| 1 | Construction of a small-caliber tissue-engineered blood vessel using icariin-loaded β-cyclodextrin sulfate for in situ anticoagulation and endothelialization显示文摘The rapid endothelialization of tissue-engineered blood vessels(TEBVs) can effectively prevent thrombosis and inhibit intimal hyperplasia. The traditional Chinese medicine ingredient icariin is highly promising for the treatment of cardiovascular diseases.β-cyclodextrin sulfate is a type of hollow molecule that has good biocompatibility and anticoagulation properties and exhibits a sustained release of icariin. We studied whether icariin-loaded β-cyclodextrin sulfate can promote the endothelialization of TEBVs. The experimental results showed that icariin could significantly promote the proliferation and migration of endothelial progenitor cells; at the same time, icariin could promote the migration of rat vascular endothelial cells(RAVECs). Subsequently,we used an electrostatic force to modify the surface of the TEBVs with icariin-loaded β-cyclodextrin sulfate, and these vessels were implanted into the rat common carotid artery. After 3 months, micro-CT results showed that the TEBVs modified using icariin-loaded β-cyclodextrin sulfate had a greater patency rate. Scanning electron microscopy(SEM) and CD31 immunofluorescence results showed a better degree of endothelialization. | 7 | Programming of Regulatory T Cells In Situ for Nerve Regeneration and Long-Term Patency of Vascular Grafts显示文摘Rapid integration into the host tissue is critical for long-term patency after small diameter tissue engineering vascular grafts(sdTEVGs)transplantation.Neural recognition may be required for host integration and functionalization of the graft.However,immune rejection and inflammation hinder nerve regeneration of sdTEVGs.Here,a CRISPR/dCas9-nanocarrier was used for targeted programming of regulatory T cells(Treg cells)in situ to promote nerve regeneration of sdTEVGs by preventing excessive inflammation.Treg cells and(C-C chemokine receptor)CCR2+macrophage recruitment occurred after transplantation.The nanodelivery system upregulated ten eleven translocation(TET2)in Treg cells in vitro.Reprogrammed Treg cells upregulated anti-inflammatory cytokines and decreased the proportion of CCR2+macrophages.IL-6 concentrations decreased to the levels required for nerve regeneration.Implantation of CRISPR/dCas9 nanodelivery system-modified sdTEVGs in rats resulted in Treg cell editing,control of excessive inflammation,and promoted nerve regeneration.After 3 months,nerve regeneration was similar to that observed in normal blood vessels;good immune homeostasis,consistency of hemodynamics,and matrix regeneration were observed.Neural recognition promotes further integration of the graft into the host,with unobstructed blood vessels without intimal hyperplasia.Our findings provide new insights into vascular implant functionalization by the host. | Yanhong Wang Fangchao Xue Yanzhao Li Lin Lin Yeqin Wang Shanlan Zhao Xingli Zhao Yong Liu Ju Tan Gang Li Haoran Xiao Juan Yan Hao Tian Min Liu Qiao Zhang Zhaojing Ba Lang He Wenyan Zhao Chuhong Zhu Wen Zeng | 2022 | Research2022,,4: | 1 |

| 11 | 局部晚期鼻咽癌同期放化疗顺铂单药每周方案剂量递增临床研究(英文)显示文摘Objective:The purpose of this study was to define the maximum tolerated dose(MTD) by describing the doselimiting toxicity(DLT) of weekly cisplatin concurrently with conventional plus 3-dimensional conformal radiotherapy(CT + 3DCRT) in patients with loco-regionally advanced nasopharyngeal carcinoma(NPC).Methods:Patients with loco-regionally advanced NPC(III/IVa stage) were enrolled into a dose-escalating study.Toxicity was graded according to Common Terminology Criteria for Adverse Events version 3.0(CTCAE v3.0).MTD was defined when 2 of 6 patients developed DLT.The starting dose of cisplatin was 15 mg/m2/w,with a subsequent dose escalation of 5 mg/m2/w in cohorts of 3 new patients.CT + 3DCRT was given to the nasopharynx and the upper neck;the lower neck was treated by a single anterior field irradiation.The prescription dose was 70-80 Gy by 35-40 fractions to the nasopharynx gross tumor,and 66-70 Gy by 33-35 fractions to the positive neck lymph nodes.Results:From Jun.2008 to Sep.2009,24 patients received complete chemoradiotherapy,and all of them were eligible for toxicity evaluation.On the first five dose levels of 15 mg/m2/w and 35 mg/m2/w,no patient experienced DLT.On the next dose level of 40 mg/m2/w,1 patient experienced DLT of grade 3 myelosuppression for 1.4 weeks,and among the additional 3 patients,no one developed DLT.On the seventh dose level of 45 mg/m2/w,all the patients developed grade 3 myelosuppression for more than 1 weeks,and the dose-escalating trial stopped.The 23(95.8%) patients achieved clinical complete remission(CR) in the local site;22(91.7%) achieved CR in the regional site,and all patients got CR 3 months later.After a median follow-up of 16.4 months,1 patient developed liver metastases 2 months later,1 patient developed bone metastases 10 months later and 22 kept disease-free survival.Conclusion:The MTD of cisplatin weekly with concurrent CT + 3DCRT for local advanced NPC is 40 mg/m2/w,with myelosuppression as DLT. | Daiyuan Ma Chunqiao Fu Bangxian Tan Mi Liu Xianfu Li Yeqin Zhou | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,4: | 1 |
| 12 | 三维适形放疗配合腔内后装治疗宫颈癌的临床研究(英文)显示文摘Objective: The aim of our study was to evaluate the outcome and complications of cervical cancer patients undergoing conventional intracavitary brachytherapy (ICBT) treated with 3D-conformal radiotherapy (3DCRT). Methods: Sixty cervical cancer patients were divided randomly into the conformal group and the conventional group. Thirty patients treated with 3D-conformal radiotherapy in the 3DCRT group, when the whole pelvic received DT 40 Gy, a planning CT scan of each patient was obtained and the second 3DCRT therapy plan was taken. Then, continued to irradiate to 50 Gy. At last, 3DCRT was boosted at local involved volumes to the total dose of 60 Gy. When 3DCRT was combined with intracavitary brachytherapy, the dose of brachytherapy to point A was 30 Gy/5 fractions. In the conventional group, after a total tumor dose of 40 Gy was delivered by the whole pelvic irradiation, the four-field technique was used to irradiate the total pelvic and regional nodes (median dose of 10 Gy), and the involved volumes were boosted to 60 Gy and the dose of brachytherapy to point A was 30 Gy-36 Gy/5-6 fractions. Moreover, both groups were combined with intracavitary brachytherapy respectively. Results: The 1, 2, 3-year survival rates for the 3DCRT group and the conventional group were 96.7%, 93.3%, 90.0% and 86.6%, 76.7%, 70% respectively (P = 0.04, P = 0.02 and P = 0.02). There was a statistically significant difference between the two groups. Compared to the two groups each other in toxic effects, except for the I-II grade rectal and bladder reaction and pelvic fibrosis which was lower in the 3DCRT group (P = 0. 007, P = 0. 006 and P = 0. 015), the side effects were similar and well tolerated in two groups. Conclusion: The all-course 3DCRT combined with intracavitary brachytherapy can be considered as an effective and feasible approach to cervical cancer and may significantly improve the survival rate and reduce the late toxicity. Methods:Sixty-eight cases of recurrent rectal cancer were divided randomly into two groups:34 cases of conformal radiotherapy plus FOLFOX chemotherapy group (experiment group) and 34 cases of conformal radiotherapy (control group). Researchers took chemotherapy in the first week and the fourth week after radiotherapy, with 5-fluorouracil 500 mg/m 2 , calcium leucovorin 200 mg, d1-5 with intravenous drip, Oxaliplatin 130 mg/m 2 and d1 with intravenous drip 2 h, 21 days was one cycle. Kaplan-Meier method was used for survival analysis. Results:The survival rates for 1, 2 and 3 years for experiment group and control group were 88.2%, 64.7%, 47.1% and 66.7%, 38.2%, 29.4% (P = 0.03), the 2-year rate of distant metastases was 32.4% and 58.8% (P = 0.032) respectively. The median survival time was 33 and 20 months respectively. There were some side effects between the groups, but there was no statistical difference. | 18 | Three-dimensional conformal radiotherapy plus concurrent DICE chemotherapy for early-stage nasal-type natural killer/T-cell lymphoma of Waldeyer's ring: A single-institution study显示文摘Objective Nasal-type natural killer/T-cell lymphoma of Waldeyer's ring(WR-NK/TL) has different clinicopathological characteristics from those of other subtypes of NK/T lymphoma; thus, the optimal treatment remains unclear. To find a more effective treatment model for WR-NK/TL, we conducted a single-center study of concurrent radiochemotherapy.Methods Forty-five patients with newly diagnosed stage IE to IIE WR-NKTL were randomly divided into two groups. The 23 cases in the concurrent radiochemotherapy group were treated with three-dimensional conformal radiotherapy(48–52 Gy) and 2 courses of DICE(dexamethasone, ifosfamide, cisplatin, and etoposide) synchronous chemotherapy. The 22 cases in the radiotherapy group only received three-dimensional conformal radiotherapy(50–54 Gy). The primary end points were overall survival(OS), progressionfree survival(PFS), and toxicity.Results The 1-, 3-, and 4-year OS and PFS rates were 95.5%, 65.6%, and 45.9%, and 86.4%, 56.0%, and 46.7% in the radiotherapy group, and 100%, 88.5%, and 88.5%, and 100%, 82.0%, and 73.8% in the concurrent radiochemotherapy group, respectively. The OS(P = 0.0477) and PFS rates(P = 0.0488) were higher in the concurrent radiochemotherapy group than in the radiotherapy group. The overall response rate was 100% in both the radiotherapy group [complete response(CR), 18 cases] and concurrent radiochemotherapy group(CR, 22 cases). The concurrent radiochemotherapy group had more severe side effects, especially grade 3 + 4 events, such as leukopenia, anorexia, and stomatitis. It has been recently recognized that the hydride materials containing negatively charged hydrogen(H~-) show promises for Nfixation and hydrogenation to ammonia. Herein, we report that rare earth metal hydrides such as lanthanum hydride can also fix Neither by heating to 200 °C or ball milling under ambient Npressure and temperature. The Nfixation by lanthanum hydride may proceed via an intermediate lanthanum hydride-nitride(La-H-N) structure to form the final lanthanum nitride product. The hydride ion functions as an electron donor, which provides electrons for Nactivation possibly mediated by the lanthanum atoms. It is observed that N–H bond is not formed during the Nfixation process, which is distinctly different from the alkali or alkaline earth metal hydrides. The hydrolysis of La-H-N to ammonia is feasible using water as the hydrogen source.
