| 1 | Baicalin prevents LPS-induced activation of TLR4/NF-κB p65 pathway and inflammation in mice via inhibiting the expression of CD14显示文摘Previous studies have shown that baicalin,an active ingredient of the Chinese traditional medicine Huangqin,attenuates LPS-induced inflammation by inhibiting the activation of TLR4/NF-κBp65 pathway,but how it affects this pathway is unknown.It has been shown that CD14 binds directly to LPS and plays an important role in sensitizing the cells to minute quantities of LPS via chaperoning LPS molecules to the TLR4/MD-2 signaling complex.In the present study we investigated the role of CD14 in the anti-inflammatory effects of baicalin in vitro and in vivo.Exposure to LPS(1μg/mL)induced inflammatory responses in RAW264.7 cells,evidenced by marked increases in the expression of MHC II molecules and the secretion of NO and IL-6,and by activation of MyD88/NF-κB p65 signaling pathway,as well as the expression of CD14 and TLR4.These changes were dose-dependently attenuated by pretreatment baicalin(12.5–50μM),but not by baicalin post-treatment.In RAW264.7 cells without LPS stimulation,baicalin dose-dependently inhibit the protein and mRNA expression of CD14,but not TLR4.In RAW264.7 cells with CD14 knockdown,baicalin pretreatment did not prevent inflammatory responses and activation of MyD88/NF-κB p65 pathway induced by high concentrations(1000μg/mL)of LPS.Furthermore,baicalin pretreatment also inhibited the expression of CD14 and activation of MyD88/NF-κB p65 pathway in LPS-induced hepatocyte-derived HepG2 cells and intestinal epithelial-derived HT-29 cells.In mice with intraperitoneal injection of LPS and in DSS-induced UC mice,oral administration of baicalin exerted protective effects by inhibition of CD14 expression and inflammation.Taken together,we demonstrate that baicalin pretreatment prevents LPS-induced inflammation in RAW264.7 cells in CD14-dependent manner.This study supports the therapeutic use of baicalin in preventing the progression of LPS-induced inflammatory diseases. | Ya-jun Fu Bo Xu Shao-wei Huang Xia Luo Xiang-liang Deng Shuang Luo Chang Liu Qing Wang Jin-yan Chen Lian Zhou | 2021 | Acta Pharmacologica Sinica2021,42,1: | 31 |

| 6 | Efficacy of intramuscular matrine in the treatment of chronic hepatitis B显示文摘BACKGROUND: Hepatitis B virus (HBV) infection, a glo- bal public health problem, is the leading cause of cirrhosis and hepatocellular carcinoma (HCC) worldwide. There are more than 350 million HBV carriers in the world and up to one million die annually due to hepatitis B associated liv- er disease. So far no optimal treatment is available for pa- tients with chronic hepatitis B. In the paper we investigated the efficacy of intramuscular matrine in the treatment of chronic hepatitis B. METHODS: One hundred and twenty patients with chronic hepatitis B were randomly divided into matrine treatment group (n =60) and control group (n =60). The patients of the matrine group were given intramuscularly with matrine (an alkaloid extracted from a traditional Chinese herb Radix Sophorae Flavescentis by Guangzhou Ming Xing Pharmaceu cal Factory, Guangzhou, China) of 100 mg daily for 90 days in addition to conventional liver-protective drugs in- cluding glucurone, inosine, compound vitamin B and caryophyllin. The control group received conventional liv- er-protective drugs alone. Clinical manifestations and labo- ratory parameters including liver biochemistry and serum hepatitis B virus markers were monitored before and after treatment in the two groups. RESULTS: Significant differences were seen between the two groups in terms of improvement of clinical symptoms and signs, recovery of liver functions, and serum conver- sion from hepatitis Be antigen to HBe antibody and from positive to negative serum HBV DNA (P <0.05-0.01). The result of the matrine group was more marked than that of the control group. Serious side-effects were not observed except mild pain at the site of injection of matrine in a few patients. | 12 | Influence of mesoscale eddies on primary production in the South China Sea during spring inter-monsoon period显示文摘Mesoscale eddies have been suggested to have an impact on biological carbon fixation in the South China Sea(SCS). However, their overall contribution to primary production during the spring inter-monsoon period is still unknown. Based on large-scale biological and environmental in situ observations and synchronous remote sensing data, the distribution patterns of phytoplankton biomass and the primary production, and the role of mesoscale eddies in regulating primary production in different eddy-controlled waters were investigated. The results suggested that the surface chlorophyll a concentrations and water column integrated primary production(IPP) are significantly higher in cyclonic eddies and lower in the anticyclonic eddies as compared to that in non-eddy waters. Although eddies could affect various environmental factors, such as nutrients, temperature and light availability, nutrient supply is suggested to be the most important one through which mesoscale eddies regulated the distribution patterns of phytoplankton biomass and primary production. The estimated IPP in cyclonic and anticyclonic eddies are about 29.5% higher and 16.6% lower than the total average in the whole study area, respectively, indicating that the promotion effect of mesoscale cold eddies on the primary production was much stronger than the inhibition effect of the warm eddies per unit area. | 14 | Oroxindin inhibits macrophage NLRP3 inflammasome activation in DSS-induced ulcerative colitis in mice via suppressing TXNIP-dependent NF-κB pathway显示文摘Oroxindin is a flavonoid isolated from the traditional Chinese medicine Huang-Qin,which has shown various pharmacological activities including anti-inflammatory,antitumor,antioxidant,etc.Thus far,the effect of oroxindin on colonic inflammation and the underlying mechanism remain unknown.In this study,we investigated the tissue distribution of oroxindin and its therapeutic effects on ulcerative colitis(UC)as well as the underlying mechanisms.UC model was established in mice by administrating dextran sulfate sodium(DSS)in drinking water for 7 d.We first showed that oroxindin was largely absorbed by the colon as an active ingredient after normal mice received Huang-Qin-Tang,a traditional Chinese medicine decoction.UC mice were then treated with oroxindin(12.5,25,50 mg-kg^-1·d^-1,i.g.)for 10 d.We found that oroxindin treatment greatly suppressed massive macrophages infiltration and attenuated pathological changes in colonic tissue.Furthermore,oroxindin treatment significantly inhibited the generation of IL-13 and IL-18 in the colon via inhibiting the nucleotide-binding oligomerization domain-like receptor 3(NLRP3)inflammasome formation and activation.In cultured macrophages,LPS induced NLRp3 inflammasome formation and caspase-1 activation,which were suppressed by oroxindin(12.5-50μM).In LPS-treated macrophages,oroxindin dose-dependently restored the expression of TXNIP protein,leading to suppressing TXNIP-dependent NF-κB activation.In conclusion,these results demonstrate that oroxindin could be absorbed by the colon and attenuate inflammatory responses via inhibiting NLRP3 inflammasome formation and activation,which is related to the inhibitory effect on TXNIP-dependent NF-κB-signaling pathway.Hence,oroxindin has the potential of becoming an effective drug for treating UC. | Qi Liu Rui Zuo Kai Wang Fei-fei Nong Ya-jun Fu Shao-wei Huang Zeng-feng Pan Yi Zhang Xia Luo Xiang-liang Deng Xiao-xue Zhang Lian Zhou Yang Chen | 2020 | Acta Pharmacologica Sinica2020,41,6: | 15 |

| 16 | Baicalein ameliorates ulcerative colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s显示文摘Ulcerative colitis(UC)is a chronic inflammatory disease of the gastrointestinal tract,which is closely related to gut barrier dysfunction.Emerging evidence shows that interleukin-22(IL-22)derived from group 3 innate lymphoid cells(ILC3s)confers benefits on intestinal barrier,and IL-22 expression is controlled by aryl hydrocarbon receptor(AhR).Previous studies show that baicalein protects the colon from inflammatory damage.In this study we elucidated the molecular mechanisms underlying the protective effect of baicalein on intestinal barrier function in colitis mice.Mice were administered baicalein(10,20,40 mg·kg^(-1)·d^(-1),i.g.)for 10 days;the mice freely drank 3%dextran sulfate sodium(DSS)on D1-D7 to induce colitis.We showed that baicalein administration simultaneously ameliorated gut inflammation,decreased intestinal permeability,restored tight junctions of colons possibly via promoting AhR/IL-22 pathway.Co-administration of AhR antagonist CH223191(10 mg/kg,i.p.)partially blocked the therapeutic effects of baicalein in colitis mice,whereas AhR agonist FICZ(1μg,i.p.)ameliorated symptoms and gut barrier function in colitis mice.In a murine lymphocyte line MNK-3,baicalein(5-20μM)dose-dependently increased the expression of AhR downstream target protein CYP1A1,and enhanced IL-22 production through facilitating AhR nuclear translocation,these effects were greatly diminished in shAhR-MNK3 cells,suggesting that baicalein induced IL-22 production in AhR-dependent manner.To further clarify that,we constructed an in vitro system consisting of MNK-3 and Caco-2 cells,in which MNK-3 cell supernatant treated with baicalein could decrease FITC-dextran permeability and promoted the expression of tight junction proteins ZO-1 and occluding in Caco-2 cells.In conclusion,this study demonstrates that baicalein ameliorates colitis by improving intestinal epithelial barrier via AhR/IL-22 pathway in ILC3s,thus providing a potential therapy for UC. | Yan-yang Li Xiao-jing Wang Yu-lin Su Qing Wang Shao-wei Huang Zeng-feng Pan Yan-ping Chen Jun-jie Liang Mei-ling Zhang Xue-qian Xie Zhi-yun Wu Jin-yan Chen Lian Zhou Xia Luo | 2022 | Acta Pharmacologica Sinica2022,43,6: | 14 |
| 17 | Chinese Medicine Sequential Therapy Improves Pregnancy Outcomes after Surgery for Endometriosis-Associated Infertility: A Multicenter Randomized Double-blind Placebo Parallel Controlled Clinical Trial显示文摘Objective:To evaluate the efficacy and safety of Chinese medicine(CM)improving pregnancy outcomes after surgery for endometriosis-associated infertility.Methods:A multicenter,randomized,doubleblind placebo parallel controlled clinical trial was designed.A total of 202 patients who had laparoscopy for endometriosis-associated infertility with qi stagnation and blood stasis syndrome were included and randomly divided into the CM treatment group and placebo control group at a ratio of 1:1 using a central block randomization from May 2014 to September 2017,101 patients in each group.The two groups received continuous intervention at 1–5 days after surgery,for 6 menstrual cycles.Before ovulation,the CM group was treated Huoxue Xiaoyi Granule(活血消异颗粒);after ovulation,Bushen Zhuyun Granule(补肾助孕颗粒)was involved.The control group was treated with placebo.Transvaginal ultrasonography was performed every menstrual cycle during the treatment,and female hormone levels in the follicular and luteal phases were measured during the 1 st,3 rd and 6 th menstrual cycles.The analysis was continued until pregnancy.The primary outcomes were clinical pregnancy rate and pregnancy outcome,and the secondary outcomes were follicular development and endometrial receptivity.Safety evaluations were performed before and after treatment.Results:(1)Clinical pregnancy and live birth rates:the clinical pregnancy and live birth rates of the CM group were significantly higher than those of the placebo group[44.6%(45/101)vs.29.7%(30/101),34.7%(35/101)vs.20.8%(21/101),both P<0.05].(2)Follicle development:the incidence of dominant follicles,rate of cumulative cycle ovulation,and rate of cumulative cycle mature follicle ovulation were significantly higher in the CM group than those in the placebo group[93.8%(350/373)vs.89.5%(341/381),80.4%(275/342)vs.69.1%(253/366),65.8%(181/275)vs 56.1%(142/253),P<0.05 or P<0.01]).The incidence of cumulative cycle luteinized unruptured follicle syndrome was significantly lower in the CM group than in the placebo group[11.7%(40/342)vs.17.8%(65/366),P<0.05).(3)Endometrial receptivity:after treatment,both endometrial types and endometrial blood flow types in the CM group were mainly types A and B,while those in the placebo group were mainly types B and C,with a significant difference between the two groups(both P<0.05).(4)Adverse events:the incidence of adverse events between the two groups was not significantly different(P>0.05).Conclusion:Strategies for activating blood circulation-regulating Gan(Liver)-tonifying Shen(Kidney)sequential therapy can effectively improve the clinical pregnancy rate and live birth rate of endometriosis-associated infertility with qi stagnation and blood stasis after laparoscopy,improve follicular development,promote ovulation,improve endometrial receptivity,while being a safe treatment option.(Trial registration No.NCT02676713). | ZHAO Rui-hua LIU Yong LU Dan WU Ying WANG Xiao-yun LI Wei-li ZENG Cheng MENG Qing-wei LIAN Feng-mei ZHOU Jun SHI Yun SUN Wei-wei HAN Qian TANG Yi SHI Guang | 2020 | Chinese Journal of Integrative Medicine2020,26,2: | 13 |

| 20 | Comorbidity Burden of Dementia:A Hospital-Based Retrospective Study from 2003 to 2012 in Seven Cities in China显示文摘Dementia is increasing dramatically and imposes a huge burden on society. To date, there is a lack of data on the health status of patients with dementia in China. In an attempt to investigate the comorbidity burden of dementia patients in China at the national level, we enrolled 2,938 patients with Alzheimer's disease(AD), vascular dementia(Va D), or other types of dementia, who were admitted to tertiary hospitals in seven regions of China from January2003 to December 2012. The Charlson Comorbidity Index(CCI) was used to evaluate the comorbidity burden of the patients with dementia. Among these patients, 53.4% had AD, 26.3% had Va D, and 20.3% had other types of dementia. The CCI was 3.0 ± 1.9 for all patients,3.4 ± 1.8 for those with Va D, and 3.0 ± 2.1 for those with AD. The CCI increased with age in all patients, andthe length of hospital stay and daily expenses rose with age and CCI. Males had a higher CCI and a longer stay than females. Moreover, patients admitted in the last 5 years of the study had a higher CCI than those admitted in the first 5 years. We found that the comorbidity burden of patients with dementia is heavy.
