Phosphorite generative processes around the Precambrian-Cambrian boundary in South China: An integrated study of Mo and phosphate O isotopic compositions显示文摘Large phosphorite deposits in Central Guizhou,China, were formed around the Precambrian/Cambrian boundary(PC/C),including the Ediacaran(Doushantuo stage)and early Cambrian(Gezhongwu stage).Among them,Gezhongwu phosphorite from Zhijin are enriched in rare earth elements(REE)plus yttrium(REY),reaching 3.503 million tons.Although phosphorites have attracted great attention, the specific sources P and REY remained unclear. To determine the P and REY sources and establish a phosphogenic model of PC/C phosphorite,we present an integrated dataset of Mo and phosphate O isotopes for the first time, along with carbonate C and O isotopes, geology, petrology, and geochemistry. In all samples,d18Op,Y/Ho,and Zr/Hf decreased from the Ediacaran to the early Cambrian, indicating increased terrigenous weathering fluxes while decreased upwelling water input.Furthermore, terrigenous weathering delivery significantly elevated marine REY concentrations in the Cambrian in Zhijin. The Ceanom and d98/95Mo suggest that seawater was oxidized in the later Ediacaran and became entirely oxic in the early Cambrian. The positive feedback between oxygen levels in atmosphere and primary productivity caused progressive oxygenation in ocean-atmosphere system and enable phosphorites to be formed by different mechanisms. Results show that the Lower Doushantuo consist of abiotic intraclasts and exhibited“seawaterlike”REY types, indicating abiological and mechanical reworking phosphogenesis. The Upper Doushantuo and Gezhongwu Formation contained mainly microbial debris and abiogenic intraclasts, and exhibit “hat-shaped”REY plots, suggesting microbially mediated phosphogenesis.Based on this data set,we developed a phosphogenic model illustrating formation of these two phosphorite deposits, wherein the Lower Doushantuo phosphorite formed through the reworking of pre-existing phosphatic sediments in anoxic and abiotic ocean, whereas the Upper Doushantuo and Gezhongwu phosphorite formed via microbial metabolisms in oxic and biotic conditions. Our study has implications on the PC/C phosphorite generative processes,as well as paleoenvironmental conditions.Haiying Yang Jiafei Xiao Yong Xia Zhuojun Xie Qinping Tan Jianbin Xu Shan He Shengwei Wu Xiqiang Liu Xingxiang Gong 2021Geoscience Frontiers2021,12,5:5
Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency. However, the virus'pathogenesis remains unclear, and there is no cure for the disease. We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution. We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies. In addition,we used three control samples. We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes, and natural killer cells(NKs)in patients in critical condition. The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active, and recovered gradually after patient conditions improved.Consistent with this finding, the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls. The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls, further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens. Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection, providing clues for therapeutic potentials in treating COVID-19.Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang 2021Signal Transduction and Targeted Therapy2021,6,4:4

Enrichment characteristics and sources of the critical metal yttrium in Zhijin rare earth-containing phosphorites, Guizhou Province, China显示文摘Yttrium(Y)is a critical metallic element that is used widely in modern scientific, technological, and industrial applications.Phosphorites in the Zhijin area of Guizhou Province,China, are famous for Y enrichment, but the enrichment characteristics and sources remain unclear.Previous studies suggested that the sources of rare earth elements(REEs,La-Lu)and Y(REYs,La-Lu+Y)were related to hydrothermal deposition. However, this study presents evidence refuting that hypothesis, with major and trace elemental data collected with quadrupoleinductively coupled plasma-mass spectrometry(Q-ICPMS)analysis.These data show that Y in Zhijin REYscontaining phosphorites has a normal distribution and is particularly enriched relative to other REYs.The Y enrichment degree is different at different∑REY intervals.Specifically, the Y enrichment degree is higher at lower∑REY values and lower at higher∑REY values. The REYs-containing phosphorites show features of primary phosphorites. Both REEs and Y have good correlations with P2 O5 in the phosphorites with low REYs contents(total REYs<535 ppm),whereas at high REYs contents(total REYs>535 ppm),REEs have a good correlation with P2 O5 but Y does not.Inconsistent enrichment processes of REYs are suggestive of complex sources of Y.Thus, seafloor hydrothermal fluids were not the direct source of Y.Normal seawater mixed with terrestrial sources might have contributed to the origin of Y here. This study could lead to improvements in Y mineral resource explorations and the situation involving the global REYs supply crisis.Xingxiang Gong Shengwei Wu Yong Xia Zhengwei Zhang Shan He Zhuojun Xie Jiafei Xiao Haiying Yang Qingping Tan Yi Huang Yuhong Yang 2021Acta Geochimica2021,40,3:1
Design and fabrication of superconducting transition edge sensor bolometers with background limited noise performance显示文摘We successfully designed and fabricated TES bolometers utilizing Al/Ti bilayer TESs as sensitive thermometers. To reduce the intrinsic noise level and tune the saturation power of a bolometer, the TES thermometer is placed on a suspended SiN platform which is thermally coupled to the heat bath by four long SiN beams with different geometries. The measurement results show that the detectors have background limited noise performance, with a low noise equivalent power(NEP)on the order of10-17W/Hz1/2and have a saturation power of several tens pW at a bath temperature of 320 mK.These detectors are suitable for applications in ground-based astrophysics experiments by integrating absorbers for specific wavelengths.Qingya Zhang Jianshe Liu Wenhui Dong Tianshun Wang Genfang He Tiefu Li Xingxiang Zhou Wei Chen 2014Chinese Science Bulletin2014,59,19:1
MSC based gene delivery methods and strategies improve the therapeutic efficacy of neurological diseases显示文摘Mesenchymal stem cells(MSCs)are promising seed cells for neural regeneration therapy owing to their plasticity and accessibility. They possess several inherent characteristics advantageous for the transplantation-based treatment of neurological disorders, including neural differentiation, immunosuppression, neurotrophy, and safety. However, the therapeutic efficacy of MSCs alone remains unsatisfactory in most cases. To improve some of their abilities, many studies have employed genetic engineering to transfer key genes into MSCs.Both viral and nonviral methods can be used to overexpress therapeutic proteins that complement the inherent properties. However,to date, different modes of gene transfer have specific drawbacks and advantages. In addition,MSCs can be functionalized through targeted gene modification to facilitate neural repair by promoting neural differentiation, enhancing neurotrophic and neuroprotective functions, and increasing survival and homing abilities. The methods of gene transfer and selection of delivered genes still need to be optimized for improved therapeutic and targeting efficacies while minimizing the loss of MSC function. In this review,we focus on gene transport technologies for engineering MSCs and the application of strategies for selecting optimal delivery genes.Further,we describe the prospects and challenges of their application in animal models of different neurological lesions to broaden treatment alternatives for neurological diseases.Heng Zhou Yan He Wei Xiong Shuili Jing Xingxiang Duan Zhiyun Huang Gurek SNahal Youjian Peng Mingchang Li Yaoqi Zhu Qingsong Ye 2023Bioactive Materials2023,,5:0
High-throughput sequencing detection and ensartinib treatment of lung cancer harboring NTRK1 fusion显示文摘Dear Editor,Although fusion events involving neurotrophic receptor tyrosine kinase 1,2,and 3 genes(NTRK1,NTRK2,and NTRK3,encoding TRKA/B/C respectively)were found in diverse tumor types, only 0.1%-0.3%of lung cancer patients harbor an NTRK(and mostly NTRK1)fusion as the primary oncogenic event[1].Such low prevalence may be partially due to the limited availability of first-line assays for detecting rare fusion events[2].Immunohistochemistry is limited by sensitivity and variable tissue background, and fluorescence in-situ hybridization falls short of elucidating functional significance such as the identity of the partner or structure of the transcript. While DNA next-generation sequencing(NGS)is suitable for mutation calling(single nucleotide variation[SNV]and insertion-or-deletion[indel]),and RNA NGS is particularly effective in detecting fusions[3],routinely performing these two assays together is labor-intensive.As a solution,we have developed a single NGS assay(PANO-Seq)for unified RNA/DNA target enrichment library preparation[4],which takes about 12 hours and $10 to prepare.Zhengbo Song Chunwei Xu Xingxiang Pu Youcai Zhu Wenxian Wang Xingliang Li Yanqiu Gao Wenliang Zhu Yunwei He Lin Wu Li Mao Li Chen Ming Chen 2021Cancer Communications2021,41,2:0
