3Biotinylated dextran amine anterograde tracing of the canine corticospinal tract显示文摘In this study, biotinylated dextran amine (BDA) was microinjected into the left cortical motor area of the canine brain. Fluorescence microscopy results showed that a large amount of BDA-labeled pyramidal cells were visible in the left cortical motor area after injection. In the left medulla oblongata, the BDA-labeled corticospinal tract was evenly distributed, with green fluorescence that had a clear boundary with the surrounding tissue. The BDA-positive corticospinal tract entered into the right lateral funiculus of the spinal cord and descended into the posterior part of the right lateral funiculus, close to the posterior horn, from cervical to sacral segments. There was a small amount of green fluorescence in the sacral segment. The distribution of BDA labeling in the canine central nervous system was consistent with the course of the corticospinal tract. Fluorescence labeling for BDA gradually diminished with time after injection. 8Bend family proteins mark chromatin boundaries and synergistically promote early germ cell differentiation显示文摘Understanding the regulatory networks for germ cell fate specification is necessary to developing strategies for improving the efficiency of germ cell production in vitro.In this study,we developed a coupled screening strategy that took advantage of an arrayed bi-molecular fluorescence complementation(BiFC)platform for protein-protein interaction screens and epiblast-like cell(EpiLC)-induction assays using reporter mouse embryonic stem cells(mESCs).Investigation of candidate interaction partners of core human pluripotent factors OCT4,NANOG,KLF4 and SOX2 in EpiLC differentiation assays identified novel primordial germ cell(PGC)-inducing factors including BEN-domain(BEND/Bend)family members.Through RNA-seq,ChIP-seq,and ATAC-seq analyses,we showed that Bend5 worked together with Bend4 and helped mark chromatin boundaries to promote EpiLC induction in vitro.Our findings suggest that BEND/Bend proteins represent a new family of transcriptional modulators and chromatin boundary factors that participate in gene expression regulation during early germline development.Guang Shi Yaofu Bai Xiya Zhang Junfeng Su Junjie Pang Quanyuan He Pengguihang Zeng Junjun Ding Yuanyan Xiong Jingran Zhang Jingwen Wang Dan Liu Wenbin Ma Junjiu Huang Zhou Songyang 2022Protein & Cell2022,13,10:1
9Modified HDDR Process for Producing Anisotropic NdFeB Powders显示文摘The effect of HDDR (hydrogenation-decompositon-desorption recombination process) treating conditions on magnetic ani- sotropy of Nd13FebalCo10B7Ga1Zr0.1 powders has been studied. It is found that increasing recombination temperature is effective for magnetic anisotropy formation, but at the cost of reducing the coercive force. The microstfucture analysis (TEM) indicates that the NdFeB recombination nucleation takes place markedly during the transition range from decomposition stage to recombination stage, in which both temperature and H2 pressure change sharply. In order to create a more stable treatment condition for recombination nucleation, a modified HDDR process is established in which the conventional continuous transition range is divided into several stages. 15The P124A mutation of SRP14 alters its migration on SDS-PAGE without impacting its function显示文摘SRP14 is a crucial protein subunit of the signal recognition particle(SRP),a ribonucleoprotein complex essential for co-translational translocation to the endoplasmic reticulum.During our investigation of SRP14 expression across diverse cell lines,we observe variations in its migration on sodium dodecyl sulfate-polyacrylamide gel electro-phoresis(SDS-PAGE),with some cells exhibiting slower migration and others migrating faster.However,the cause of this phenomenon remains elusive.Our research rules out alternative splicing as the cause and,instead,iden-tifies the presence of a P124A mutation in SRP14(SRP14P124A)among the faster-migrating variants,while the slower-migrating variants lack this mutation.Subsequent ectopic expression of wild-type SRP14P124 or SRP14WT and SRP14P124A in various cell lines confirms that the P124A mutation indeed leads to faster migration of SRP14.Further mutagenesis analysis shows that the P117A and A121P mutations within the alanine-rich domain at the C-terminus of SRP14 are responsible for migration alterations on SDS-PAGE,whereas mutations outside this domain,such as P39A,Y27F,and T45A,have no such effect.Furthermore,the ectopic expression of SRP14WT and SRP14P124A yields similar outcomes in terms of SRP RNA stability,cell morphology,and cell growth,indicating that SRP14P124A represents a natural variant of SRP14 and retains comparable functionality.In conclusion,the substitution of proline for alanine in the alanine-rich tail of SRP14 results in faster migration on SDS-PAGE,but has little effect on its function.Yaofu Liu Jinqiu Zhou 2024Acta Biochimica et Biophysica Sinica2024,56,2:0

17The environmental effect of technical change:A spatial model with city-level data in China显示文摘This paper studies the environmental effects of technical change using a spatial model with panel data from 284 prefecture-cities over 2004-2015 in China.We find that the effects of technical change vary across different dimensions of technical change and different pollution indicators.Furthermore,we also provide robust evidence for the existence of the spatial effects of technical change on environmental pollution across cities.First,indigenous technical change displays three patterns of effects on the four pollutants:a positive effect on wastewater,a negative effect on PM_(2.5)concentrations,and an inverted U-shaped relationship with SO_(2)and soot emissions.The spatial effect of indigenous technical change promotes cleaner industrial productions(fewer emissions of SO_(2),soot and wastewater)but higher PM_(2.5)concentrations.Second,technology transfers from foreign direct investment are associated with less pollution except for wastewater,and their spatial effects are unanimously negative on all pollutants.Finally,absorptive capacity can also promote cleaner industrial production,but its spatial effects can do otherwise.Accordingly,the government should take the spatial spillover effects of technical change into account when implementing specific policies,pin down specific pollutants to make full use of the pollution-reducing effects of technical change,and improve the absorptive capacity of domestic firms.Peng Li Yaofu Ouyang Dan Shi 2021Chinese Journal of Population,Resources and Environment2021,19,1:0

19Anisotropic characteristic in HDDR Nd_(13)Fe_(80)B_7显示文摘The function of disproportionation and desorption stages of hydrogenation-disproportionation-desorption-recombination(HDDR) process in the formation of magnetic anisotropy was studied. The results showed that degree of anisotropy (DOA) ofNd12Fe80B7 induced by appropriate HDDR process is comparable with that of Nd13Fe80-xB7M1 (M=Co, Zr or Ga), however, the moder-ate coercive force of Nd13Fe80B7 magnetic powder led to an undesirable (BH) of 104 kJ/m3 in bonded magnet. Experiments demon-strated that a relatively short disproportionation time, combined with a high desorption temperature and slow desorption rate, duringHDDR process is beneficial to anisotropy attainment, but HDDR Nd13Fe80B7 magnetic powder becomes isotropic if the dispropor-tionation time is longer than a certain period. This phenomenon strongly indicated that the origin of anisotropy in NdFeB-typeHDDR material is contributed by disproportionation stage. XRD (X-ray Diffraction) analysis and TEM (Transmission Electron Mi-croscope) observation told that no other phase except NdH2, α-Fe and Fe2B has been found in the disproportionation step.
