1The Reference Genome of Tea Plant and Resequencing of 81 Diverse Accessions Provide Insights into Its Genome Evolution and Adaptation显示文摘Tea plant is an important economic crop,which is used to produce the world's oldest and most widely consumed tea beverages.Here,we present a high-quality reference genome assembly of the tea plant(Camellia sinensis var.sinensis)consisting of 15 pseudo-chromosomes.LTR retrotransposons(LTR-RTs)account for 70.38%of the genome,and we present evidence that LTR-RTS play critical roles in genome size expansion and the transcriptional diversification of tea plant genes through preferential insertion in promoter regions and introns.Genes,particularly those coding for terpene biosynthesis pro-teins,associated with tea aroma and stress resistance were significantly amplified through recent tandem duplications and exist as gene clusters in tea plant genome.Phylogenetic analysis of the sequences of 81 tea plant accessions with diverse origins revealed three well-differentiated tea plant populations,support-ing the proposition for the southwest origin of the Chinese cultivated tea plant and its later spread to western Asia through introduction.Domestication and modern breeding left significant signatures on hundreds of genes in the tea plant genome,particularly those associated with tea quality and stress resis-tance.The genomic sequences of the reported reference and resequenced tea plant accessions provide valuable resources for future functional genomics study and molecular breeding of improved cul-tivars of tea plants.Enhua Xia Wei Tong Yan Hou Yanlin An Linbo Chen Qiong Wu Yunlong Liu Jie Yu Fangdong Li Ruopei Li Penghui Li Huijuan Zhao Ruoheng Ge Jin Huang Ali Inayat Mallano Yanrui Zhang Shengrui Liu Weiwei Deng Chuankui Song Zhaoliang Zhang Jian Zhao Shu Wei Zhengzhu Zhang Tao Xia Chaoling Wei Xiaochun Wan 2020Molecular Plant2020,13,7:51
2Maspin expression and its clinicopathological significance in tumorigenesis and progression of gastric cancer显示文摘AIM:To investigate maspin expression in tumorigenesis and progression of gastric cancer and to explore its relevant molecular mechanisms.METHODS:Formalin-fixed and paraffin-embedded tissues from normal mucosa (n=182), dysplasia (n=69), cancer (n=l13) of the stomach were studied for maspin expression by immunohistochemistry. Microvessel density (MVD) in gastric cancer was labeled using anti-CD34 antibody. Maspin expression was compared with clinical parameters and MVD of tumors. Caspase-3 expression was also detected in gastric carcinoma by immunohistochemistry. The relationship between Caspase-3 and maspin expression was concerned as well.RESULTS:The positive rates of maspin expression were 79.8% (145/182), 75.4%(52/69) and 50.4%(57/113) in normal mucosa, dysplasia and cancer of the stomach,respectively.Cancer less frequently expressed maspin than normal mucosa and dysplasia (P<0.05).Maspin expression showed a significantly negative association with invasive depth, metastasis, Lauren's and Nakamura's classification (P<0.05),but not with tumor size, Borrmann's classification,growth pattern or TNIVl staging (P>0.05). The positive rate of Caspase-3 was significantly lower in gastric cancer than in normal gastric mucosa (P<0.05,32.7% vs 50.4%). It was noteworthy that maspin expression was negatively correlated with MVD, but positively correlated with expression of Caspase-3 in gastric cancer (P<0.05).CONCLUSION:Down-regulated maspin expression is a late molecular event in gastric carcinogenesis. Reduced expression of maspin contributes to progression of gastric cancer probably by inhibiting cell adhesion, enhandng cell mobility,decreasing cell apoptosis and facilitating angiogenesis.Additionally altered expression of maspin underlies the molecular mechanism of differentiation of gastric cancer and supports the different histogenetic pathways of intestinal and diffuse gastric cancers. 4Intercropping of Sonchus asper and Vicia faba affects plant cadmium accumulation and root responses显示文摘The cadmium(Cd)pollution of farmland soil is serious in the world.The present study investigated the effects of intercropping Vicia faba and the hyperaccumulator Sonchus asper on the Cd accumulation and root responses(morphology and secreted organic acids)of plants grown on soil from a mining area in Yunnan Province,China,under different Cd stress levels(0,50,100,and 200 mg kg-1).Intercropping increased the biomass of both S.asper and V.faba,as well as the Cd accumulation and Cd transfer coefficient from roots to shoots of S.asper,but decreased those of V.faba in the 200 mg kg-1Cd treatment.The Cd concentrations in roots,shoots,and grains from intercropped V.faba plants were positively correlated(P<0.05)with the N,N-diethyl phenylacetamide-extractable Cd content in soil.In the 50 mg kg-1Cd treatment,intercropping decreased citric and malic acids in root secretions of S.asper.Intercropped V.faba secreted more citric,oxalic,acetic,and malic acid compared with S.asper.Intercropping also increased root length and root surface area of both S.asper and V.faba.At 50 mg kg-1Cd,root length for S.asper was higher than that at 0 mg kg-1Cd,whereas V.faba roots had significantly decreased length and mean diameter.Intercropping of S.asper and V.faba is suggested as an in situ phytoremediation strategy of Cd-contaminated soils and may improve the food quality of V.faba.Yanqun ZU Li QIN Fangdong ZHAN Jiong WU Yuan LI Jianjun CHEN Jixiu WANG Wenyou HU 2020Pedosphere2020,30,4:6
5Construction of a bacterial artificial chromosome library of S-type CMS maize mitochondria显示文摘In order to isolate mitochondrial genes easily, we have developed a new method to construct S-type CMS maize mitochondrial gene library by means of embedding mitochondria and enzymatic digesting mitochondria in situ, preparing mtDNA by electrophoresis, digesting LMP agarose with β-agarase, using BAC vector and electroporation. About 2 500 white clones of Mo17 CMS-J mitochondrial gene library were obtained with the average size of 18.24 kb, ranging from 5 to 40 kb, 63.6% inserts came from mitochondrial genome and represented 48 × mitochondrial genome equivalents. All the probes had detected the positive clones in the gene library. 6Electroporation: A New Approach Enhancing Antitumor Effects of Cytoxan显示文摘Electrochemotherapy (ECT) is a novel cancer treatment in which electric pulses (EPs) inducing cell membrane pored (electroporation) are used as a means of delivering antitumor drugs to the cytoplasm of cancer cells. In vitro, with scan electromicroscope (SEM) and Trypan blue staining examination, the best parameter of EPs of electroporation is studied by the S-180 cells exposed to EP with various voltages, pulses , capacitance. The best parameter of EP of electroporation is 600V/cm, 6 pulses, 10 μF. In the in vivo study, ECT is studied with the Cytoxan (CTX) injected directly into the tumor followed immediately by a local EP at the tumor site. Four parameters, which include the tumor inhibitory ratio, the curing ratio and the vas capillare of tumor, the tumor’s histopathological characteristics are determined and compared among the ECT group, the control group, the EP-only group and the drug-only group. 9Validation of an operational forecasting system of sea dike risk in the southern Zhejiang Province, South China显示文摘In this study,an operational forecasting system of sea dike risk in the southern Zhejiang Province,South China was developed based on a coupled storm-surge and wave model.This forecasting system is important because of the high cost of storm-surge damage and the need for rapid emergency planning.A comparison with astronomical tides in 2016 and the validation of storm surges and high water marks of 20 typhoons verified that the forecast system has a good simulation ability.The system can forecast relatively realistic water levels and wave heights as shown under the parametric atmospheric forces simulated in a case study;the sea dikes in credible high risk were mainly located in the estuaries,rivers,and around the islands in the southern Zhejiang.Therefore,the forecast system is applicable in the southern Zhejiang with a support to the effective prevention from typhoon storm-surge damage.LI Tao WANG Fangdong HOU Jingming CHE Zhumei DONG Jianxi 2019Journal of Oceanology and Limnology2019,37,6:2
10Differential SW16.1 allelic effects and genetic backgrounds contributed to increased seed weight after soybean domestication显示文摘Although seed weight has increased following domestication from wild soybean(Glycine soja) to cultivated soybean(Glycine max), the genetic basis underlying this change is unclear. Using mapping populations derived from chromosome segment substitution lines of wild soybean, we identified SW16.1 as the causative gene underlying a major quantitative trait locus controlling seed weight.SW16.1 encodes a nucleus-localized LIM domaincontaining protein. Importantly, the GsSW16.1 allele from wild soybean accession N24852 had a negative effect on seed weight, whereas the GmSW16.1 allele from cultivar NN1138-2 had a positive effect. Gene expression network analysis,reverse-transcription quantitative polymerase chain reaction, and promoter-luciferase reporter transient expression assays suggested that SW16.1 regulates the transcription of MT4, a positive regulator of seed weight. The natural variations in SW16.1 and other known seed weight genes were analyzed in soybean germplasm. The SW16.1 polymorphism was associated with seed weight in 247 soybean accessions, showing much higher frequency of positive-effect alleles in cultivated soybean than in wild soybean. Interestingly,gene allele matrix analysis of the known seed weight genes revealed that G. max has lost 38.5%of the G. soja alleles and that most of the lost alleles had negative effects on seed weight. 12Impact of the Qinghai-Tibet Railway on Population Genetic Structure of the Toad-Headed Lizard,Phrynocephalus vlangalii显示文摘Using data from nine microsatellite DNA loci and a population genetic approach,we evaluate the barrier effect of the Qinghai-Tibet Railway on toad-headed lizard,Phrynocephalus vlangalii. The study area is along a 20 km stretch of the railway on northern Qinghai-Tibet Plateau,and this section of the railway was constructed between 1958–1979. Both assignment tests and analysis of molecular variance(AMOVA) were used for data analysis. We found significant genetic differentiation between the populations from the study area and those from a further southeastern area,which are separated by a 20 km gap. This suggests the existence of population substructure at a fine-scale. However,we did not detect any difference between samples from the western and eastern sides of the railway within the study area,and concluded that the railway may not impose a significant barrier effect on these lizard populations at the present time. Available suitable habitat alongside the railway and bridge underpasses may have facilitated the gene exchange between the sides. The relatively short time since the completion of the railway may not allow the differentiation to accumulate to a detectable level.
