1From mandatory icebreaker guiding to a permission regime: changes to the new Russian legislation of the Northern Sea Route显示文摘This article focuses on two issues. The first concerns definitions of the Northern Sea Route(NSR) in old and new Russian legislation, and the second relates to Russian rules on icebreaker guiding. Based on a comprehensive comparative analysis of relevant Russian legal provisions enacted in 2013 and previous laws in this area, we offer the following conclusions.(1) Our legal analysis indicates that Russia's view of the NSR as a historical national transportation route has not changed. However, the new law redefines the scope and coverage of the NSR, which now comprises the internal waters, territorial sea, adjacent zone, and the exclusive economic zone of the Russian Federation. In fact, the new law resolves previous ambiguity regarding extension of the NSR boundary to the high seas.(2) Based on an analysis of the new rules on icebreaker guiding, the article concludes that NSR is transitioning from a mandatory icebreaker guiding regime into a permit regime. This is particularly evident in its provision of a concrete, practical, and predictable clause on permissible or impermissible conditions relating to independent navigation. According to the new rules, it is possible for foreign ships to undertake independent navigation in the NSR. 2Ammonium Impacts on Vibrations of Hydroxyl and Lattice of Phengite at High Temperature and High Pressure显示文摘Dehydration in hydrous phases of the downgoing slab controls water release processes in subduction zones. Interplay between volatiles in hydrous minerals has complicated the previous knowledge of their dehydration. Phengite is an essential mineral carrying both hydrogen and nitrogen to the deep Earth. To further understand the link between nitrogen and dehydration process of phengite at an atomic level, we here carry out high temperature and high pressure vibrational spectroscopic investigations on hydroxyl and lattice of ammonium-bearing and ammonium-free phengites. The results show that heating to 800 oC hardly influences hydroxyl bonding strength, whereas pressure induces strengthening of hydrogen bonding until 10 GPa. Moreover, hydrogen transits between the sites with increasing temperature and pressure. The lattice modes soften with increasing temperature and stiffen under compression. Ammonium has no effect on hydroxyl bonding strength, but hinders hydrogen transition at high temperatures and high pressures. Ammonium does not influence the lattice at high pressures either, but delays softening of the lattice at high temperatures. 3Reactivity,Regioselectivity,and Synthetic Application of 2-Pyrenyl Units in Scholl Reactions显示文摘We herein report the reactivity and regioselectivity of 2-pyrenyl as a coupling unit in Scholl reactions.On the basis of the Scholl reactions of hexaarylbenzene substrates,we have found that pyrenyl units are preferably oxidized over naphthyl and phenyl units under appropriate Scholl reaction conditions,allowing divergent synthesis through a highly controllable intramolecular coupling sequence.We find that the C1 and C3 positions of the 2-pyrenyl unit are the favorable sites for intramolecular coupling while C4 is not reactive to allow further coupling.The reactivity and regioselectivity pattern can be explained by the spin density distribution,which shows that carbon-carbon bonds preferably form at sites with higher positive spin density.Guided by these findings,we successfully synthesized a double helicene and a sextuple helicene through the controlled Scholl reactions of 2-pyrenyl units.Sai Ho Pun Ethan Chi Ho Wen Zeming Xia Han Chen Felix RFischer Qian Miao 2023CCS Chemistry2023,5,9:0
4A PD-L1-targeting chimeric switch receptor enhances efficacy of CAR-T cell for pleural and peritoneal metastasis显示文摘Pleural and peritoneal metastasis accompanied by malignant pleural effusion(MPE)or malignant ascites(MA)is frequent in patients with advanced solid tumors that originate from the lung,breast,gastrointestinal tract and ovary.Regional delivery of CAR-T cells represents a new strategy to control tumor dissemination in serous cavities.However,malignant effusions constitute an immune-suppressive environment that potentially induces CAR-T cell dysfunction.Here,we demonstrated that the anti-tumor cytotoxicity of conventional 2nd-generation CAR-T cells was significantly inhibited by both the cellular and non-cellular components of MPE/MA,which was primarily attributed to impaired CAR-T cell proliferation and cytokine production in MPE/MA environment.Interestingly,we found that PD-L1 was widely expressed on freshly-isolated MPE/MA cells.Based on this feature,a novel PD-L1-targeting chimeric switch receptor(PD-L1.BB CSR)was designed,which can bind to PD-L1,switching the inhibitory signal into an additional 4-1BB signal.When co-expressed with a 2nd-generation CAR,PD-L1.BB CSR-modified CAR-T cells displayed superior fitness and enhanced functions in both culture medium and MPE/MA environment,causing rapid and durable eradication of pleural and peritoneal metastatic tumors in xenograft models.Further investigations revealed elevated expressions of T-cell activation,proliferation,and cytotoxicity-related genes,and we confirmed that PD-L1 scFv and 4-1BB intracellular domain,the two important components of PD-L1.BB CSR,were both necessary for the functional improvements of CAR-T cells.Overall,our study shed light on the clinical application of PD-L1.BB CSR-modified dual-targeting CAR-T cells.Based on this study,a phase I clinical trial was initiated in patients with pleural or peritoneal metastasis(NCT04684459).Qizhi Ma Xia He Benxia Zhang Fuchun Guo Xuejin Ou Qiyu Yang Pei Shu Yue Chen Kai Li Ge Gao Yajuan Zhu Diyuan Qin Jie Tang Xiaoyu Li Meng Jing Jian Zhao Zeming Mo Ning Liu Yao Zeng Kexun Zhou Mingyang Feng Weiting Liao Wanting Lei Qiu Li Dan Li Yongsheng Wang 2022Signal Transduction and Targeted Therapy2022,7,12:0
