| 4 | CCL5 deficiency promotes liver repair by improving inflammation resolution and liver regeneration through M2 macrophage polarization显示文摘Despite the diverse etiologies of drug-induced liver injury(DILI),innate immunity activation is a common feature involved in DILI progression.However,the involvement of innate immunity regulation in inflammation resolution and liver regeneration in DILI remains obscure.Herein,we identified the chemokine CCL5 as a central mediator of innate immunity regulation in the pathogenesis of DILI.First,we showed that serum and hepatic CCL5 levels are elevated in both DILI patients and an APAP-induced liver injury(AILI)mouse model.Interestingly,both nonparenchymal cells and stressed hepatocytes are cell sources of CCL5 induction in response to liver injury.Functional experiments showed that CCL5 deficiency has no effect on the early phase of AILI but promotes liver repair in the late phase mainly by promoting inflammation resolution and liver regeneration,which are associated with an increased number of hepatic M2 macrophages.Mechanistically,CCL5 can directly activate M1 polarization and impede M2 polarization through the CCR1-and CCR5-mediated activation of the MAPK and NF-κB pathways.We then showed that CCL5 inhibition mediated by either a CCL5-neutralizing antibody or the antagonist Met-CCL5 can greatly alleviate liver injury and improve survival in an AILI mouse model.Our data demonstrate CCL5 induction during DILI,identify CCL5 as a novel innate immunity regulator in macrophage polarization,and suggest that CCL5 blockage is a promising therapeutic strategy for the treatment of DILI. | Meng Li Xuehua Sun Jie Zhao Lei Xia Jichang Li Min Xu Bingrui Wang Han Guo Chang Yu Yueqiu Gao Hailong Wu Xiaoni Kong Qiang Xia | 2020 | Cellular & Molecular Immunology2020,17,7: | 13 |
| 5 | Heterotic Group Classification of 63 Inbred Lines and Hybrid Purity Identification by Using SSR Markers in Winter Cabbage(Brassica Oleracea L. var. capitata)显示文摘Winter cabbage is an important crop cultivated through winter in the region near the Yangtze River, enabling the supply of fresh cabbage there at that time of year. However, a problem has emerged regarding the newly developed parents of winter cabbage, which is completely different from spring and autumn cabbage, namely, how to combine these parents to breed an elite hybrid. To classify the heterotic groups and improve the efficiency of parent selection in winter cabbage breeding, 20 polymorphic SSR markers were selected to screen 63 winter cabbage inbred lines. Seventeen pairs among the 20 SSR markers amplified polymorphic bands. These primers amplified two to six bands,with an average of 2.8 bands per primer, and a total of 47 polymorphic bands were generated in the 63 inbred lines. These lines included flatheaded morphotype and round-headed morphotype, thus they were separately classified into heterotic groups based on the SSR markers. The flat-headed morphotype contained 21 inbred lines and was classified into three heterotic groups, named Hanchun 4, Jiali, and Dongsheng, in accordance with the representative germplasm contained in each group. The round-headed morphotype contained 42 inbred lines and was classified into five heterotic groups, named Parte, Bejo1039, YK-143, SCA002, and Golden B90. Meanwhile, parent analysis of 20 developed elite combinations showed that their parents were all distributed in different heterotic groups, indicating that the group classification was reasonable,which can provide a basis for further parent selection in winter cabbage breeding. | 6 | Quality assessment of randomized controlled trials reporting on knee osteoarthritis treated with warming needle moxibustion显示文摘OBJECTIVE: To evaluate the quality of randomized controlled trials(RCTs) testing the efficacy of warming needle moxibustion on knee osteoarthritis(KOA).METHODS: Nine databases were searched systematically including: the Cochrane Library(1/1993-10/2012), Pub Med(1/1980-10/2012), EMBASE(1/1990-10/2012), SCI Expanded(1/1986-10/2012), Science Direct(1/1979-10/2012), Chinese Biomedical Literature Database disc(1/1978-10/2012), China National Knowledge Infrastructure Database(1/1979-10/2012), China Science and Technology Journal Database(a full text issue database of China, 1/1989-10/2012), and Wanfang Database(a full text issue database of China, 1/1990-10/2012). Traditional reference retrieval was also conducted. Language was limited to Chinese and English. We identified 37 RCTs that used warming needle moxibustion as an intervention and they were assessed with the Consolidated Standards for Reporting of Trials Statement 2010(CONSORT 2010) and Standards for Reporting Interventions Controlled Trials of Acupuncture 2010(STRICTA 2010).RESULTS: Of the 37 reports, only seventeen(45.95%) mentioned the random allocation of sequence methods, nine used unacceptable methods, and only one(2.70%) gave the description of the mechanism of allocation concealment. One study(2.70%) mentioned blinding, one(2.70%) had a sample size calculation, and twenty-three(62.16%) used reasonable statistical methods. Thirteen(35.14%) described the background and reason for the RCTs, three(8.11%) gave a description of lost or eliminated patients, and two(5.41%) reported accidental situations. Only three(8.11%)gave analysis of the RCT limitations. No report mentioned intentional analysis.CONCLUSION: The quality of the RCTs assessed in this study was from moderate to low. The design of RCTs, the methods of statistical analysis, and the description of reports information needs to be improved. | 8 | Utilization of Ogura CMS germplasm with the clubroot resistance gene by fertility restoration and cytoplasm replacement in Brassica oleracea L显示文摘Clubroot disease,a major plant root disease caused by Plasmodiophora brassicae,has become one of the most destructive diseases among cultivated cruciferous vegetables.However,clubroot-resistant Brassica oleracea materials are rare.A few clubroot-resistant cabbage varieties are available on the market,but all are Ogura cytoplasmic male sterile(CMS)types.Therefore,in this study,to reutilize the clubroot-resistant Ogura CMS germplasm of cabbage,a new fertility-restored Ogura CMS material,16Q2-11,was used as a bridge to transfer the clubroot resistance(CR)gene from the Ogura CMS cytoplasm to the normal cytoplasm by a two-step method(a fertility restoration and cytoplasm replacement method).In the first cross for fertility restoration of Ogura CMS clubroot-resistant cabbage(FRCRC),16Q2-11 was used as a restorer to cross with Ogura CMS materials containing the CR gene CRb2.Eleven Rfo-positive progenies were generated,of which four contained CRb2:F8-514,F8-620,F8-732 and F8-839.After inoculation with race 4 of P.brassicae,these four CRb2-positive individuals showed resistance.Furthermore,F8-514 and F8-839 were then used as male parents in the second cross of FRCRC to cross with cabbage inbred lines,resulting in the successful introgression of the CRb2 gene into the inbred lines.All offspring produced from this step of cross,which had a normal cytoplasm,showed a high resistance to race 4 of P.brassicae and could be utilized for the breeding of clubrootresistant cabbage varieties in the future.This is the first time that the Ogura CMS restorer has been used to restore the fertility of Ogura CMS clubroot-resistant cabbages,which could improve germplasm diversity in cabbage and provide a reference method for using CMS germplasm in Brassica crops. | Wenjing Ren Zhiyuan Li Fengqing Han Bin Zhang Xing Li Zhiyuan Fang Limei Yang Mu Zhuang Honghao Lv Yumei Liu Yong Wang Hailong Yu Yangyong Zhang | 2020 | Horticulture Research2020,7,1: | 7 |

| 10 | A Quantitative Evaluation of Shale Gas Content in Different Occurrence States of the Longmaxi Formation: A New Insight from Well JY-A in the Fuling Shale Gas Field,Sichuan Basin显示文摘Comprehensive quantitative evaluation of shale gas content and the controlling factors in different occurrence states is of great significance for accurately assessing gas-bearing capacity and providing effective well-production strategies. A total of 122 core samples from well JY-A in the Fuling shale gas field were studied to reveal the characteristics of S_1 l shale,15 of which were selected to further predict the shale gas content in different occurrence states, which are dependent on geological factors in the thermal evolution process. Geological parameters were researched by a number of laboratory programs, and the factors influential in controlling shale gas content were extracted by both PCA and GRA methods and prediction models were confirmed by the BE method using SPSS software. Results reveal that the adsorbed gas content is mainly controlled by TOC, Ro, SSA, PD and pyrite content, and the free gas content is mainly controlled by S_2, quartz content, gas saturation and formation pressure for S_1 l in well JY-A. Three methods, including the on-site gas desorption method, the empirical formula method, and the multiple regression analysis method were used in combination to evaluate the shale gas capacity of well JY-A, all of which show that the overall shale gas content of well JY-A is in the range of 2.0–5.0 m^3/t and that the free gas ratio is about 50%, lower than that of well JY-1. | 11 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 12 | Two regimes of Atlantic multidecadal oscillation: cross-basin dependent or Atlantic-intrinsic显示文摘The Atlantic Multidedal Oscillation(AMO) is a prominent mode of sea surface temperature variability in the Atlantic and incurs significant global influence. Most coupled models failed to reproduce the observed 50–80-year AMO, but were overwhelmed by a 10–30-year AMO. Here we show that the 50–80-year AMO and 10–30-year AMO represent two different AMO regimes. The key differences are:(1) the 50–80-year AMO involves transport of warm and saline Atlantic water into the Greenland-Iceland-Norwegian(GIN)Seas prior to reaching its maximum positive phase, while such a transport is weak for the 10–30-year AMO;(2) the zonality of atmospheric variability associated with the 50–80 year AMO favors the transport of warm and saline water into the GIN Seas;(3) the disappearance of Pacific variability weakens the zonality of atmospheric variability and the transport of warm and saline water into the GIN Seas, leading to the weakening of the 50–80-year AMO. In contrast, the 10–30-year AMO does not show dependence on the variability in Pacific and in the GIN Seas and may be an Atlantic-intrinsic mode. | 14 | Acetylglutamate kinase is required for both gametophyte function and embryo development in Arabidopsis thaliana显示文摘The specific functions of the genes encoding arginine biosynthesis enzymes in plants are not well characterized. We report the isolation and characterization of Arabidopsis thaliana N-acetylglutamate kinase(NAGK), which catalyzes the second step of arginine biosynthesis. NAGK is a plastid-localized protein and is expressed during most developmental processes in Arabidopsis. Heterologous expression of the Arabidopsis NAGK gene in a NAGK-deficient Escherichia coli strain fully restores bacterial growth on arginine-deficient medium. nagk mutant pollen tubes grow more slowly than wild type pollen tubes and the phenotype is restored by either specifically through complementation by NAGK in pollen, or exogenous supplementation of arginine. nagk female gametophytes are defective in micropylar pollen tube guidance due to the fact that female gametophyte cell fate specification was specifically affected. Expression of NAGK in synergid cells rescues the defect of nagk female gametophytes. Lossof-function of NAGK results in Arabidopsis embryos not developing beyond the four-celled embryo stage. The embryo-defective phenotype in nagk/NAGK plants cannot be rescued by watering nagk/NAGK plants with arginine or ornithine supplementation. | 17 | Comparative Transcriptome Analysis Reveals Different Host Cell Responses to Acute and Persistent Foot-and-Mouth Disease Virus Infection显示文摘Foot-and-mouth disease virus(FMDV) rapidly causes cytopathic effects in susceptible cells. Incomplete viral clearance during the acute infection leads to persistent infection. The relationship between host gene expression and the persistent infection remains unclear. In this study, we analyzed the transcriptome profiles of BHK-21 cells acutely and persistently infected with FMDV to identify differences in gene expression. GO and KEGG enrichment analyses showed that the 8,378 differentially expressed genes were significantly enriched in categories including metabolism, biosynthesis, ribosome function, and endocytosis. In persistently infected BHK-21 cells, ribosome-and translation-related genes were significantly down-regulated. There were more differentially expressed immune-related genes during persistent infection than during acute infection. Two hundred and seventy-four genes were differentially expressed in both acutely and persistently infected BHK-21 cells. Among these genes, heat shock protein family B member 1(Hspb1) knockdown significantly inhibited FMDV replication. | 18 | Reduced graphene oxide/silicon nanowire hetero- structures with enhanced photoactivity and superior photoelectrochemical stability显示文摘Silicon nanowires (SiNWs) have been widely studied as light harvesting antennas in photocatalysts due to their ability to absorb broad-spectrum solar radiation, but they are typically limited by poor photoelectrochemical stability. Here, we report the synthesis of reduced graphene oxide-SiNW (rGO-SiNW) heterostructures to achieve greatly improved photocatalytic activity and stability. The SiNWs were synthesized through a metal-assisted electroless etching process and functionalized with reduced graphene oxide (rGO) flakes through a chemical absorption process. Here, the rGO not only functions as a physical protection layer to isolate the SiNWs from the harsh electrochemical environment but also serves as a charge mediator to facilitate the charge separation and transport processes. Furthermore, the rGO may also function as a redox catalyst to ensure efficient utilization of photo-carriers for the desired chemical reactions. Photocatalytic dye degradation studies show that the photoactivity of the heterostructures can be significantly enhanced with an initial activation process and maintained without apparent decay over repeated reaction cycles. Electrochemical and photo- electrochemical studies indicate that the enhanced photoactivity and photostability can be attributed to the more efficient separation of photoexcited charge carriers in SiNWs and the reduced self-oxidation of the surface of the SiNWs during the photocatalytic dye degradation process. | 20 | Indonesian Throughflow in an eddy-resolving ocean model显示文摘The performance of the eddy-resolving LICOM2.0 in simulating the Indonesian Throughflow has been evaluated against the INSTANT data in the present study.The mean vertical structures of the along strait velocities are simulated well in LICOM2.0,but the large velocities at the bottom of the Lifamatola Passage and the Timor Passage cannot be reproduced by LICOM2.0.The causes are considered to be both the errors in the topography and the tidal mixing at the bottom.Despite several biases in the mean velocities,the mean inflow and outflow volume transports in LICOM2.0 are almost identical to the INSTANT data.Compared with the lower resolution LICOM,the most significant improvement is the better simulation of the partitions of the inflow and outflow transports in individual straits.The outflow for low-resolution LICOM is mainly through the Ombai and Lombok Strait,whereas that for LICOM2.0 is mainly through the Timor Passage.The variability of the vertical structure of velocities and the volume transport are also investigated.LICOM2.0 overestimates the magnitude of the upper-layer currents and the amplitude of the variation.We also found that the largest correlation coefficient occurs in the shallowest strait,the Lombok,whereas the lowest occurs in the Timor Passage,especially in the upper layer.The latter may be caused by the unrealistic transport through the Torres Strait in LICOM2.0. | FENG Xue LIU HaiLong WANG FuChang YU YongQiang YUAN DongLiang | 2013 | Chinese Science Bulletin2013,58,35: | 4 |
