| 2 | Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). | 3 | A Brief Introduction to BNU-HESM1.0 and Its Earth Surface Temperature Simulations显示文摘Integrated assessment models and coupled earth system models both have their limitations in understanding the interactions between human activity and the physical earth system. In this paper,a new human–earth system model,BNUHESM1.0,constructed by combining the economic and climate damage components of the Dynamic Integrated Model of Climate Change and Economy to the BNU-ESM model,is introduced. The ability of BNU-HESM1.0 in simulating the global CO2 concentration and surface temperature is also evaluated. We find that,compared to observation,BNU-HESM1.0underestimates the global CO2 concentration and its rising trend during 1965–2005,due to the uncertainty in the economic components. However,the surface temperature simulated by BNU-HESM1.0 is much closer to observation,resulting from the overestimates of surface temperature by the original BNU-ESM model. The uncertainty of BNU-ESM falls within the range of present earth system uncertainty,so it is the economic and climate damage component of BNU-HESM1.0 that needs to be improved through further study. However,the main purpose of this paper is to introduce a new approach to investigate the complex relationship between human activity and the earth system. | 4 | Numerical simulation of sedimentation of rectangular particle in Newtonian fluid显示文摘The sedimentation of a rectangular particle falling in a two-dimensional channel filled with Newtonian fluid was simulated with finite element arbitrary Lagrangian-Eulerian domain method. The numerical procedure was validated by comparison of the simulation results with existing numerical work. Moreover, good agreement was obtained between the simulation results and experimental measurements performed in the current study. The equilibrium position, stable orientation and drag coefficient of a rect- angular particle for different particle Reynolds numbers (Re p ) were studied. The results show that there is a critical particle Reynolds number for the preferred orientation of a rectangular particle falling in a Newtonian fluid. When Re p is smaller than the critical value, the particle falls with its long side parallel to gravity; otherwise the particle falls with its long side perpendicular to gravity. The critical particle Reynolds number is a decreasing function of the blockage ratio and aspect ratio. The distributions of pressure and shear stress on rectangular particle surface were analyzed. | 5 | Expression,structure and function analysis of the sperm-oocyte fusion genes Juno and Izumo1 in sheep(Ovis aries)显示文摘Background:JUNO and IZUMO1 are the first receptor-ligand protein pairs discovered to be essential for spermoocyte fusion;their interaction is indispensable for fertilization.Methods:PCR was used to clone the full-length DNA sequence of the Juno gene in sheep.The single nucleotide polymorphism(SNP)loci of Juno were genotyped by Sequenom MassARRAY®.PCR combined with rapid amplification of cDNA Ends were used to clone the full-length cDNA sequence of Juno and Izumo1.Reverse transcriptase-PCR(RT-PCR)and real time-quantitative-PCR(RT-qPCR)were used to analyze the genes’expression in tissues of sheep,and single cell RNA-seq was used to analyze the genes’expression in oocytes,granulosa cells and follicular theca of polytocous and monotocous Small Tail Han ewes.Bioinformatics was used to analyze advanced structure and phylogeny of JUNO and IZUMO1 proteins.Results:The full-length DNA sequence of the Juno gene in sheep was cloned and nine SNPs were screened.We found a significant association between the g.848253 C>A locus of Juno and litter size of Small Tail Han sheep(P<0.05).The full-length cDNA sequence of Juno and Izumo1 genes from Small Tail Han sheep were obtained.We found a new segment of the Izumo1 CDS consisting of 35 bp,and we confirmed the Izumo1 gene has 9 exons,not 8.RT-qPCR showed that Juno and Izumo1 genes were highly expressed in ovarian and testicular tissues,respectively(P<0.01).Single cell RNA-seq showed Juno was specifically expressed in oocytes,but not in granulosa cells or follicular theca,while Izumo1 displayed little to no expression in all three cell types.There was no difference in expression of the Juno gene in oocyte and ovarian tissue in sheep with different litter sizes,indicating expression of Juno is not related to litter size traits.Bioinformatic analysis revealed the g.848253 C>A locus of Juno results in a nonconservative missense point mutation leading to a change from Phe to Leu at position 219 in the amino acid sequence.Conclusions:For the first time,this study systematically analyzed the expression,structure and function of Juno and Izumo1 genes and their encoded proteins in Small Tail Han sheep,providing the basis for future studies of the regulatory mechanisms of Juno and Izumo1 genes. | Wenping Hu Xinlong Dong Zhilong Tian Zhuangbiao Zhang Jishun Tang Benmeng Liang Qiuyue Liu Mingxing Chu | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 3 |

| 12 | Mitochondria-targeting NIR fluorescent probe for rapid,highly sensitive and selective visualization of nitroxyl in live cells,tissues and mice显示文摘Nitroxyl(HNO)has been reported to possess unique biological and pharmacological performances,and emerged as a novel therapy for congestive heart failure.Recent studies also suggest that HNO may be produced and involved in important metabolisms in mitochondria.However,due to its high reactivity and short life properties,fast,sensitive and selective observation and monitoring of HNO related dynamic changes in mitochondria still remains a great challenge.Herein,we synthesized a mitochondria-targeting near-infrared(NIR)fluorescent probe(DCMHNO)for rapid detection of HNO with remarkably high sensitivity,selectivity and photostability.DCMHNO shows fast response(about 4 min)towards HNO via 2-(diphenylphosphino)benzoyl group through the Staudinger reaction to boost the bright NIR emission(700 nm)with excellent sensitivity(detection limit of 13 nM),high p H stability and very low interference from other species.DCMHNO can selectively locate in mitochondria and visualize exogenous and endogenous HNO in live He La cells with high biocompatibility and photostability.The probe could also monitor the interaction between NO and H2 S that gives rise to the generation of HNO in live He La cells.In addition,DCMHNO was further utilized in ex vivo NIR imaging of HNO in live mouse liver tissues at the depth of about 50μm.In vivo imaging of HNO with high signal-to-noise ratio in live mice was also realized by using DCMHNO.These remarkable imaging performances could render NIR DCMNHNO as a useful tool to reveal HNO related dynamic changes in live samples. | Jianguo Wang Wenping Zhu Chunbin Li Pengfei Zhang Guoyu Jiang Guangle Niu Ben Zhong Tang | 2020 | Science China Chemistry2020,63,2: | 0 |

| 14 | Ligand effects on electronic and optoelectronic properties of two-dimensional PbS necking percolative superlattices显示文摘The inter-nanocrystal (NC) distance, necking degree, ordering level, and NC surface ligands all affect the electronic and optoelectronic properties of NC solids. Herein, we introduce a unique PbS structure of necking percolative superlattices to exclude the morphological factors and study the effect of ligands on the NC properties. X-ray photoelectron spectroscopy data indicate that 1,2-ethanedithiol (EDT), oxalic acid, mercaptopropionic acid, and NH4SCN (SCN) ligands were attached to the surface of NCs by substrate-supported ligand exchange. Field-effect transistors were tested and photodetector measurements were performed to compare these NC solids. An SCN-treated film had the highest mobility and responsivity under high-power intensity irradiation owing to its high carrier density, whereas an EDT-treated film had the lowest mobility, photocurrent, and dark current.
