| 1 | Late Holocene hydroclimatic variations and possible forcing mechanisms over the eastern Central Asia显示文摘Hydroclimatic variations over the eastern Central Asia are highly sensitive to changes in hemispheric-scale atmospheric circulation systems. To fully understand the long-term variability and relationship between hydroclimate and atmospheric circulation system, we present a high-resolution lascustrine record of late Holocene hydroclimate from Lake Sayram, Central Tianshan Mountains, China, based on the total organic carbon, total nitrogen, and carbonate contents, carbon/nitrogen ratios, and grain size. Our results reveal four periods of substantially increased precipitation at the interval of 4000–3780, 3590–3210, 2800–2160, and 890–280 cal yr BP, and one period of slightly increased precipitation from 1700–1370 cal yr BP. These wetter periods broadly coincide with those identified in other records from the mid-latitude Westerlies-dominated eastern Central Asia, including the northern Tibetan Plateau. As such, a similar hydroclimatic pattern existed over this entire region during the late Holocene. Based on a close similarity of our record with reconstruction of North Atlantic Oscillation indices and solar irradiance, we propose that decreased solar irradiance and southern migration of the entire circum-North Atlantic circulation system, particularly the main pathway of the mid-latitude Westerlies, significantly influenced hydroclimate in eastern Central Asia during the late Holocene. | 2 | Oncolytic adenoviruses expressing checkpoint inhibitors for cancer therapy显示文摘Despite the remarkable success of immune checkpoint inhibitors(ICIs),primary resistance to ICIs causes only subsets of patients to achieve durable responses due to the complex tumor microenvironment(TME).Oncolytic viruses(OVs)can overcome the immunosuppressive TME and promote systemic antitumor immunity in hosts.Engineered OVs armed with ICIs would likely have improved effectiveness as a cancer therapy.According to the diverse immune cell landscapes among different types of tumors,we rationally and precisely generated three recombinant oncolytic adenoviruses(OAds):OAd-SIRPα-Fc,OAd-Siglec10-Fc and OAd-TIGIT-Fc.These viruses were designed to locally deliver SIRPα-Fc,Siglec10-Fc or TIGIT-Fc fusion proteins recognizing CD47,CD24 or CD155,respectively,in the TME to achieve enhanced antitumor effects.Our results suggested that OAd-SIRPα-Fc and OAd-Siglec10-Fc both showed outstanding efficacy in tumor suppression of macrophage-dominated tumors,while OAd-TIGIT-Fc showed the best antitumor immunity in CD8+T-cell-dominated tumors.Importantly,the recombinant OAds activated an inflammatory immune response and generated long-term antitumor memory.In addition,the combination of OAd-Siglec10-Fc with anti-PD-1 significantly enhanced the antitumor effect in a 4T1 tumor model by remodeling the TME.In summary,rationally designed OAds expressing ICIs tailored to the immune cell landscape in the TME can precisely achieve tumor-specific immunotherapy of cancer. | Daoyuan Xie Yaomei Tian Die Hu Yuanda Wang Yuling Yang Bailing Zhou Rui Zhang Zhixiang Ren Mohan Liu Jie Xu Chunyan Dong Binyan Zhao Li Yang | 2023 | Signal Transduction and Targeted Therapy2023,8,12: | 0 |
| 3 | Angiopoietin-1 mRNA expression in estradiol-treated ovariectomized rats with focal cerebral ischemia after reperfusion显示文摘BACKGROUND: Epidemiologic studies have indicated that the incidence of stroke in premenopausal females is lower than in males at the same age, but it significantly rises in postmenopausal females. Estrogen is used clinically to alleviate injury caused by cerebral ischemia. It has been hypothesized that the neuroprotective role of estrogen relates to angiopoietin (Angpt), which plays an important role in vascularization, vascular remodeling and maturation. OBJECTIVE: To observe and validate the effect of estradiol on angiopoietin-1 (Angpt1) mRNA expression in ovariectomized rats with focal cerebral ischemia after reperfusion, so as to explore the molecular mechanisms of estradiol-mediated protection from cerebral ischemic damage. DESIGN, TIME AND SETTING: Randomized, controlled, molecular biology, prospective animal study. The experiment was performed at the Central Laboratory of Chongqing Medical University from September to December 2005. MATERIALS: Fifty healthy female wild type (WT) rats aged 6 months and fifty female rats aged 6 months with knockout of the estrogen-alpha receptor gene (ERKO). METHODS: WT rats and ERKO rats were divided into estradiol and control groups (n = 25), and injected in- tramuscularly with estradiol benzoate (100 μg/kg per day) or corn oil (1 mL/kg per day) for 7 days, 30 days after bilateral ovariectomy. Rat models of cerebral ischemia/reperfusion were established with the middle cerebral artery occlusion method. After 30 minutes of middle cerebral artery occlusion, rats from the estradiol and control groups were injected intramuscularly with estradiol benzoate or corn oil at the above dose. MAIN OUTCOME MEASURES: We used radio-immunity analysis and laser-Doppler flowmetry to measure plasma estradiol levels and changes in cerebral blood flow. We used immunohistochemical staining of CD34 epitopes to measure changes in the capillary density in brain following cerebral ischemia/reperfusion, and quantitative RT-PCR analysis to assess mRNA expression levels of Angpt1, Angpt2, Tie2, vascular endothelial growth factor (Vegf), VegfR1, and VegfR2. RESULTS: In WT ovariectomized rats treated with estradiol, the change in cerebral blood flow following cerebral ischemia/reperfusion, capillary density in the basal nuclei and parietal lobe cortex and Angpt1 mRNA level were significantly higher than in the control group (P < 0.01). We did not identify any such changes in ERKO rats treated with estradiol. In addition, the plasma estradiol levels in WT and ERKO ovariectomized rats treated with estradiol were remarkably higher than in their corresponding control groups (P < 0.01). CONCLUSION: Angpt1 is a critical factor in many processes during the repair of cerebral ischemia/reperfusion injury. For example, it confers estrogen-mediated protection, restoration of cerebral blood flow and increases in brain capillary density. | 4 | Angiopoietin-1 mRNA and Bcl-2 expression following estradiol treatment in ovariectomized rats with focal cerebral ischemia/reperfusion injury显示文摘BACKGROUND:Estrogen has been clinically demonstrated to attenuate ischemic brain injury. However,the precise mechanisms remain controversial. OBJECTIVE:To investigate the effects of estradiol on angiopoietin-1 mRNA and Bcl-2 expression, as well as apoptosis and cerebral blood flow,in ovariectomized rats with focal cerebral ischemia following reperfusion. DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment.The study was performed at the Central Laboratory,Chongqing Medical University from September to December 2005. MATERIALS:Estradiol benzoate was purchased from Shanghai Ninth Pharmaceutical Factory; com oil was purchased from Walmart Supercenter;TUNEL kit,rabbit anti-rat Bcl-2 polyclonal antibody,and biotin-labeled goat anti-rabbit antibody were purchased from Wuhan Boster,China. METHODS:Healthy,female,6-month-old Wistar rats-wild-type and estrogen alpha receptor gene knockout(ERKO)-were randomly divided into estradiol and control groups with 25 animals in each group.The rats were intramuscularly injected with estradiol benzoate(100μg/kg per day) at 30 days following bilateral ovariectomy or corn oil(1 mL/kg per day) for seven consecutive days.Following administration,cerebral ischemia/reperfusion models were established using the right middle cerebral artery occlusion(MCAO) method.After 30 minutes of MCAO,estradiol and control groups were separately injected with estradiol benzoate and corn oil with the above-mentioned doses. MAIN OUTCOME MEASURES:Cell apoptosis was determined by TUNEL;angiopoietin-1 mRNA and Bcl-2 gene expression was determined,respectively,by immunohistochemical staining and RT-PCR.In addition,changes in cerebral blood flow were measured by laser Doppler flowmetry. RESULTS:Changes in angiopoietin-1 mRNA and cerebral blood flow in estradiol-treated,wild-type, MCAO rats following ischemia/reperfusion were greater than in control rats(P<0.01 or 0.05). However,no significant difference was observed between estradiol-treated ERKO MCAO rats and control rats.In addition,estradiol-treated wild-type and ERKO MCAO rats exhibited significantly increased Bcl-2 expression(P<0.05) and decreased number of apoptotic cells in brain tissues compared with control groups(P<0.05). | 6 | Clipping discrete multi-tone for peak-power-constraint IM/DD optical systems显示文摘High peak-to-average power ratio(PAPR)is a major drawback of discrete multi-tone(DMT)in peak-power-constraint(PPC)intensity-modulation/direct-detection(IM/DD)optical systems.Symmetric clipping operation is a frequently-used and practical method for mitigating the PAPR of the DMT signal.However,clipping noise is inevitably induced by the symmetric clipping operation,thereby deteriorating the system’s performance.In this paper,we study the statistical characteristics of the clipping noise in detail.Depending on the statistical characteristics,clipping-noise-cancellation(CNC)algorithms are proposed to mitigate the clipping noise in the clipping DMT systems.Theoretically,the clipping noise can be completely removed using the low-density-parity-check-assisted CNC algorithm(LDPC-assisted CNC algorithm).To verify the superiority of the clipping DMT with the CNC algorithm,we conduct an experiment with a50 Gb/s DMT system using 10G-class commercial devices for the scenario of PPC passive optical networks.The receiver sensitivity of the clipping DMT system with the CNC algorithm can reach-24 d Bm at a 20%soft-decision forward-error-correction limit,which is 2.5 d B higher than that of the DMT system without clipping operation.The clipping operation and the CNC algorithm are added after the original digital signal processing,thereby demonstrating the potential to efectively overcome the drawback in DMT signals and other high-PAPR signals. | Ji ZHOU Liangchuan LI Jiale HE Xiaofeng LU Yu BO Guanyu WANG Yuanda HUANG Gengchen LIU Yanzhao LU Shecheng GAO Yuanhua FENG Shancheng ZHAO Weiping LIU Changyuan YU Zhaohui LI | 2023 | Science China(Information Sciences)2023,66,5: | 0 |
