1Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis.Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang 2020Acta Biochimica et Biophysica Sinica2020,52,8:16
2Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 3Efficacy of Rebixiao Chinese herbal tablets and Chinese formula granules in acute gout arthritis patients:a randomized,multicenter,double-blind,controlled trial显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,controlled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three centers.The total effective rates of the joint symptom score,Traditional Chinese Medicine(TCM)symptoms score,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical efficacy.Safety assessments were also performed.RESULTS:Of the 165 enrolled patients,147 completed the clinical observation.There was no difference in baseline between the two groups.The total effective rates of the joint symptom score were 94.36%and 97.36%,and the total effective rates of the TCM symptoms score were 95.77%and 97.36%in the CFG group and CHT group,respectively.No statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were consistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).CONCLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects.Wang Shasha Zhang Ying Dai Min Yang Min Leng Wenfei Peng Xiufeng Li Guojun Yang Jinbing Gan Jianping Zhu Fenglin Li Yanping Wu Bin 2020Journal of Traditional Chinese Medicine2020,40,4:3

6N_(x)O emissions in response to the irrigation lower limits under different irrigation modes in a lettuce field显示文摘Irrigation has a significant impact on N_(x)O(N_(2)O and NO)emissions from cultivated land,yet the N_(2)O or NO emission among the irrigation lower limits under different irrigation modes has not been well compared.In an irrigated lettuce field,three DR(drip irrigation)lower limits were designed,including 75%(DR1),65%(DR2)and 55%(DR3)field capacity,and one FI(furrow irrigation)lower limit(65%field capacity).The N_(2)O and NO emission fluxes and soil nitrogen(N)forms were determined,and the linear correlation between these indicators was analyzed.Results showed that under the same irrigation regime,the N_(2)O and NO emissions from furrow irrigation treatment increased by 36.8%and 45.2%respectively compared to that from drip irrigation treatment.The cumulative N_(2)O and NO emissions under DR3 were 30.2%and 28.6%higher than under DR1,respectively.Moreover,DR1 was also the lowest among the four treatments in soil-N concentration.The N_(2)O and NO emission fluxes were more correlated to soil NH_(4)^(+)-N(r=0.88 and 0.76)or NO_(2)^(-)-N(r=0.90 and 0.80)concentration than soil-N and soluble organic N,indicating that N_(2)O and NO were mainly produced by the soil nitrification process.When the irrigation regime was the same,N_(2)O and NO emissions were lower with drip irrigation than with furrow irrigation.Besides,drip irrigation with small quota but high frequency reduced N_(2)O and NO emission compared to that with large quota but low frequency.Maomao Hou Ying Xiao Qinyuan Zhu Jingnan Chen Huan Huang Qiu Jin Lin Zhu Fenglin Zhong 2023International Journal of Agricultural and Biological Engineering2023,16,4:1
7Geographical Information Visualization on a Panoramic Sphere in an Immersive Environment显示文摘In this paper,we propose a novel approach to visualizing global geographical information:a panoramic sphere in an immersive environment.The whole geographical surface can be observed through the rotating of heads as the viewpoint of the panoramic sphere is inside the sphere.We compared three approaches to visualizing the earth for rendering the geographical information in a virtual reality environment.On the tasks of terrestrial and marine geographical information,we compare the visualization effects on a)a globe,b)a flat map and c)a panoramic sphere.Terrestrial geographical information tasks include the area comparison and direction determination.Marine geographical information tasks contain the visualization of sea surface temperature and sea surface currents.For terrestrial geographical information tasks,the experimental results show that the panoramic sphere outperforms the globe and the flat map,with a higher average accuracy and a shorter time.On marine geographical information task,the panoramic sphere visualization is also superior to the flat map and the globe in an immersive environment for the sea surface temperature data and the sea surface current fields.In all three visualization experiments,the panoramic sphere is most preferred by the participants,particularly for global,transcontinental and transoceanic needs.MA Ying TIAN Fenglin LIU Wen TANG Moran LI Jinyu CHEN Ge 2023Journal of Ocean University of China2023,22,4:0

10Inflammation as a Mediator of Microbiome Dysbiosis-Associated DNA Methylation Changes in Gastric Premalignant Lesions显示文摘Evidence for the influence of chronic inflammation induced by microbial dysbiosis on aberrant DNA methylation supports a plausible connexion between disordered microbiota and precancerous lesions of gastric cancer(PLGC).Here,a comprehensive study including multi-omics data was performed to estimate the relationships amongst the gastric microbiome,inflammatory proteins and DNA methylation alterations and their roles in PLGC development.The results demonstrated that gastric dysbacteriosis increased the risk of PLGC and DNA methylation alterations in related tumour suppressor genes.Seven inflammatory biomarkers were identified for antrum and corpus tissues,respectively,amongst which the expression levels of several biomarkers were significantly correlated with the microbial dysbiosis index(MDI)and methylation status of specific tumour suppressor genes.Notably,mediation analysis revealed that microbial dysbiosis partially contributed to DNA methylation changes in the stomach via the inflammatory cytokines C-C motif chemokine 20(CCL20)and tumour necrosis factor receptor superfamily member 9(TNFRSF9).Overall,these results may provide new insights into the mechanisms that might link the gastric microbiome to PLGC.Lingjun Yan Wanxin Li Fenglin Chen Junzhuo Wang Jianshun Chen Ying Chen Weimin Ye 2023Phenomics2023,3,5:0
