1Characterization and Fine Mapping of a Novel Rice Narrow Leaf Mutant nal9显示文摘A narrow leaf mutant was isolated from transgenic rice(Oryza sativa L.)lines carrying a T-DNA insertion.The mutant is characterized by narrow leaves during its whole growth period,and was named nal9(narrow leaf 9).The mutant also has other phenotypes,such as light green leaves at the seedling stage,reduced plant height,a small panicle and increased tillering.Genetic analysis revealed that the mutation is controlled by a single recessive gene.A hygromycin resistance assay showed that the mutation was not caused by T-DNA insertion,so a map-based cloning strategy was employed to isolate the nal9 gene.The mutant individuals from the F2generations of a cross between the nal9 mutant and Longtepu were used for mapping.With 24 F2mutants,the nal9 gene was preliminarily mapped near the marker RM156 on the chromosome 3.New INDEL markers were then designed based on the sequence differences between japonica and indica at the region near RM156.The nal9 gene was fnally located in a 69.3 kb region between the markers V239B and V239G within BAC OJ1212_C05 by chromosome walking.Sequence and expression analysis showed that an ATP-dependent Clp protease proteolytic subunit gene(ClpP)was most likely to be the nal9 gene.Furthermore,the nal9 mutation was rescued by transformation of the ClpP cDNA driven by the 35S promoter.Accordingly,the ClpP gene was identifed as the NAL9 gene.Our results provide a basis for functional studies of NAL9 in future work.Wei Li Chao Wu Guocheng Hu Li Xing Wenjing Qian Huamin Si Zongxiu Sun Xingchun Wang Yaping Fu Wenzhen Liu 2013Journal of Integrative Plant Biology2013,55,11:8
2A comparative study on different regeneration processes of Pt-Sn/γ-Al2O3 catalysts for propane dehydrogenation显示文摘Three different regeneration processes including hydrogen or nitrogen purging and coke-burning treatment were used to restore the Pt-Sn/γ-Al_2O_3 catalysts, through which propane dehydrogenation reaction was performed in a consecutive reaction-regeneration mode. It was found that the catalyst using hydrogen regeneration showed the best stability compared with those regenerated by nitrogen purging and coke-burning treatment, suggesting that hydrogen regeneration is an effective approach for maintaining the performance of Pt-Sn/γ-Al_2O_3 catalysts in propane dehydrogenation reaction. The effect of different regeneration atmospheres on the metal active center and the coke deposition was investigated by XRD,TEM, N_2-physisorption, TPO, TG and Raman technologies, and the results revealed that hydrogen or nitrogen regeneration resulted in little impact on the size and structure of metal active center, retaining the effective Pt Sn phase over the catalyst. Moreover, hydrogen regeneration not only removed the low dense components of the coke, but also altered the property of the residual coke through hydrogenation, leading to a higher mobility of coke, and thus a higher accessibility of the metal active centers. Whereas nitrogen regeneration only removed the low dense components of the coke. 3The role of China's terrestrial carbon sequestration 2010-2060 in offsetting energy-related CO_(2) emissions显示文摘Energy consumption dominates annual CO_(2) emissions in China.It is essential to significantly reduce CO_(2) emissions from energy consumption to reach national carbon neutrality by 2060,while the role of terrestrial carbon sequestration in offsetting energy-related CO_(2) emissions cannot be underestimated.Natural climate solutions(NCS),including improvements in terrestrial carbon sequestration,represent readily deployable options to offset anthropogenic greenhouse gas emissions.However,the extent to which China’s terrestrial carbon sequestration in the future,especially when target-oriented managements(TOMs)are implemented,can help to mitigate energy-related CO_(2) emissions is far from certain.By synthesizing available findings and using several parameter-sparse empirical models that have been calibrated and/or fitted against contemporary measurements,we assessed China’s terrestrial carbon sequestration over 2010-2060 and its contribution to offsetting national energy-related CO_(2) emissions.We show that terrestrial C sequestration in China will increase from 0.375±0.056(mean±standard deviation)Pg C yr^(-1) in the 2010s to 0.458±0.100 Pg C yr^(-1)under RCP2.6 and 0.493±0.108 Pg C yr^(-1) under the RCP4.5 scenario in the 2050s,when TOMs are implemented.The majority of carbon sequestration comes from forest,accounting for 67.8-71.4%of the total amount.China’s terrestrial ecosystems can offset 12.2-15.0%and 13.4-17.8%of energy-related peak CO_(2) emissions in 2030 and 2060,respectively.The implementation of TOMs contributes 11.9%of the overall terrestrial carbon sequestration in the 2020s and 23.7%in the 2050s.The most likely strategy to maximize future NCS effectiveness is a full implementation of all applicable cost-effective NCS pathways in China.Our findings highlight the role of terrestrial carbon sequestration in offsetting energy-related CO_(2) emissions and put forward future needs in the context of carbon neutrality.Yao Huang Wenjuan Sun Zhangcai Qin Wen Zhang Yongqiang Yu Tingting Li Qing Zhang Guocheng Wang Lingfei Yu Yijie Wang Fan Ding Ping Zhang 2022National Science Review2022,9,8:4

10Most root-derived carbon inputs do not contribute to long-term global soil carbon storage显示文摘Plant root-derived carbon(C)inputs(I_(root))are the primary source of C in mineral bulk soil.However,a fraction of I_(root)may lose quickly(I_(loss),e.g.,via rhizosphere microbial respiration,leaching and fauna feeding)without contributing to long-term bulk soil C storage,yet this loss has never been quantified,particularly on a global scale.In this study we integrated three observational global data sets including soil radiocarbon content,allocation of photo synthetically assimilated C,and root biomass distribution in 2,034 soil profiles to quantify I_(root)and its contribution to the bulk soil C pool.We show that global average I_(root)in the 0-200 cm soil profile is 3.5 Mg ha^(-1)yr^(-1),~80%of which(i.e.,I_(loss))is lost rather than co ntributing to long-term bulk soil C storage.I_(root)decreases exponentially with soil depth,and the top 20 cm soil contains>60%of total I_(root).Actual C input contributing to long-term bulk soil storage(i.e.,I_(root)-I_(loss))shows a similar depth distribution to I_(root).We also map I_(loss)and its depth distribution across the globe.Our results demonstrate the global significance of direct C losses which limit the contribution of I_(root)to bulk soil C storage;and provide spatially explicit data to facilitate reliable soil C predictions via separating direct C losses from total root-derived C inputs.Guocheng WANG Liujun XIAO Ziqi LIN Qing ZHANG Xiaowei GUO Annette COWIE Shuai ZHANG Mingming WANG Songchao CHEN Ganlin ZHANG Zhou SHI Wenjuan SUN Zhongkui LUO 2023Science China Earth Sciences2023,66,5:0
11Inhibition of VEGF-A expression in hypoxia-exposed fetal retinal microvascular endothelial cells by exosomes derived from human umbilical cord mesenchymal stem cells显示文摘Objective:This study aimed to investigate the potential of human umbilical cord mesenchymal stem cell(hucMSC)-derived exosomes(hucMSC-Exos)in inhibiting hypoxia-induced cell hyper proliferation and overexpression of vascular endothelial growth factor A(VEGF-A)in immature human fetal retinal microvascular endothelial cells(hfRMECs).Methods:Exosomes were isolated from hucMSCs using cryogenic ultracentrifugation and characterized through various techniques,including transmission electron microscopy,nanoparticle tracking analysis,bicinchoninic acid assays,and western blotting.The hfRMECs were identified using von Willebrand factor(vWF)co-staining and divided into four groups:a control group cultured under normoxic condition,a hypoxic model group,a hypoxic group treated with low-concentration hucMSC-Exos(75μg/mL)and a hypoxic group treated with high-concentration hucMSC-Exos(100μg/mL).Cell viability and proliferation were assessed using Cell Counting Kit-8(CCK-8)assay and EdU(5-ethynyl-2′-deoxyuridine)assay respectively.Expression levels of VEGF-A were evaluated using RT-PCR,western blotting and immunofluorescence.Results:Hypoxia significantly increased hfRMECs’viability and proliferation by upregulating VEGF-A levels.The administration of hucMSC-Exos effectively reversed this response,with the high-concentration group exhibiting greater efficacy compared to the lowconcentration group.Conclusion:In conclusion,hucMSC-Exos can dose-dependently inhibit hypoxia-induced hyperproliferation and VEGF-A overexpression in immature fetal retinal microvascular endothelial cells.JING LI WANWAN FAN LILI HAO YONGSHENG LI GUOCHENG YU WEI SUN XIANQIONG LUO JINGXIANG ZHONG 2023BIOCELL2023,47,11:0
12Efficient stereoselective hydroxylation of deoxycholic acid by the robust whole-cell cytochrome P450 CYP107D1 biocatalyst显示文摘Deoxycholic acid(DCA)has been authorized by the Federal Drug Agency for cosmetic reduction of redundant submental fat.The hydroxylated product(6β-OH DCA)was developed to improve the solubility and pharmaceutic properties of DCA for further applications.Herein,a combinatorial catalytic strategy was applied to construct a powerful Cytochrome P450 biocatalyst(CYP107D1,OleP)to convert DCA to 6β-OH DCA.Firstly,the weak expression of OleP was significantly improved using pRSFDuet-1 plasmid in the E.coli C41(DE3)strain.Next,the supply of heme was enhanced by the moderate overexpression of crucial genes in the heme biosynthetic pathway.In addition,a new biosensor was developed to select the appropriate redox partner.Furthermore,a cost-effective whole-cell catalytic system was constructed,resulting in the highest reported conversion rate of 6β-OH DCA(from 4.8%to 99.1%).The combinatorial catalytic strategies applied in this study provide an efficient method to synthesize high-value-added hydroxylated compounds by P450s.Chixiang Sun Baodong Hu Yanchun Li Zhimeng Wu Jingwen Zhou Jianghua Li Jian Chen Guocheng Du Xinrui Zhao 2023Synthetic and Systems Biotechnology2023,8,4:0
