1A four-compartment model for the COVID-19 infection—implications on infection kinetics,control measures,and lockdown exit strategies显示文摘Objective:To analyse the impact and repercussions of the surge in healthcare demand in response to the COVID-19 pandemic,assess the potential effectiveness of various infection/disease control measures,and make projections on the best approach to exit from the current lockdown.Design:A four-compartment model was constructed for SARS-CoV-2 infection based on the Wuhan data and validated with data collected in Italy,the UK,and the US.The model captures the effectiveness of various disease suppression measures in three modifiable factors:(a)the per capita contact rate(β)that can be lowered by means of social distancing,(b)infection probability upon contacting infectious individuals that can be lowered by wearing facemasks,personal hygiene,etc.,and(c)the population of infectious individuals in contact with the susceptible population,which can be lowered by quarantine.The model was used to make projections on the best approach to exit from the current lockdown.Results:The model was applied to evaluate the epidemiological data and hospital burden in Italy,the UK,and the US.The control measures were identified as the key drivers for the observed epidemiological data through sensitivity analyses.Analysing the different lockdown exit strategies showed that a lockdown exit strategy with a combination of social separation/general facemask use may work,but this needs to be supported by intense monitoring whichwould allowre-introduction/tightening of the controlmeasures if the number of newinfected subjects increases again.Conclusions and relevance:Governments should act early in a swift and decisive manner for containment policies.Any lockdown exit will need to be monitored closely,with regards to the potential of lockdown reimplementation.This mathematical model provides a framework for major pandemics in the future.Tianbing Wang Yanqiu Wu Johnson Yiu-Nam Lau Yingqi Yu Liyu Liu Jing Li Kang Zhang Weiwei Tong Baoguo Jiang 2020Precision Clinical Medicine2020,3,2:5

3A general bimetal-ion adsorption strategy to prepare nickel single atom catalysts anchored on graphene for efficient oxygen evolution reaction显示文摘Single-atom catalysts (SACs) supported on two-dimensional (2D) materials are highly attractive for maximizing their catalytic activity.However,graphene based SACs are primarily bonded with nitrogen and carbon sites,resulting in poor performance for the oxygen evolution reaction (OER).Herein,we develop a general bimetal-ion adsorption strategy for the synthesis of individually dispersed Ni SACs anchored on the oxygenated sites of ultrathin reduced graphene oxide as efficient OER electrocatalysts.The resultant Ni SACs for OER in alkaline electrolyte exhibit a highly stable overpotential of 328 mV at the current density of 10 mA cm^-2,and Tafel slope of 84 mV dec^-1 together with long-term durability and negligible degradation for 50 h,which is greatly outperform its counterparts of nitrogen bonded Ni SACs (564 mV,364 mV dec^-1) and Ni(OH)2 nanoparticles anchored on graphene (450 mV,142 mV dec^-1),and most reported Ni based OER electrocatalysts.Furthermore,the extended X-ray absorption fine structure at the Ni K-edge and theoretical simulation reveal that the nickel-oxygen coordination significantly boost OER performance.Therefore,this work will open numerous opportunities for creating novel-type 2D SACs via oxygen-metal bonding as highly robust OER catalysts.Yingqi Xu Weifeng Zhang Yaguang Li Pengfei Lu Zhong-Shuai Wu 2020Journal of Energy Chemistry2020,29,4:3

5Monascus vinegar alleviates high-fat-diet-induced inflammation in rats by regulating the NF-κB and PI3K/AKT/mTOR pathways显示文摘Monascus vinegar(MV) is a typical fermented food with various health-promoting effects. This study aimed to evaluate the role of MV in alleviating high-fat-diet-induced inflammation in rats with hyperlipidemia and elucidate the possible regulatory mechanisms. In the study, serum lipid profiles, liver pathology and liver inflammatory cytokines were analyzed in hyperlipidemia rats with MV(0.5 mL/kg mb, 2 mL/kg mb). Results showed that the administration of MV alleviated dyslipidemia by decreasing the serum and liver levels of triglyceride and total cholesterol. Increase in hepatic lipase and carnitine palmitoyl transferase 1(CPT-1)levels and decrease in hepatocyte steatosis, nephritis, and intestinal tissue injury in the HD group showed that high-dose MV can significantly suppress hepatic lipid accumulation and steatosis. In addition, compared with the model(MOD) group, the HD group showed significantly down-regulated the level of serum or hepatic alanine aminotransferase(ALT), aspartate aminotransferase(AST), CPT-1, interleukin(IL)-2, IL-6, IL-12,and tumor necrosis factor α(TNF-α). Moreover, the HD group showed repressed hepatic nuclear factor κB(NF-κB) pathway and inactivated phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR) pathway mitigated liver inflammation. Similar results were obtained from cell experiments. 6Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy.Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang 2023Signal Transduction and Targeted Therapy2023,8,4:2
7Efficient and high-fidelity base editor with expanded PAM compatibility for cytidine dinucleotide显示文摘Cytidine base editor(CBE),which is composed of a cytidine deaminase fused to Cas9 nickase,has been widely used to induce C-to-T conversions in a wide range of organisms.However,the targeting scope of current CBEs is largely restricted to protospacer adjacent motif(PAM)sequences containing G,T,or A bases.In this study,we developed a new base editor termed“nNme2-CBE”with excellent PAM compatibility for cytidine dinucleotide,significantly expanding the genome-targeting scope of CBEs.Using nNme2-CBE,targeted editing efficiencies of 29.0%-55.0%and 17.3%—52.5%were generated in human cells and rabbit embryos,respectively.In contrast to conventional nSp-CBE,the nNme2-CBE is a natural high-fidelity base editing platform with minimal DNA off-targeting detected in vivo.Significantly increased efficiency in GC context and precision were determined by combining nNme2Cas9 with rationally engineered cytidine deaminases.In addition,the Founder rabbits with accurate single-base substitutions at Fgf5 gene loci were successfully generated by using the nNme2-CBE system.These novel nNme2-CBEs with expanded PAM compatibility and high fidelity will expand the base editing toolset for efficient gene modification and therapeutic applications.Zhiquan Liu Siyu Chen Yingqi Jia Huanhuan Shan Mao Chen Yuning Song Liangxue Lai Zhanjun Li 2021Science China(Life Sciences)2021,64,8:2

10Spatio-Temporal Cellular Network Traffic Prediction Using Multi-Task Deep Learning for AI-Enabled 6G显示文摘Spatio-temporal cellular network traffic prediction at wide-area level plays an important role in resource reconfiguration,traffic scheduling and intrusion detection,thus potentially supporting connected intelligence of the sixth generation of mobile communications technology(6G).However,the existing studies just focus on the spatio-temporal modeling of traffic data of single network service,such as short message,call,or Internet.It is not conducive to accurate prediction of traffic data,characterised by diverse network service,spatio-temporality and supersize volume.To address this issue,a novel multi-task deep learning framework is developed for citywide cellular network traffic prediction.Functionally,this framework mainly consists of a dual modular feature sharing layer and a multi-task learning layer(DMFS-MT).The former aims at mining long-term spatio-temporal dependencies and local spatio-temporal fluctuation trends in data,respectively,via a new combination of convolutional gated recurrent unit(ConvGRU)and 3-dimensional convolutional neural network(3D-CNN).For the latter,each task is performed for predicting service-specific traffic data based on a fully connected network.On the real-world Telecom Italia dataset,simulation results demonstrate the effectiveness of our proposal through prediction performance measure,spatial pattern comparison and statistical distribution verification.Xiaochuan Sun Biao Wei Jiahui Gao Difei Cao Zhigang Li Yingqi Li 2022Journal of Beijing Institute of Technology2022,31,5:1
11Effect of haemodynamics on the risk of ischaemic stroke in patients with severe vertebral artery stenosis显示文摘Objectives Endovascular treatment strategies to optimise individualised care for patients with vertebral artery(VA)stenosis need to be revisited.This study aimed to investigate the relationship between net VA flow volume(NVAFV)and the risk of posterior circulation infarction(PCI)in a high-risk patient population.Methods We screened 1239 patients with extracranial VA stenosis,of whom 321 patients with severe VA V1 segment stenosis(≥70%)were enrolled in our study.We restratified the patients based on NVAFV and contralateral VA stenosis grades to analyse the proportion of each PCI mechanism-large artery atherosclerosis and branch artery occlusive disease.Furthermore,we estimated the incidence of recurrent ischaemic stroke between groups with different NVAFV over a follow-up period of 2 years.Results NVAFV was lower in the PCI group.Multiple logistic regression analysis showed that NVAFV is an independent risk factor for PCI and that the OR for PCI for the lowest NVAFV(<112.8 mL/min)was 4.19(1.76 to 9.95,p=0.001).In patients with severe carotid artery disease,the OR for the lowest NVAFV was 14.03(3.18 to 61.92,p<0.001).The lower NVAFV group had a higher incidence of recurrent ischaemic stroke events than the higher NVAFV group(HR 2.978,95%CIs 1.414 to 6.272).Conclusion Our study demonstrated that NVAFV,as estimated by colour duplex ultrasonography,was associated with the incidence of PCI and subsequent ischaemic events and that a high-risk population could be identified for further posterior circulation revascularisation.Qing Li Yinghua Zhou Yingqi Xing Jie Yang Yang Hua 2022Stroke & Vascular Neurology2022,7,3:1

The strict specifications of caprolactam products make the refining process of caprolactam extremely important. According to the different rearrangement process, the refining process of caprolactam is also different. The main refining sequence of liquid phase rearrangement products includes extraction, ion exchange, hydrogenation, and distillation. The refining process of gas phase rearrangement products is conducted mainly through distillation, crystallization, and hydrogenation.
