| 1 | One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients显示文摘The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. | 2 | Microcalorimetric investigation of effect of berberine alkaloids from Coptis chinensis Franch on intestinal diagnostic flora growth显示文摘The inhibitory effect of three berberine alkaloids (BAs) from Coptis chinensis Franch, a traditional Chinese medicinal (TCM) herb, on Bifidobacterium adolescentis growth was investigated by micro- calorimetry. The power-time curves of B. adolescentis with and without BAs were acquired, meanwhile the extent and duration of inhibitory effect on the metabolism were evaluated by the growth rate con- stant (k), half inhibitory ratio (IC50), maximum heat-output power (Pmax), peak time of maximum heat-output power (tp) and total heat production (Qt). k, Pmax and Qt decreased, and tp was prolonged with the increase of BAs concentration. The IC50 of BAs is 806 μg/mL for berberine, 341 μg/mL for cop- tisine and 236 μg/mL for palmatine. The sequence of antimicrobial activity of BAs is berberine < cop- tisine < palmatine. Combined with previous studies, it could be shown that the sequences of antim- icrobial activity of BAs on both Bacillus shigae and Escherichia coli are berberine > coptisine > pal- matine. The structure-function relationship of BAs indicates that the functional group methylenedioxy or methoxyl at C2 and C3 might be the major group inducing the activities of BAs on E. coli and B. adolescentis. | 3 | Genome-wide Analysis of MYB Gene Family in Potato Provides Insights into Tissue-specific Regulation of Anthocyanin Biosynthesis显示文摘The MYB transcription factor is one of the largest gene families in plants,playing an important role in regulating plant growth,development,response to stress,senescence,and especially the anthocyanin biosynthesis.In this study,A total of 217 MYB genes,including 901R-MYBs,124 R2R3-MYBs,and 3 R1R2R3-MYBs have been identified from the potato genome.The 1R-MYB and R2R3-MYB family members could be divided into 20 and 35 subgroups respectively.Analysis of gene structure and protein motifs revealed that members within the same subgroup presented similar exon/intron and motif organization,further supporting the results of phylogenetic analysis.Potato is an ideal plant to reveal the tissue-specific anthocyanins biosynthesis regulated by MYB,as the anthocyanins could be accumulated in different tissues,showing colorful phenotypes.Five pairs of colored and colorless tissues,stigma,petal,stem,leaf,and tuber flesh,were applied to the transcriptomic analysis.A total of 70 MYB genes were found to be differentially expressed between colored and colorless tissues,and these differentially expressed genes were suspected to regulate the biosynthesis of anthocyanin of different tissues.Co-expression analysis identified numerous potential interactive regulators of anthocyanins biosynthesis,involving 39 MYBs,24 bHLHs,2 WD-repeats,and 29 biosynthesis genes.Genome-wide association study(GWAS)of tuber flesh color revealed amajor signal at the end of Chromosome 10,which was co-localized with reported I gene(StMYB88),controlling tuber peel color.Analyses of DEGs(Differentially Expression Genes)revealed that both StMYB88 and StMYB89 were closely related to regulating anthocyanin biosynthesis of tuber flesh.This work offers a comprehensive overview of the MYB family in potato and will lay a foundation for the functional validation of these genes in the tissue-specific regulation of anthocyanin biosynthesis. | Yumei Li Jun Liang Xiangzhi Zeng Han Guo Youwei Luo Philip Kear Shouming Zhang Guangtao Zhu | 2021 | Horticultural Plant Journal2021,7,2: | 8 |
| 4 | Utilization of Ogura CMS germplasm with the clubroot resistance gene by fertility restoration and cytoplasm replacement in Brassica oleracea L显示文摘Clubroot disease,a major plant root disease caused by Plasmodiophora brassicae,has become one of the most destructive diseases among cultivated cruciferous vegetables.However,clubroot-resistant Brassica oleracea materials are rare.A few clubroot-resistant cabbage varieties are available on the market,but all are Ogura cytoplasmic male sterile(CMS)types.Therefore,in this study,to reutilize the clubroot-resistant Ogura CMS germplasm of cabbage,a new fertility-restored Ogura CMS material,16Q2-11,was used as a bridge to transfer the clubroot resistance(CR)gene from the Ogura CMS cytoplasm to the normal cytoplasm by a two-step method(a fertility restoration and cytoplasm replacement method).In the first cross for fertility restoration of Ogura CMS clubroot-resistant cabbage(FRCRC),16Q2-11 was used as a restorer to cross with Ogura CMS materials containing the CR gene CRb2.Eleven Rfo-positive progenies were generated,of which four contained CRb2:F8-514,F8-620,F8-732 and F8-839.After inoculation with race 4 of P.brassicae,these four CRb2-positive individuals showed resistance.Furthermore,F8-514 and F8-839 were then used as male parents in the second cross of FRCRC to cross with cabbage inbred lines,resulting in the successful introgression of the CRb2 gene into the inbred lines.All offspring produced from this step of cross,which had a normal cytoplasm,showed a high resistance to race 4 of P.brassicae and could be utilized for the breeding of clubrootresistant cabbage varieties in the future.This is the first time that the Ogura CMS restorer has been used to restore the fertility of Ogura CMS clubroot-resistant cabbages,which could improve germplasm diversity in cabbage and provide a reference method for using CMS germplasm in Brassica crops. | Wenjing Ren Zhiyuan Li Fengqing Han Bin Zhang Xing Li Zhiyuan Fang Limei Yang Mu Zhuang Honghao Lv Yumei Liu Yong Wang Hailong Yu Yangyong Zhang | 2020 | Horticulture Research2020,7,1: | 7 |
| 5 | The action of medicinal animal horns on Escherichia coli growth investigated by microcalorimetry and chemometric analysis显示文摘The action of Cornu Cervi Pantotrichum (CCP), Cornu Cervi (CC) and Cornu Saigae Tataricae (CST) on Escherichia coli growth were investigated using microcalorimetry to find the heat change regularity of microbial growth. The similarity of thermogenic curves and thermodynamics parameters were investigated as evaluation index, such as the growth rate constant in the first expo- nential phase (k1), maximum power in the first exponential phase (P1), maximum power in the secondary exponential phase (P2), peak time in the first exponential phase (T1), peak time in the stationary phase (T2) and the total heat production in stage 1 (Q1), and the total heat production in stage 2 (Q2). Chemometric analysis was used as a reference for the bioactivity evaluation of medicinal animal horns. The results indicated that the similarity between CST and the control was smaller than that between CCP, CC and the control. Both CCP and CC could increase the heat in the microbial growth, whereas CST decreased it. | 8 | Effect of Wheel Load on Wheel Vibration and Sound Radiation显示文摘The current researches of wheel vibration and sound radiation mainly focus on the low noise damped wheel. Compared with the traditional research, the relationship between the sound and wheel/rail contact is difficulty and worth studying. However, there are few studies on the effect of wheel load on wheel vibration and sound radiation. In this paper, laboratory test carried out in a semi-anechoic room investigates the effect of wheel load on wheel natural frequencies, damping ratios, wheel vibration and its sound radiation. The laboratory test results show that the vibration of the wheel and total sound radiation decrease significantly with the increase of the wheel load from 0 t to 1 t. The sound energy level of the wheel decreases by 3.7 d B. When the wheel load exceeds 1 t, the attenuation trend of the vibration and sound radiation of the wheel becomes slow. And the increase of the wheel load causes the growth of the wheel natural frequencies and the mode damping ratios. Based on the finite element method(FEM) and boundary element method(BEM), a rolling noise prediction model is developed to calculate the influence of wheel load on the wheel vibration and sound radiation. In the calculation, the used wheel/rail excitation is the measured wheel/rail roughness. The calculated results show that the sound power level of the wheel decreases by about 0.4 dB when the wheel load increases by 0.5 t. The sound radiation of the wheel decreases slowly with wheel load increase, and this conclusion is verified by the field test. | 18 | A genome wide association study between copy number variation (CNV) and human height in Chinese population显示文摘Copy number variation (CNV) is a type of genetic variation which may have important roles in phenotypic variability and disease susceptibility. To hunt for genetic variants underlying human height variation, we performed a genome wide CNV association study for human height in 618 Chinese unrelated subjects using Affymetrix 500K array set. After adjusting for age and sex, we found that four CNVs at 6p21.3, 8p23.3-23.2, 9p23 and 16p12.1 were associated with human height (with borderline significant p value: 0.013, 0.011, 0.024, 0.049; respectively). However, after multiple tests correction, none of them was associated with human height. We observed that the gain of copy number (more than 2 copies) at 8p23.3-23.2 was associated with lower height (normal copy number vs. gain of copy number: 161.2 cm vs. 153.7 cm, p = 0.011), which accounted for 0.9% of height variation. Loss of copy number (less than 2 copies) at 6p21.3 was associated with 0.8% lower height (loss of copy number vs. normal copy number: 154.5 cm vs. 161.1 cm, p = 0.013). Since no important genes influencing height located in CNVs at loci of 8p23.3-23.2 and 6p21.3, the two CNVs may cause the structural rear- rangements of neighbored important candidate genes, thus regulates the variation of height.
