| 3 | Toll-Like Receptor 4 Deficiency Causes Reduced Exploratory Behavior in Mice Under Approach-Avoidance Conflict显示文摘Abnormal approach-avoidance behavior has been linked to deficits in the mesolimbic dopamine(DA)system of the brain. Recently, increasing evidence has indicated that toll-like receptor 4(TLR4), an important pattern-recognition receptor in the innate immune system,can be directly activated by substances of abuse, resulting in an increase of the extracellular DA level in the nucleus accumbens. We thus hypothesized that TLR4-dependent signaling might regulate approach-avoidance behavior. To test this hypothesis, we compared the novelty-seeking and social interaction behaviors of TLR4-deficient(TLR4^(-/-))and wild-type(WT) mice in an approach-avoidance conflict situation in which the positive motivation to explore a novel object or interact with an unfamiliar mouse was counteracted by the negative motivation to hide in exposed,large spaces. We found that TLR4^(-/-)mice exhibitedreduced novelty-seeking and social interaction in the large open spaces. In less stressful test apparatuses similar in size to the mouse cage, however, TLR4^(-/-)mice performed normally in both novelty-seeking and social interaction tests. The reduced exploratory behaviors under approachavoidance conflict were not due to a high anxiety level or an enhanced fear response in the TLR4^(-/-)mice, as these mice showed normal anxiety and fear responses in the open field and passive avoidance tests, respectively. Importantly,the novelty-seeking behavior in the large open field induced a higher level of c-Fos activation in the nucleus accumbens shell(NAc Sh) in TLR4^(-/-)mice than in WT mice. Partially inactivating the NAc Sh via infusion of GABA receptor agonists restored the novelty-seeking behavior of TLR4^(-/-)mice. These data suggested that TLR4 is crucial for positive motivational behavior under approach-avoidance conflict. | 18 | Numerical study of vortex breaker optimization in a first stage oxygen tank显示文摘One of the crucial factors affecting the carrying capacity of the cryogenic liquid launch vehicle is the effective volume of the tank.Theoretical and experimental investigations on vortex breaker mechanisms have proposed promising schemes applied in the oxygen tank of the liquid-propellant launch vehicle to ensure the normal operation of the engine.In this paper,the liquid surface profile functions of the laminar core when the vortex generates were derived based on the Rankine vortex model.The dimensionless residual volume V/d3 and the Froude number were applied to compare the theoretical prediction of critical height with the actual simulation data of liquid oxygen.This comparison method can improve the model’s accuracy.The efficiency of different basic shapes of vortex breakers was tested by conducting CFD modelling on a non-vertical outflow tank under a specific operating condition.Simulation results suggest negligible effects of heat transfer and surface tension.A circular plate is considered the optimal vortex breaker shape in traditional vertical outflow tanks,while a higher optimize efficiency was discovered in the half baffle basic shape in a non-vertical outflow tank by comparing the dimensionless residual volume and flow coefficient.A 34.26%reduction in flow resistance of half baffle breaker can be reached when applying a twenty-degree outlet pipe chamfering setting compared to a zero-degree chamfer.Considering practical operating limitations,it is concluded that a vortex breaker mechanism in a half baffle basic shape with a radius of 2.5d and a height of 4/d is the optimal scheme,which is suitable for all types of tanks.Its optimization efficiency of the residual volume reduction is about 56.68%compared to a nobreaker installation case.Lastly,a general equation based on CFD simulation for predicting the residual volume under a certain outflow velocity was proposed:V=d3yaFr0:3,which trend is consistent with that of mathematical prediction V=d3yaFr1=3.This consistency proves the accuracy and applicability of optimization strategy in this paper. | Yixiu Shen Yimeng Li Zhenggang Du | 2023 | Propulsion and Power Research2023,12,3: | 0 |

| 20 | Multitranscriptome analyses of keloid fibroblasts reveal the role of the HIF-1α/HOXC6/ERK axis in keloid development显示文摘Background:A keloid is a disease of excessive fibrosis that is characterized by the aberrant prolifer-ation of fibroblasts.However,the molecular mechanisms of fibroblasts during the development of keloids remain unclear.This study aims to identify new molecular targets that promote the proliferation and migration of keloid fibroblasts,providing new ideas for the prevention and treatment of keloids.Methods:We utilized bioinformatics tools to analyze data from keloid fibroblasts(KFs)available in the Gene Expression Omnibus(GEO)database to identify the key genes involved in keloid development.Homeobox C6(HOXC6)emerged as a hub gene in KFs from the GEO database was verified in keloid tissue samples and KFs using reverse transcription-quantitative polymerase chain reaction,western blot(WB)and immunohistochemistry.Subsequently,the effects of down-regulated HOXC6 expression on the cellular behaviors of KFs were examined by performing Cell Counting Kit-8,flow cytometry,transwell migration and WB assays.Meanwhile,we performed transcriptome sequencing and gene set enrichment analysis(GSEA)to further explore HOXC6-related mechanisms and validated the signaling pathways by performing a series of experiments.Results:HOXC6 was the top-ranking hub gene of KFs in microarray datasets from GEO and was upregulated in keloid tissue samples and KFs.Downregulation of HOXC6 inhibited proliferation,migration and extracellular matrix(ECM)accumulation and promoted KF apoptosis.GSEA pre-dicted that the hypoxia signaling pathway was associated with HOXC6 in KFs.Transcriptome sequencing suggested that the extracellular regulated protein kinase(ERK)pathway was one of the downstream pathways of HOXC6 in KFs.Our experiments confirmed that hypoxia-inducible factor-1α(HIF-1α)upregulates HOXC6,contributing to KFs proliferation,migration,apoptosis inhibition and collagen accumulation through the ERK signaling pathway.Conclusions:Our findings first revealed that HOXC6 acts as an oncogenic driver in the molec-ular mechanisms of fibroblasts in keloids.The HIF-1α/HOXC6/ERK axis promotes proliferation,migration and ECM production by KFs,contributing to the progression of keloids.Taken together,HOXC6 may serve as a promising novel therapeutic target and new focus for research designed to understand the pathogenesis of keloids. | Qi Wang Yixiu Zhong Zhijia Li Dingheng Zhu Hongyan Lu Pingjiao Chen Changxing Li Xuebiao Peng Qian Li Kang Zeng | 2022 | Burns & Trauma2022,10,1: | 0 |
