| 3 | Changes in circulating Foxp3^+ regulatory T cells and interleukin-17-producing T helper cells during HBV-related acute-on-chronic liver failure显示文摘AIM:To longitudinally investigate cytokine gene expression and protein levels in Th17 and Treg cells,to observe T-cell phenotypes during hepatitis B virus(HBV)-related acute-on-chronic liver failure(ACHBLF)and to analyze changes in Th17 and Treg phenotypes during disease progression.METHODS:We measured the expression of seven Th17/Treg differentiation-related genes and serum concentrations of the corresponding cytokines in 18ACHBLF,18 chronic hepatitis B(CHB)disease controls and 10 healthy controls(HCs)by real-time quantitative pCR and enzyme linked immunosorbent assay.peripheral Th17 and Treg cell frequencies were analyzed by flow cytometry.RESULTS:From the onset of ACHBLF,patients presented with a conductive Th17 differentiation cytokine environment accompanied by high Th17 frequency and high serum IL-17 levels,which were sustained throughout the disease course.The Treg-related cytokine IL-2and Foxp3 were also up-regulated from disease onset,and Foxp3 gene expression showed a gradually increasing trend during ACHBLF.The circular phenotype of Treg and Th17 cells showed changes from the onset of ACHGLF.At disease onset,Th17 frequency increased significantly compared with both CHB and HCs,but Treg cell frequency decreased significantly compared with CHB.During the ACHBLF event,Th17 frequency remained higher compared with HCs,but decreased sharply from the peak point to the recovery point;Treg cell frequency increased gradually during the ACHBLF event.Treg and Th17 cell counts correlated with ACHBLF development;in all patients,serum IL-17 levels significantly correlated with patient serum ALT levels.In survivors,Th17 frequency at the onset point and the Treg to Th17 ratio at the peak point correlated with the patient’s model for end stage liver disease(MELD)plus sodium(MELD-Na)score.The Treg to Th17 ratio and the Th17 frequency at onset were significant predictors of patient survival.Low Treg/Th17 cell ratios at the onset predicted poor survival.Survivors exhibited an initial decrease in the circulating Treg/Th17 ratio from the onset to the peak time,and subsequently displayed a continuous increase.CONCLUSION:Treg and Th17 cells showed changes in genes,protein levels and T cell phenotypes during ACHBLF events.An increased Treg/Th17 ratio was associated with the survival of ACHBLF patients. | Xue-Song Liang Cheng-Zhong Li Yin Zhou Wei Yin Ya Yun Liu Wen-Han Fan | 2014 | World Journal of Gastroenterology2014，20，26： | 44 |
| 4 | Inhibition of CXCR4 activity with AMD3100 decreases invasion of human colorectal cancer cells in vitro显示文摘AIM: To investigate the effect and mechanism of blockade of the CXC chemokine receptor-4 (CXCR4) signaling pathway by AMD3100, a small non-peptide CXCR4 inhibitor, on invasion and metastasis of colorectal cancer cells in vitro. METHODS: Human colorectal cancer cell line SW480 was treated with AMD3100 at different final concentrations. 3-(4,5-dimethylthiazole-2-yl)-2.5-dipheny-ltetrazolium bromide (MTT) assay was used to detect the effect of AMD3100 on cell proliferation. The invasion ability of SW480 cells was determined by cell invasion assay kit. In the presence of AMD3100, the CXCL12-mediated migratory response of SW480 cells was tested by classical chemotaxis assays. RT-PCR analysis and Western blotting were used to detect the expression of vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 (MMP-2) and -9 (MMP-9) in SW480 cells. RESULTS: Cell viability was significantly suppressed by AMD3100 in a dose-dependent manner. AMD3100 (100 and 1000 ng/mL) significantly inhibited the invasion ability of SW480 cells. Treatment with AMD3100 markedly reduced the expression of VEGF and MMP-9 but not MMP-2 in SW480 cells. CONCLUSION: The CXCL12/CXCR4 system is an important mediator of proliferation and invasion of CXCR4-expressing colorectal cancer cells. | 8 | Discovery of Paleogene marine stratum along the southern side of Yarlung-Zangbo suture zone and its implications in tectonics显示文摘The Jiachala Formatiom is a new lithostratigraphic unit set up in this paper. This is the first report on the Paleogene marine sequences and microfaunas in Gyangze to the southern side of the Yarlung-Zangbo suture zone. On the basis of fossil abundance, species diversity, dominant species and first and last occurrences of key species, three dinoflagellate assemblages and three polynological assemblages were recognized in the Jiachala Formatiom. They are in ascending order as follows: Apectodinium quinquelatum-Apectodinium hyperacanthum, Cannin- gia chinensis-Palaeoperidinium pyrophorum and Cymatiosphaera reticulosa-Samlandia chla- mydophora dinoflagellate assemblages, and Arliaceoipollenites baculatus-Anacolosidites sub- trudens, Aglaoreidia cyclops-Pinuspollenites microinsigis, and Elaeangnacites asper-Ilexpol- lenites iliacus polynological Assemblages. According to the assemblages, the age of the Jiachala Formation is referred to Paleocene-early Eocene. The preliminary study on the Jiachala Forma- tion shows that this group of Paleogene deposits accumulated in an underfilled peripheral fore- land basin which was in response to the load of the crust thickening resulted from the Asia-India collision. The stratigraphic evidences provided by the evolution of foreland basin indicate that the India-Asia initional collision should occure near the Cretaceous/Paleogene boundary. The de- velopment and evolution of the southern Tibet foreland basin was under the control of the stress field from the subduct-collision between India and Asia and the structure pattern of the basement. Results showed that ecotoxicological effects under the combined pollution were not only related to chemical proper-ties of pollutants but also dependent on the concentration level of pollutants, in particular on the combination of concentrations of pollutants in ecosystems. Additionally, species of organisms, especially the type of ecosystem, determined the influences. | 10 | Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. | 11 | Changes in Waist Circumference and Abdominal Obesity among Chinese Adults over a Ten-year Period显示文摘Objective The aim of this study was to describe changes in waist circumference(WC) and prevalence of abdominal obesity over a period of 10 years among Chinese adults in different socio-economic status(SES). Methods Data derived from the China Nutrition and Health Surveillance during 2002 and 2010-2012. We calculated the mean WC and the prevalence of abdominal obesity by gender, place of residence, SES indicators(education, income, and marital status), and body mass index(BMI) categoriesand used pooled t-tests to assess the differences between the two time periods. Results 26.0% of men and 25.3% of women had abdominal obesity in 2010-2012. The age-adjusted mean WC increased by 2.7 cm among men and 2.1 cm among women; the age-adjusted prevalence of abdominal obesity increased by 7.7% among men and 5.3% among women. The rising trends were observed in all subgroups except for a negative growth in high-income women. People living in rural areas with low education and income and with a BMI of 18.5 to 23.9 kg/m^2 had a greater absolute and relative increase in WC. People living in rural areas with low income had a greater relative increase in abdominal obesity. Conclusion The mean WC and prevalence of abdominal obesity among Chinese adults have increased during the past 10 years. | 13 | N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. | 14 | Comparison of Undernutrition Prevalence of Children under 5 Years in China between 2002 and 2013显示文摘Objective To describe the undernutrition status of children under 5-year in China, and study the trend between 2002 and 2013. Methods The study was based on two national surveys. Undernutrition was determined against WHO's 2006 growth standards. The prevalence in 2013 and 2002 was weighted by China sixth National Population Census(2010). The relationship between undernutrition and gender/age groups/different areas use weighted logistic regression. Results The results indicated the overall prevalence of stunting, underweight, and wasting of Chinese children under 5-year was 8.1%, 2.4%, and 1.9% in 2013, respectively. The prevalence of stunting was higher for children aged 12-47 month, while underweight was higher for children aged 48-59 month. The prevalence of undernutrition was higher in rural areas than in urban areas, especially in poor rural areas. There was a decline of stunting, underweight, and wasting between 2002 and 2013 among the children, with greater reduction in rural areas than in urban areas. Conclusion The prevalence of undernutrition of children under 5-year remains high in rural areas especially in poor rural areas in China. | 17 | Properties and Applications of the Eccentric-gear Drive显示文摘The eccentric-gear can be used for variable speed transmission.But,due to the vibrations caused by variation of backlash in working process,the eccentric-gear is seldom applied in engineering project.There is just a little study about the eccentric-gear.And it is necessary to take a further research on eccentric-gear transmission for the applications in the transplanting mechanism of high-speed rice transplanter.The key of this paper is to extend understanding of three following characteristics of this drive:(1) The rotation-center-distance and the geometrical-center-distance of the two meshing eccentric gears are respectively the hypotenuse and the straight edge of a right triangle in a cycle.(2) The geometrical center line of two meshing gears divides the linking-line of the two rotational centers equally.(3) When two times the eccentricity of gear,the rotation-center-distance and the geometrical-center-distance form a right triangle,the optimal value of rotation-center-distance can be determined.In addition,the kinematic analysis,such as the relationship between contact point and midpoint of the linking-line of two gears' rotational centers,transmission ratio between the driving-gear and the driven-gear,contact angle and coordinate of contact point are be made,further the formula for calculation of contact force,flow chart for writing optimization program and curve for analysis of data are also be developed.The relationship between the rotation-center-distance and the geometrical-center-distance,which is determined by three characteristics mentioned above, is a key for the rational design and application of eccentric-gear transmission.Particularly,the presented right triangle property of eccentric gear drive is an important reference for effective analysis of dynamic characteristic of eccentric-gear mechanism and its reliable design. | ZHAO Yun SUN Liang YU Gaohong | 2011 | Chinese Journal of Mechanical Engineering2011，24，2： | 16 |

In this study, Corynebacterium glutamicum ATCC 13032 was engineered de novo by blocking and attenuating the conversion of L-serine to pyruvate and glycine, releasing the feedback inhibition by L-serine to 3-phosphoglycerate dehydrogenase (PGDH), in combination with the co-expression of 3-phosphoglycerate kinase (PGK) and feedback-resistant PGDH (PGDHr). The resulting strain, SER-8, exhibited a lower specific growth rate and significant differences in L-serine levels from Phase I to Phase V as determined for fed-batch fermentation. The intracellular L-serine pool reached (14.22±1.41) μmol gCDM 1, which was higher than glycine pool, contrary to fermentation with the wild-type strain. Furthermore, metabolic flux analysis demonstrated that the over-expression of PGK directed the flux of the pentose phosphate pathway (PPP) towards the glycolysis pathway (EMP), and the expression of PGDHr improved the L-serine biosynthesis pathway. In addition, the flux from L-serine to glycine dropped by 24%, indicating that the deletion of the activator GlyR resulted in down-regulation of serine hydroxymethyltransferase (SHMT) expression. | 19 | Ionic liquid based dispersive liquid-liquid microextraction of aromatic amines in water samples显示文摘In this work,a new microextraction method termed ionic liquid based dispersive liquid-liquid microextraction(IL-DLLME) was demonstrated for the extraction of 2-methylaniline,4-chloroaniline,1-naphthylamine and 4-aminobiphenyl in aqueous matrices.After extraction the ionic liquid(IL)phase was injected directly into the high performance liquid chromatography(HPLC) system for determination.Some parameters that might affect the extraction efficiency were optimized.Under the optimum conditions,good linear relationship,sensitivity and reproducibility were obtained.The limits of detection(LOD,S/N=3)for the four analytes were in the range of 0.45-2.6μg L^(-1).The relative standard deviations(R.S.D.,n=6)were in the range of 6.2-9.8%.
