| 1 | A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking,one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents.In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches,including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps.The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size,and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families,respectively,accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry.Moreover,the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine,forage,papermaking,and barkcloth making.We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively,these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition.Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making,and as forage.These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18 |
| 2 | Function of the auxin-responsive gene TaSAUR75 under salt and drought stress显示文摘Small auxin-upregulated RNAs(SAURs) are genes regulated by auxin and environmental factors. In this study, we identified a SAUR gene in wheat, Ta SAUR75. Under salt stress,Ta SAUR75 is downregulated in wheat roots. Subcellular localization revealed that Ta SAUR75 was localized in both the cytoplasm and nucleus. Overexpression of Ta SAUR75 increased drought and salt tolerance in Arabidopsis. Transgenic lines showed higher root length and survival rate and higher expression of some stress-responsive genes than control plants under salt and drought stress. Less H_2O_2 accumulated in transgenic lines than in control plants under drought stress. | 3 | RNA-seq analysis of unintended effects in transgenic wheat overexpressing the transcription factor GmDREB1显示文摘The engineering of plants with enhanced tolerance to abiotic stresses typically involves complex multigene networks and may therefore have a greater potential to introduce unintended effects than the genetic modification for simple monogenic traits. For this reason, it is essential to study the unintended effects in transgenic plants engineered for stress tolerance. We selected drought-and salt-tolerant transgenic wheat overexpressing the transcription factor, GmDREB1, to investigate unintended pleiotropic effects using RNA-seq analysis. We compared the transcriptome alteration of transgenic plants with that of wild-type plants subjected to salt stress as a control. We found that GmDREB1 overexpression had a minimal impact on gene expression under normal conditions.GmDREB1 overexpression resulted in transcriptional reprogramming of the salt response,but many of the genes with differential expression are known to mitigate salt stress and contribute incrementally to the enhanced stress tolerance of transgenic wheat. GmDREB1 overexpression did not activate unintended gene networks with respect to gene expression in the roots of transgenic wheat. | 4 | Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL. | Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen | 2023 | Cancer Communications2023,43,2: | 3 |

| 6 | Dual effects of spatial updating in both real and virtual environments显示文摘When people move around in their environment, spatial updating, which is an automatic cognitive process, is essential to ensuring people can keep track of their relations between them and the surrounding objects, and to 'recalculating' the relative position and orientation of those objects with regard to the current position of the persons. Despite the facilitating effect of spatial updating to people's mental representation in most circumstances as demonstrated in most of the existing literature, the effect sometimes can be adversarial. For instance, some research suggested that even though people were asked to ignore their locomotion, it is difficult to suppress updating of the spatial representation during movement. The current two studies were conducted to systematically investigate the dual effects of spatial updating in both real and virtual environments. We used a typical spatial updating paradigm to explore the effects of scene familiarity (familiar vs. novel) and person's locomotion (stationary vs. moving) on change detection accuracy (target object moved or not). | 7 | Metabolic profiling of DREB-overexpressing transgenic wheat seeds by liquid chromatographymass spectrometry显示文摘DREBs are transcription factors that regulate abiotic stress tolerance in plants.Previously,we reported that wheat transgenic lines overexpressing Gm DREB1 showed increased tolerance to drought and salt stress.However,the molecular basis of increased tolerance is still poorly understood,and whether the overexpression of DREB will cause unexpected effects is also of concern.We performed seed metabolic profiling of the genetically modified(GM)wheat T349 and three non-GM cultivars with LC-MS to identify the metabolic basis of stress tolerance and to assess the unexpected effects of exogenous gene insertion.Although we did not note the appearance of novel metabolites or the disappearance of existing metabolites,overexpression of the transcription factor Gm DREB1 in T349 wheat influenced metabolite levels in seeds.Increased levels of stress tolerance-associated metabolites were found in the stress-sensitive non-transgenic acceptor counterpart J19,while metabolites associated with cell membrane structure and stability accumulated in T349.Among these metabolites in T349,most showed levels similar to those in the non-GM wheats.Overexpression of Gm DREB1 in T349 may cause a shift in its metabolic profile leading to down-regulation of several energy-consuming processes to favor increased yield under stress conditions,which is a reasonable expectation of breeders while creating the GM wheat and Gm DREB1 overexpression did not cause unexpected effects in T349 seeds.These results may be helpful for GM crop research and risk assessment. | Fengjuan Niu Qiyan Jiang Xianjun Sun Zheng Hu Lixia Wang Hui Zhang | 2020 | The Crop Journal2020,8,6: | 2 |
| 8 | Antihepatofibrotic effect of Guizhifuling pill(桂枝茯苓丸) on carbon tetrachloride-induced liver fibrosis in mice显示文摘OBJECTIVE: To evaluate the protective effects and the underlying mechanism of Guizhifuling pill(桂枝茯苓丸, GZFL) on carbon tetrachloride(CCl4)-induced hepatic fibrosis in mice. METHODS: Male ICR mice by intraperitoneally administered with 20% CCl4(mixed 1∶4 in soybean oil) to induce liver fibrosis. Mice that underwent CCl4 were orally with GZFL. Using hematoxylin and eosin and Masson staining to examine the pathological changes in liver tissue. Serum biochemical parameters, antioxidant enzyme activity and proinflammatory cytokines was assessed. Nuclear factor-kappa B(NF-κB) pathway and nuclear factor-erythroid 2-related factor 2(Nrf2) family members were evaluated by Western blotting. RESULTS: Our findings indicated that GZFL could effectively suppress the progression of liver fibrosis in mice, which was determined based on the improvement in liver function and reduction of collagen deposition. GZFL treatment also decreased the level of cytokines and increased the activity of antioxidant enzymes in liver tissue. Moreover, GZFL exerted anti-inflammatory and antioxidant effects through regulating the Nrf2-mediated antioxidant system and inhibiting the NF-κB pathway. | 9 | FGFBPI-mediated crosstalk between fibroblasts and pancreatic cancer cells via FGF22/FGFR2 promotes invasion and metastasis of pancreatic cancer显示文摘Fibroblast growth factor-binding protein 1(FGFBP1)promotes fibroblast growth factor(FGF)activity by releasing FGFs from extracellular matrix storage.We previously reported that the tumor suppressor F-box and WD repeat domain-containing 7 suppresses FGFBP1 by reducing expression of c-Myc,which inhibits the proliferation and migration of pancreatic cancer cells.However,the potential mechanism by which FGFBP1 facilitates pancreatic ductal adenocarcinoma(PDAC)remains unexplored.In this study,we focused on the function of FGFBP1 in the interplay between cancer-associated fibroblasts(CAFs)and pancreatic cancer cells(PCCs).Decreased FGF22 expression was detected in CAFs co-cultured with PCCs with FGFBP1 abrogation,which was verified in the cell culture medium by enzyme-linked immunosorbent assay.Active cytokine FGF22 significantly facilitated the migration and invasion of PANC-1 and Mia PaCa-2 cells.The number of penetrating PCCs cocultured with CAFs with FGF22 abrogation was significantly less than that of the control group.Interestingly,higher expressions of FGF22 and fibroblast growth factor receptor 2(FGFR2)were associated with worse prognosis of patients with PDAC and FGFR2,an independent prognostic marker of PDAC.The PANC-1 and Mia PaCa-2 cells with silenced FGFR2 showed weaker invasion and metastasis,even if these cells were simultaneously treated with cytokine FGF22.These results revealed that FGFBPI-mediated interaction between CAFs and PCCs via FGF22/FGFR2 facilitates the migration and invasion of PCCs.FGFR2 could act as a prognostic marker for patients with PDAC. | Zheng Zhang Yi Qin Shunrong Ji Wenyan Xu Mengqi Liu Qiangsheng Hu Zeng Ye Guixiong Fan Xianjun Yu Wensheng Liu Xiaowu Xu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,8: | 2 |
