treatment in seed germination is conversely changed by SlAN11-overexpressed or-downregulated lines.Yeast two-hybrid assays demonstrated that SlAN11 interacted with bHLH but not with MYB proteins in the ternary MBW complex,whereas bHLH interacted with MYB in tomato.Our results indicated that low level of anthocyanins in tomato fruits,with low expression of bHLH(SlTT8)and MYB(SlANT1 and SlAN2)genes,remain unchanged upon modification of SlAN11 gene alone in the transgenic lines.These results suggest that the tomato WD40 protein SlAN11,coordinating with bHLH and MYB proteins,plays a crucial role in the fine adjustment of the flavonoid biosynthesis and seed dormancy in tomato.Yongfeng Gao Jikai Liu Yongfu Chen Hai Tang Yang Wang Yongmei He Yongbin Ou Xiaochun Sun Songhu Wang Yinan Yao 2018Horticulture Research2018,5,1:13
9Endoplasmic reticulum stress-induced apoptosis in the penumbra aggravates secondary damage in rats with traumatic brain injury显示文摘Neuronal apoptosis is mediated by intrinsic and extrinsic signaling pathways such as the membrane-mediated,mitochondrial,and endoplasmic reticulum stress pathways.Few studies have examined the endoplasmic reticulum-mediated apoptosis pathway in the penumbra after traumatic brain injury,and it remains unclear whether endoplasmic reticulum stress can activate the caspase-12-dependent apoptotic pathway in the traumatic penumbra.Here,we established rat models of fluid percussion-induced traumatic brain injury and found that protein expression of caspase-12,caspase-3 and the endoplasmic reticulum stress marker 78 k Da glucose-regulated protein increased in the traumatic penumbra 6 hours after injury and peaked at 24 hours.Furthermore,numbers of terminal deoxynucleotidyl transferase-mediated d UTP nick end labeling-positive cells in the traumatic penumbra also reached peak levels 24 hours after injury.These findings suggest that caspase-12-mediated endoplasmic reticulum-related apoptosis is activated in the traumatic penumbra,and may play an important role in the pathophysiology of secondary brain injury.Guo-zhu Sun Fen-fei Gao Zong-mao Zhao Hai Sun Wei Xu Li-wei Wu Yong-chang He 2016Neural Regeneration Research2016,11,8:12

12One-pot synthesis and characterization of a new, branched polycarbosilane bearing allyl groups显示文摘A highly branched polycarbosilane bearing allyl groups has been prepared by a one-pot synthesis with chloromethyltrichlor-osilane, chloromethylmethyldichlorosilane and allyl chloride as the starting materials. The resultant polymer, with the approximate formula [SiH1.2(CH3)0.71(CH2CH=CH2)0.09CH2]n, has been characterized by 1H, 13C, and 29Si NMR, GPC, TG and elemental analysis. It could be cross-linked thermally at 170℃in the absence of oxygen. 13Effect of heparin on apoptosis in human nasopharyngeal carcinoma CNE2 cells显示文摘In order to study the mechanism of the effect of heparin on apoptosis in carcinoma cells, the nasopharyngeal carcinoma cell line CNE2 was used to identify the effect of heparin on apoptosis associated with the expression of c-myc, bax, bcl-2 proteins by use of Hoechst 33258 staining, terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), agarose gel electrophoresis, and flow cytometry, as well as Western blot analysis. The results showed that heparin induced apoptosis of CNE2 cells including the morphologic changes such as reduction in the volume, and the nuclear chromatin condensation, as well as the “ladder pattern” revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner.The number of TUNEL-positive cells was dramatically increased to 33.6±1.2% from 2.8±0.3% by treat ment with heparin in different concentrations (10~40 kU/L). The apoptotic index was increased to 32.5% from 3.5% by detecting SubG1 peaks on flow cytometry. 15Programmed Death Ligand-1 on Microglia Regulates Th1 Differentiation via Nitric Oxide in Experimental Autoimmune Encephalomyelitis显示文摘Microglia are considered to be potential antigen-presenting cells and have the ability to present antigen under pathological conditions. Nevertheless, whether and how microglia are involved in immune regulation are largely unknown. Here, we investigated the suppressive activity of microglia during experimental autoimmune encephalomyelitis(EAE) induced by myelin oligodendrocyte glycoprotein, with the goal of understanding their role in regulating the T cell reaction. Using flow cytometric analysis, we found that microglia were characterized by increased cell number and up-regulated programmed death ligand-1(PD-L1) at the peak phase of EAE. Meanwhile,both the CD^(4+)T cells and microglia that infiltrated the central nervous system expressed higher levels of PD1, the receptor for PD-L1, accompanied by a decline of Th1 cells.In an ex vivo co-culture system, microglia from EAE mice inhibited the proliferation of antigen-specific CD^(4+)T cells and the differentiation of Th1 cells, and this was significantly inhibited by PD-L1 blockade. Further,microglia suppressed Th1 cells via nitric oxide(NO), the production of which was dependent on PD-L1. 16Autophagy Accompanied with Bisdemethoxycurcumin-induced Apoptosis in Non-small Cell Lung Cancer Cells显示文摘Objective To investigate the effects of bisdemethoxycurcumin(BDMC) on non-small cell lung cancer(NSCLC) cell line, A549, and the highly metastatic lung cancer 95 D cells. Methods CCK-8 assay was used to assess the effect of BDMC on cytotoxicity. Flow cytometry was used to evaluate apoptosis. Western blot analysis, electron microscopy, and quantification of GFP-LC3 punctuates were used to test the effect of BDMC on autophagy and apoptosis of lung cancer cells. Results BDMC inhibited the viability of NSCLC cells, but had no cytotoxic effects on lung small airway epithelial cells(SAECs). The apoptotic cell death induced by BDMC was accompanied with the induction of autophagy in NSCLC cells. Blockage of autophagy by the autophagy inhibitor 3-methyladenine(3-MA) repressed the growth inhibitory effects and induction of apoptosis by BDMC. In addition, BDMC treatment significantly decreased smoothened(SMO) and the transcription factor glioma-associated oncogene 1(Gli1) expression. Furthermore, depletion of Gli1 by si RNA and cyclopamine(a specific SMO inhibitor) induced autophagy. Conclusion Aberrant activation of Hedgehog(Hh) signaling has been implicated in several human cancers, including lung cancers. 18Magnetic nanoparticles based cancer therapy: current status and applications显示文摘Nanotechnology holds a promising potential for developing biomedical nanoplatforms in cancer therapy. The magnetic nanoparticles, which integrate uniquely appealing features of magnetic manipulation, nanoscale heat generator, localized magnetic field and enzyme-mimics, prompt the development and application of magnetic nanoparticles-based cancer medicine. Considerable success has been achieved in improving the magnetic resonance imaging(MRI) sensitivity, and the therapeutic function of the magnetic nanoparticles should be given adequate attention. This work reviews the current status and applications of magnetic nanoparticles based cancer therapy.
