Therefore, apoptosis is a stringent and effective cell quality controlling system, which reduces the number of harmful cells to the maximum, such as auto-immunity cell, cells infected by virus, tumor cells by cell suicide111. | 2 | SRSF1 plays a critical role in invariant natural killer T cell development and function显示文摘Invariant natural killer T(iNKT)cells are highly conserved innate-like T lymphocytes that originate from CD4^(+)CD8^(+)double-positive(DP)thymocytes.Here,we report that serine/arginine splicing factor 1(SRSF1)intrinsically regulates iNKT cell development by directly targeting Myb and balancing the abundance of short and long isoforms.Conditional ablation of SRSF1 in DP cells led to a substantially diminished iNKT cell pool due to defects in proliferation,survival,and TCRαrearrangement.The transition from stage 0 to stage 1 of iNKT cells was substantially blocked,and the iNKT2 subset was notably diminished in SRSF1-deficient mice.SRSF1 deficiency resulted in aberrant expression of a series of regulators that are tightly correlated with iNKT cell development and iNKT2 differentiation,including Myb,PLZF,Gata3,ICOS,and CD5.In particular,we found that SRSF1 directly binds and regulates pre-mRNA alternative splicing of Myb and that the expression of the short isoform of Myb is substantially reduced in SRSF1-deficient DP and iNKT cells.Strikingly,ectopic expression of the Myb short isoform partially rectified the defects caused by ablation of SRSF1.Furthermore,we confirmed that the SRSF1-deficient mice exhibited resistance to acute liver injury uponα-GalCer and Con A induction.Our findings thus uncovered a previously unknown role of SRSF1 as an essential post-transcriptional regulator in iNKT cell development and functional differentiation,providing new clinical insights into iNKT-correlated disease. | Jingjing Liu Menghao You Yingpeng Yao Ce Ji Zhao Wang Fang Wang Di Wang Zhihong Qi Guotao Yu Zhen Sun Wenhui Guo Juanjuan Liu Shumin Li Yipeng Jin Tianyan Zhao Hai-Hui Xue Yuanchao Xue Shuyang Yu | 2021 | Cellular & Molecular Immunology2021,18,11: | 3 |

| 6 | Evaluation of menopausal status among breast cancer patients with chemotherapy-induced amenorrhea显示文摘Objective: In patients with chemotherapy-induced amenorrhea(CIA), the menopausal status is ambiguous and difficult to evaluate. This study aimed to establish a discriminative model to predict and classify the menopausal status of breast cancer patients with CIA.Methods: This is a single center hospital-based study from 2013 to 2016. The menopausal age distribution and accumulated incidence rate of CIA are described. Multivariate models were adjusted for established and potential confounding factors including age, serum concentration of estradiol(E2) and follicle-stimulating hormone(FSH),feeding, pregnancy, parity, abortions, and body mass index(BMI). The odds ratio(OR) and 95% confidence interval(95% CI) of different risk factors were estimated.Results: A total of 1,796 breast cancer patients were included in this study, among whom, 1,175(65.42%) were premenopausal patients and 621(34.58%) were post-menopause patients. Five hundred and fifty patients were included in CIA analysis, and a cumulative CIA rate of 81.64% was found in them. Age(OR: 1.856, 95% CI:1.732-1.990), serum concentration of E2(OR: 0.976, 95% CI: 0.972-0.980) and FSH(OR: 1.060, 95% CI:1.053-1.066), and menarche age(OR: 1.074, 95% CI: 1.009-1.144) were found to be associated with the patients' menopausal status. According to multivariate analysis, the discriminative model to predict the menopausal status is Logit(P)=-28.396+0.536 Age-0.014 E2+0.031 FSH. The sensitivities for this model were higher than 85%, and its specificities were higher than 89%.Conclusions: The discriminative model obtained from this study for predicting menstrual state is important for premenopausal patients with CIA. | 14 | Analysis of proteins that interact with nucleocapsid protein of SARS-CoV using 15-mer phage-displayed library显示文摘Analysis of proteins that interact with N protein of SARS-CoV using 15-mer phage-displayed library will help to explore the virus pathogenesis and to develop new drugs and vaccines against SARS. In this study,we cloned,expressed and purified N protein of SARS-CoV. This 46-kD N protein was verified by SDS-PAGE and Western-blot. Then,the peptides binding-specific to N protein were identified using 15-mer phage-displayed library. Surprisingly,all of the 89 clones from monoclonal ELISA were positive (S/N>2.1) and the result was further confirmed experimentally once again. Six N protein-binding pep-tides,designated separately as SNA1,SNA2,SNA4,SNA5,SNA9 and SNG11,were selected for se-quencing. Sequence analysis suggested that SNA5 shared approximatively 100% sequence identity to SNA4,SNA2,SNA9 and SNA1. In addition,the binding specificity of the 15-mer peptides with the SARS-CoV N protein was further demonstrated by blocking ELISA using the synthetical 15-mer peptide according to the deduced amino acid sequence of SNA5. Also,the deduced amino sequence of SNA5 was compared with proteins in translated database using the tblastx program,and the results showed that the proteins with the highest homology were Ubiquinol-cytochrome c reductase iron-sulfur sub-units (UCRI or UQCR),otherwise known as the Rieske iron-sulfur proteins (RISP). Notablely,in the 2Fe-2S redox centre of UCRI,there were 6 residues GGW(Y)F(Y)CP compatible to the residues (po-sition 2→7,GGWFCP7) of the NH2-terminal of the 15-mer peptide,which indicated higher binding specificity between the N protein of SARS-CoV and the redox centre of UCRI to some extent. | 15 | Cloning and identification of measles virus receptor gene from marmoset cells显示文摘The measles virus (MV) strains with mutated hemagglutinin gene (ha) lost the capacity to infect its sensitive host cells (Vero cells), but it may infect the marmoset B-lymphoblastoid cell line B95a. From above, we can presume that there is a novel cellular receptor for those measles virus strains on B95a cells. Using the yeast two-hybrid system, we screened and cloned a novel gene-bip (B-lymphoblastoid interaction protein of marmoset) from B95a cell cDNA library, which encoded a protein interacting with measles virus hemagglutinin protein (Ha). The bip cDNA was 1540 base pairs in length and contained a unique open reading frame (ORF) of 1011 base pairs encoding a transmembrane protein of 337 amino acid residues. The primary structure of amino acids residue is predicted that the Bip comprised a hydrophobic transmembrane domain and a hydrophobic leader region. Ectopically expressed P49 protein had the capacity to inhibit the activity of Caspases, being the executioner of apoptosis. Digestion of P49 with human Caspase or Bm-Caspase both yielded 10 and 40 kD fragments. Checking the sequence of P49, we found that the motif 91-TVTDG-95 of P49 was the sequence recog-nized by Caspases. The mutant of P49, Asp94Ala, could not be cut by both caspases and lost its caspases inhibition. Meanwhile, Thr91Ala mutant permitted the cleavage and partially retained its activity of caspases inhibition.
