1Fingolimod alters inflammatory mediators and vascular permeability in intracerebral hemorrhage显示文摘Intracerebral hemorrhage(ICH) leads to high rates of death and disability. The pronounced infl ammatory reactions that rapidly follow ICH contribute to disease progression. Our recent clinical trial demonstrated that oral administration of an immune modulator fingolimod restrained secondary injury derived from initial hematoma,but the mechanisms remain unknown. In this study,we aim to investigate the effects of fingolimod on inflammatory mediators and vascular permeability in the clinical trial of oral fingolimod for intracerebral hemorrhage(ICH). The results showed that fingolimod decreased the numbers of circulating CD4^+ T,CD8^+ T,CD(19)^+B,NK,and NKT cells and they recovered quickly after the drug was stopped. The plasma ICAM level was decreased and IL-10 was increased by fingolimod. Interestingly,fingolimod protected vascular permeability as indicated by a decreased plasma level of MMP9 and the reduced r T1%. In conclusion,modulation of systemic inflammation by fingolimod demonstrates that it is an effective therapeutic agent for ICH. 2Modeling the circulation and sediment transport in the Beibu Gulf显示文摘Water circulation and sediment transport in the Beibu Gulf are important for its environmental protection and resource exploitation.By employing the Regional Ocean Modeling System(ROMS),we studied the seasonal variation of circulation,sediment transport and long-term morphological evolution in the Beibu Gulf.The simulation results show that the circulation induced by tide and wind is cyclonic both in winter and summer in the gulf and that the wind-driven circulation is stronger in winter than that in summer.The sediment concentration is higher in the Qiongzhou Strait,west of the Hainan Island and the coast of Vietnam and the Leizhou Peninsula.The sediment is transported westwards in winter and eastwards in summer in the Qiongzhou Strait.The west entrance of the Qiongzhou Strait is dominated by westward transport all the year round.The sediment discharged by rivers is deposited near the river mouths.The simulated result demonstrates that the sediment transport is mainly controlled by tidal induced bottom resuspension in the Beibu Gulf.Four characteristics are summarized for the distribution patterns of erosion and deposition.(1) The erosion and deposition are insignificant in most area of the gulf.(2) Sediment deposition is more significant in the mouths of Qiongzhou Strait.(3) The erosion is observed in the seabed of Qiongzhou Strait.(4) Erosion and deposition occur alternatively in the west of Hainan Island.CHENG Gaolei GONG Wenping WANG Yaping XIA Zhen 2017Acta Oceanologica Sinica2017,36,4:5
3Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. 4Breast cancer-specific serum peptide profiles显示文摘Early detection of breast cancer is paramount to successful clinical therapy. Yet, early-stage breast cancer lacks specific symptoms or biomarkers. With the emerging of the mass spectrometric (MS) -based signatures as biomarkers, we investigated breast cancer-related serum profile pattern through class prediction and independent validation, and used Fourier transfer MS to identify breast cancer signature. We now show a distinctive serum peptide pattern that discriminates breast cancer from healthy controls with 93.2% sensitivity and 95.4% specificity. m/z 5901.70 and 4465.74 of ion fragment of FPA and alpha1-antichymotrypsin are found in the signatures that predominantly discriminate breast cancer from healthy individuals. 5Synthesis of MoX2 (X =Se or S) monolayers with high-concentration 1T'phase on 4H/fcc-Au nanorods for hydrogen evolution显示文摘Controlled synthesis of transition metal dichalcogenide (TMD) monolayers with unusual crystal phases has attracted increasing attention due to their promising applications in electrocatalysis.However,the facile and large-scale preparation of TMD monolayers with high-concentration unusual crystal phase still remains a challenge.Herein,we report the synthesis of MoX2 (X =Se or S) monolayers with high-concentration semimetallic 1T'phase by using the 4H/face-centered cubic (fcc)-Au nanorod as template to form the 4H/fcc-Au@MoX2 nanocomposite.The concentrations of 1T'phase in the prepared MoSe2 and MoS2 monolayers are up to 86% and 81%,respectively.As a proof-of-concept application,the obtained Au@MoS2 nanocomposite is used for the electrocatalytic hydrogen evolution reaction (HER) in acid medium,exhibiting excellent performance with a low overpotential of 178 mV at the current density of 10 mNcm^2,a small Tafel slope of 43.3 mV/dec,and excellent HER stability.This work paves a way for direct synthesis of TMD monolayers with high-concentration of unusual crystal phase for the electrocatalytic application.Zhengqing Liu Xiao Zhang Yue Gong Qipeng Lu Zhicheng Zhang Hongfei Cheng Qinglang Ma Junze Chen Meiting Zhao Bo Chen Ye Chen Xue-Jun Wu Pengfei Yin Lin Gu Yaping Du Hua Zhang 2019Nano Research2019,12,6:3

7Determination of key components in automobile braking systems based on ABC classification and FMECA显示文摘As one of the critical issues in operations management, spare parts planning classification systems based only on cost may not withstand the test of time because of continuing technological advancement or environmental degradation. This study, however, emphasizes using failure mode effect and criticality analysis(FMECA) as a basis for designing an ABC classification system that is capable of more accurately determining critical equipment and making maintenance more efficient. In the proposed methodology, risk priority numbers(RPN) and criticality as an index of safety and reliability can be obtained using a structured failure analysis technique. These indexes can be added to the economic indicator of maintenance cost, and then a classification model of spare parts can be established according to a comprehensive scoring method. The model is calibrated by using case data from a bus braking system, and the components of the braking system are compared and analyzed. The results show that the improved ABC classification method reduces the proportion of key and main components. 8A TLP approach for BGP based on local speculation显示文摘With the explosive growth of Internet,the low efficiency of BGP has seriously in-fluenced its usability. In this work,a TLP(thread level parallelism) approach with local speculation is proposed to improve the BGP performance. The thread parti-tion is locally performed on each separated sub-module at route processing,and the speculation strategy is implemented to guarantee the memory consistency and sequential commit. Experiments on Intel Quad-core server show that this approach reaches an average speedup of 1.46 under single peer,multi-peers and route flap-ping. 10Numerical analysis for contribution of the Tibetan Plateau to dust aerosols in the atmosphere over the East Asia显示文摘Although the Tibetan Plateau is widely thought as a potential dust source to the atmosphere over East Asia,little is known about the temporal changes of Tibetan dust activities and Tibetan dust source strength.In this study,we address these two issues by analyzing dust storm frequencies and aerosol index through remote sensing data and by means of numerical simulation.The findings indicate that monthly dust profiles over the Tibetan Plateau vary significantly with time.Near the surface,dust concentration increases from October,reaches its maximum in February March,and then decreases.In the middle to upper troposphere,dust concentration increases from January,reaches its maximum in May June,and decreases thereafter.Although Tibetan dust sources are important contributors to dust in the atmosphere over the Tibetan Plateau,their contribution to dust in the troposphere over eastern China is weaker.The contribution of Tibetan dust sources to dust in the atmosphere over the Tibetan Plateau decreases sharply with height,from 69% at the surface,40% in the lower troposphere,and 5% in the middle troposphere.Furthermore,the contribution shows seasonal changes,with dust sources at the surface at approximately 80% between November and May and 45% between June and September;in the middle and upper troposphere,dust sources are between 21% from February to March and less than 5% in the other months.Overall,dust aerosols originating from the Tibetan Plateau contribute to less than 10% of dust in East Asia.MAO Rui GONG DaoYi SHAO YaPing WU GuangJian BAO JingDong 2013Science China Earth Sciences2013,56,2:1

18Lenvatinib-and vadimezan-loaded synthetic high-density lipoprotein for combinational immunochemotherapy of metastatic triple-negative breast cancer显示文摘Metastatic triple-negative breast cancer(TNBC)is the most aggressive type of breast cancer.Combination of systemic chemotherapy and immune checkpoint blockade is effective but of limited benefit due to insufficient intratumoral infiltration of cytotoxic T lymphocytes(CTLs)and the accumulation of immunosuppressive cells.Herein,we designed a lenvatinib-and vadimezan-loaded synthetic high-density lipoprotein(LV-sHDL)for combinational immunochemotherapy of metastatic TNBC.The LV-sHDL targeted scavenger receptor class B type 1-overexpressing 4T1 cells in the tumor after intravenous injection.The multitargeted tyrosine kinase inhibitor(TKI)lenvatinib induced immunogenic cell death of the cancer cells,and the stimulator of interferon genes(STING)agonist vadimezan triggered local inflammation to facilitate dendritic cell maturation and antitumor macrophage differentiation,which synergistically improved the intratumoral infiltration of total and active CTLs by 33-and 13-fold,respectively.LV-sHDL inhibited the growth of orthotopic 4T1 tumors,reduced pulmonary metastasis,and prolonged the survival of animals.The efficacy could be further improved when LV-sHDL was used in combination with antibody against programmed cell death ligand 1.This study highlights the combination use of multitargeted TKI and STING agonist a promising treatment for metastatic TNBC.Chao Zheng Wen Zhang Jinming Wang Yihui Zha Fengqin Xiong Ying Cai Xiang Gong Binyu Zhu Helen He Zhu Hao Wang Yaping Li Pengcheng Zhang 2022Acta Pharmaceutica Sinica B2022,12,9:1
19Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology.
