1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 4Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 7Deep genotyping of the gene GmSNAP facilitates pyramiding resistance to cyst nematode in soybean显示文摘Soybean cyst nematode(SCN)is a highly destructive pathogen.The soybean host genome harbors at least two major genes for resistance(rhg1 and Rhg4),as well as a minor locus(SCN3-11).In the present study,a splicing site in GmSNAP11,the potential causal gene of SCN3-11,was identified by comparison of the GmSNAP11 cDNA sequences generated from resistant and susceptible soybean accessions.The sequence information was used to design a codominant CAPS marker,GmSNAP11-2565,which was used to genotype a panel of 209 soybean accessions varying with respect to SCN resistance.Analyses of the effect of the haplotypes formed by GmSNAP11-2565 and another large-effect(nonsynonymous)locus,GmSNAP11-2307,previously identified in GmSNAP11,revealed linkage disequilibrium(P<0.0001)between the two loci,suggesting that GmSNAP11-2565 could be used as a marker for GmSNAP11.GmSNAP11-2565 was accordingly used,along with established markers for GmSNAP18(rhg1)and GmSHMT(Rhg4),to characterize the panel accessions.The mean SCN female index of accessions carrying only the GmSNAP11 allele associated with resistance(20.3%)was higher than that associated with accessions carrying alleles for resistance at both GmSNAP11 and GmSNAP18(12.4%),while the index for accessions carrying alleles for resistance at all of GmSNAP11,GmSNAP18,and GmSHMT was very low(1.9%).Selection on all three markers was effective for maintaining a high level of resistance to SCN race 3.Yu Tian Bo Liu Xuehui Shi Jochen C.Reif Rongxia Guan Ying-hui Li Li-juan Qiu 2019The Crop Journal2019,7,5:2

9Plant pan-genomics and its applications显示文摘Plant genomes are so highly diverse that a substantial proportion of genomic sequences are not shared among individuals.The variable DNA sequences,along with the conserved core sequences,compose the more sophisticated pan-genome that represents the collection of all non-redundant DNA in a species.With rapid progress in genome sequencing technologies,pan-genome research in plants is now accelerating.Here we review recent advances in plant pan-genomics,including major driving forces of structural variations that constitute the variable sequences,methodological innovations for representing the pan-genome,and major successes in constructing plant pan-genomes.We also summarize recent efforts toward decoding the remaining dark matter in telomere-to-telomere or gapless plant genomes.These new genome resources,which have remarkable advantages over numerous previously assembled less-than-perfect genomes,are expected to become new references for genetic studies and plant breeding.Junpeng Shi Zhixi Tian Jinsheng Lai Xuehui Huang 2023Molecular Plant2023,16,1:1

15Chlorophyll-a concentrations in 82 large alpine lakes on the Tibetan Plateau during 2003–2017: temporal–spatial variations and influencing factors显示文摘As essential parts of the unique ecosystem of Tibetan Plateau(TP),the sizes and associated physical properties of alpine lakes have long been investigated.However,little is known about one of the most critical biogeochemical properties,i.e.the Chlorophyll-a(Chl-a)concentrations.Here,for the first time,we presented a comprehensive investigation of the temporal–spatial variations in Chl-a in 82 lakes(>50 km^(2))across the entire TP region,based on MODIS observations in the period of 2003–2017.The results showed that the 82 lakes exhibited an average longterm mean Chl-a of 3.3±4.3 mg m^(−3),with high Chl-a lakes concentrated in the eastern and southern inner TP basin and northeastern parts of the TP.An interannual trend analysis revealed that lakes exhibiting(significantly)decreasing Chl-a trends and(significantly)increasing Chl-a trends were comparable in numbers but differed in distribution patterns.A correlation analysis indicated that at least 70%of the interannual variability in Chl-a values of lakes was significantly correlated with one of the four environmental factors(wind speed,ice cover duration,lake water surface temperature and surface runoff)and lake size.In addition,glacier meltwater tended to reduce lake Chl-a while salinity levels showed minor influences.Xuehui Pi Lian Feng Weifeng Li Junguo Liu Xingxing Kuang Kun Shi Wei Qi Deliang Chen Jing Tang 2021International Journal of Digital Earth2021,14,6:0
16Beamforming design for RIS-aided amplify-and-forward relay networks显示文摘The use of a reconfigurable intelligent surface(RIS)in the enhancement of the rate performance is considered to involve the limitation of the RIS being a passive reflector.To address this issue,we propose a RIS-aided amplify-and-forward(AF)relay network in this paper.By jointly optimizing the beamforming matrix at AF relay and the phase-shift matrices at RIS,two schemes are put forward to address a maximizing signal-to-noise ratio(SNR)problem.First,aiming at achieving a high rate,a high-performance alternating optimization(AO)method based on Charnes–Cooper transformation and semidefinite programming(CCT-SDP)is proposed,where the optimization problem is decomposed into three subproblems solved using CCT-SDP,and rank-one solutions can be recovered using Gaussian randomization.However,the optimization variables in the CCT-SDP method are matrices,leading to extremely high complexity.To reduce the complexity,a low-complexity AO scheme based on Dinkelbachs transformation and successive convex approximation(DT-SCA)is proposed,where the variables are represented in vector form,and the three decoupling subproblems are solved using DT-SCA.Simulation results verify that compared to three benchmarks(i.e.,a RIS-assisted AF relay network with random phase,an AF relay network without RIS,and a RIS-aided network without AF relay),the proposed CCT-SDP and DT-SCA schemes can harvest better rate performance.Furthermore,it is revealed that the rate of the low-complexity DT-SCA method is close to that of the CCT-SDP method.Xuehui WANG Feng SHU Riqing CHEN Peng ZHANG Qi ZHANG Guiyang XIA Weiping SHI Jiangzhou WANG 2023Frontiers of Information Technology & Electronic Engineering2023,24,12:0
