Erianin,a novel dibenzyl compound in Dendrobium extract, inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases, including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment. Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl, exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells, which was accompanied by ROS accumulation, lipid peroxidation, and GSH depletion. The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death, indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together, our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration, and erianin will hopefully serve as a prospective compound for lung cancer treatment. | Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 42
Prediction of undrained shear strength using extreme gradient boosting and random forest based on Bayesian optimization显示文摘Accurate assessment of undrained shear strength(USS)for soft sensitive clays is a great concern in geotechnical engineering practice. This study applies novel data-driven extreme gradient boosting(XGBoost)and random forest(RF)ensemble learning methods for capturing the relationships between the USS and various basic soil parameters.Based on the soil data sets from TC304 database,a general approach is developed to predict the USS of soft clays using the two machine learning methods above, where five feature variables including the preconsolidation stress(PS),vertical effective stress(VES),liquid limit(LL),plastic limit(PL)and natural water content(W)are adopted. To reduce the dependence on the rule of thumb and inefficient brute-force search, the Bayesian optimization method is applied to determine the appropriate model hyper-parameters of both XGBoost and RF.The developed models are comprehensively compared with three comparison machine learning methods and two transformation models with respect to predictive accuracy and robustness under 5-fold cross-validation(CV).It is shown that XGBoost-based and RF-based methods outperform these approaches.Besides, the XGBoostbased model provides feature importance ranks, which makes it a promising tool in the prediction of geotechnical parameters and enhances the interpretability of model. | Wengang Zhang Chongzhi Wu Haiyi Zhong Yongqin Li Lin Wang | 2021 | Geoscience Frontiers2021,12,1: | 33
Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently, the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however, the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown. In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo, accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment. The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death, indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together, our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis, which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23
Single-atom catalyst: a rising star for green synthesis of fine chemicals显示文摘The green synthesis of fine chemicals calls for a new generation of efficient and robust catalysts. Single-atom catalysts(SACs), in which all metal species are atomically dispersed on a solid support, and which often consist of well-defined mononuclear active sites, are expected to bridge homogeneous and heterogeneous catalysts for liquid-phase organic transformations. This review summarizes major advances in the SAC-catalysed green synthesis of fine chemicals in the past several years, with a focus on the catalytic activity, selectivity and reusability of SACs in various organic reactions. The relationship between catalytic performance and the active site structure is discussed in terms of the valence state, coordination environment and anchoring chemistry of single atoms to the support, in an effort to guide the rational design of SACs in this special area, which has traditionally been dominated by homogeneous catalysis. Finally, the challenges remaining in this research area are discussed and possible future research directions are proposed. | Leilei Zhang Yujing Ren Wengang Liu Aiqin Wang Tao Zhang | 2018 | National Science Review2018,5,5: | 15
Temperature field distribution of burnt surrounding rock in UCG stope显示文摘In order to study temperature field distribution in burnt surrounding rock and to determine ranges of burnt surrounding rock, coal-wall coking cycle and heat influence in the underground coal gasification(UCG) stope, based on the Laplace transform and inversion formula, we studied the temperature analytical solution of one-dimensional unsteady heat conduction for multi-layer overlying strata under the first and the forth kinds of boundary conditions, and we also carried out a numerical simulation of twodimensional unsteady heat conduction by the COMSOL multiphysics. The results show that when the boundary temperature of surrounding rock has a linear decrease because of a directional movement of heat source in the UCG flame working face, the temperature in surrounding rock increases first and then decreases with time, the peak of temperature curve decreases gradually and its position moves inside surrounding rock from the boundary. In the surrounding rock of UCG stope, there is an envelope curve of temperature curve clusters. We analyzed the influence of thermophysical parameters on envelope curves and put forward to take envelope curve as the calculation basis for ranges of burnt surrounding rock, coal-wall coking cycle and heat influence. Finally, the concrete numerical values are given by determining those judgement standards and temperature thresholds, which basically tally with the field geophysical prospecting results. | Xin Lin Wang Zuotang Huang Wengang Kang Guojun Lu Xuefeng Zhang Peng Wang Jianhua | 2014 | International Journal of Mining Science and Technology2014,24,4: | 10
A new strawberry harvesting robot for elevated-trough culture显示文摘In order to improve robotic harvesting and reduce production cost,a harvesting robot system for strawberry on the elevated-trough culture was designed. It was supposed to serve for sightseeing agriculture and technological education.Based on the sonar-camera sensor,an autonomous navigation system of the harvesting robot was built to move along the trough lines independently. The mature fruits were recognized according to the H(Hue)and S(Saturation)color feature and the picking-point were located by the binocular-vision unit.A nondestructive end-effector,used to suck the fruit, hold and cut the fruit-stem,was designed to prevent pericarp damage and disease infection.A joint-type industrial manipulator with six degrees-of-freedom(DOF)was utilized to carry the end-effector.The key points and time steps for the collision-free and rapid motion of manipulator were planned.Experimental results showed that all the 100 mature strawberry targets were recognized automatically in the harvesting test. The success harvesting rate was 86%,and the success harvesting operation cost 31.3 seconds on average, including a single harvest operation of 10 seconds. The average error for fruit location was less than 4.6 mm. | Feng Qingchun Wang Xiu Zheng Wengang Qiu Quan Jiang Kai | 2012 | International Journal of Agricultural and Biological Engineering2012,5,2: | 8
Effect of butanol on flotation separation of quartz from hematite with N-dodecyl ethylenediamine显示文摘In order to investigate the effect of butanol on quartz flotation when N-dodecyl ethylenediamine(ND)was used as collector, single mineral flotation and artificial mixed mineral(hematite and quartz were mixed at a mass ratio of 3:2) separation were conducted in the laboratory. Experimental results indicated that addition of butanol could improve the collecting performance of ND on quartz and enhance the floatability of quartz. Best flotation recovery of quartz was obtained when butanol was mixed with ND at a mass ratio of 1:1. Moreover, the molecular structure of alcohols had a significant effect on mineral recovery. Best separation efficiency could be obtained when tert-butanol was added as it had the largest cross-sectional area. Zeta potential measurements indicated that alcohols could strengthen electrostatic adsorption between quartz and collector. Molecular dynamic simulations revealed that co-adsorption of alcohols along with ND had taken place on the quartz surface, and ND/tert-butyl combinations were more easily absorbed on the quartz surface. | Liu Wengang Liu Wenbao Wang Xinyang Wei Dezhou Zhang Hao Liu Wei | 2016 | International Journal of Mining Science and Technology2016,26,6: | 7

Neutralization of interleukin-17A alleviates burn-induced intestinal barrier disruption via reducing pro-inflammatory cytokines in a mouse model显示文摘Background:The intestinal barrier integrity can be disrupted due to burn injury, which is responsible for local and systemic inflammatory responses.Anti-inflammation strategy is one of the proposed therapeutic approaches to control inflammatory cascade at an early stage.Interleukin-17A(IL-17A)plays a critical role in inflammatory diseases. However, the role of IL-17A in the progression of burn-induced intestinal inflammation is poorly understood. In this study,we aimed to investigate the effect of IL-17A and associated pro-inflammatory cytokines that were deeply involved in the pathogenesis of burn-induced intestinal inflammatory injury, and furthermore,we sought to determine the early source of IL-17A in the intestine. Methods:Mouse burn model was successfully established with infliction of 30%total body surface area scald burn. The histopathological manifestation, intestinal permeability, zonula occludens-1 expression,pro-inflammatory cytokines were determined with or without IL-17A-neutralization.Flow cytometry was used to detect the major source of IL-17A^(+)cells in the intestine. Results:Burn caused intestinal barrier damage, increase of intestinal permeability, alteration of zonula occludens-1 expressions, elevation of IL-17A,IL-6,IL-1βand tumor necrosis factor-α(TNF-α),whereas IL-17A neutralization dramatically alleviated burn-induced intestinal barrier disruption, maintained zonula occludens-1 expression, and noticeably, inhibited pro-inflammatory cytokines elevation. In addition,we observed that the proportion of intestinal IL-17A^(+)Vγ4^(+)T subtype cells(but not IL-17A^(+)Vγ1^(+)T subtype cells)were increased in burn group, and neutralization of IL-17A suppressed this increase. Conclusions:The main original findings of this study are intestinal mucosa barrier is disrupted after burn through affecting the expression of pro-inflammatory cytokines, and a protective role of IL-17A neutralization for intestinal mucosa barrier is determined.Furthermore,Vγ4^(+)T cells are identified as the major early producers of IL-17A that orchestrate an inflammatory response in the burn model. These data suggest that IL-17A blockage may provide a unique target for therapeutic intervention to treat intestinal insult after burn. | Yajun Song Yang Li Ya Xiao Wengang Hu Xu Wang Pei Wang Xiaorong Zhang Jiacai Yang Yong Huang Weifeng He Chibing Huang | 2019 | Burns & Trauma2019,7,1: | 5
Influence of groundwater drawdown on excavation responses e A case history in Bukit Timah granitic residual soils显示文摘Performances of a braced cut-and-cover excavation system for mass rapid transit(MRT) stations of the Downtown Line Stage 2 in Singapore are presented. The excavation was carried out in the Bukit Timah granitic(BTG) residual soils and characterized by significant groundwater drawdown, due to dewatering work in complex site conditions, insufficient effective waterproof measures and more permeable soils. A two-dimensional numerical model was developed for back analysis of retaining wall movement and ground surface settlement. Comparisons of these measured excavation responses with the calculated performances were carried out, upon which the numerical simulation procedures were calibrated. In addition, the influences of groundwater drawdown on the wall deflection and ground surface settlement were numerically investigated and summarized. The performances were also compared with some commonly used empirical charts, and the results indicated that these charts are less applicable for cases with significant groundwater drawdowns. It is expected that these general behaviors will provide useful references and insights for future projects involving excavation in BTG residual soils under significant groundwater drawdowns. | Wengang Zhang Wei Wang Dong Zhou Runhong Zhang A.T.C. | 12 | Genome wide association study identifies SNPs associated with fatty acid composition in Chinese Wagyu cattle显示文摘Background: Fatty acids are important traits that affect meat quality and nutritive values in beef cattle. Detection of genetic variants for fatty acid composition can help to elucidate the genetic mechanism underpinning these traits and promote the improvement of fatty acid profiles. In this study, we performed a genome-wide association study(GWAS) on fatty acid composition using high-density single nucleotide polymorphism(SNP) arrays in Chinese Wagyu cattle. Results: In total, we detected 15 and 8 significant genome-wide SNPs for individual fatty acids and fatty acid groups in Chinese Wagyu cattle, respectively. Also, we identified nine candidate genes based on 100 kb regions around associated SNPs. Four SNPs significantly associated with C14:1 cis-9 were embedded with stearoyl-CoA desaturase(SCD), while three SNPs in total were identified for C22:6 n-3 within Phospholipid scramblase family member 5(PLSCR5), Cytoplasmic linker associated protein 1(CLASP1), and Chymosin(CYM). Notably, we found the top candidate SNP within SCD can explain ~ 7.37% of phenotypic variance for C14:1 cis-9.Moreover, we detected several blocks with high LD in the 100 kb region around SCD. In addition, we found three significant SNPs within a 100 kb region showing pleiotropic effects related to multiple FA groups(PUFA,n-6, and PUFA/SFA), which contains BAI1 associated protein 2 like 2(BAIAP2 L2), MAF bZIP transcription factor F(MAFF),and transmembrane protein 184 B(TMEM184 B).Conclusions: Our study identified several significant SNPs and candidate genes for individual fatty acids and fatty acid groups in Chinese Wagyu cattle, and these findings will further assist the design of breeding programs for meat quality in cattle. | Zezhao Wang Bo Zhu Hong Niu Wengang Zhang Ling Xu Lei Xu Yan Chen Lupei Zhang Xue Gao Huijiang Gao Shengli Zhang Lingyang Xu Junya Li | 2019 | Journal of Animal Science and Biotechnology2019,10,2: | 4
PCDH17 increases the sensitivity of colorectal cancer to 5-fluorouracil treatment by inducing apoptosis and autophagic cell death显示文摘5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical success. Our previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in CRC.However, the relationship between PCDH17 and 5-FU resistance in CRC remains unclear. Here,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues, and high expression of PCDH17 was correlated with high BECN1 expression. Moreover, this expression profile contributed to superior prognosis and increased survival in CRC patients.Restoring PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell death.Furthermore, autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor Z-VAD-FMK.PCDH17 inhibition by siRNA decreased the autophagy response and 5-FU sensitivity.Mechanistically,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced autophagy. The compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC cells.Taken together, our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell death.PCDH17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients. | Shuiping Liu Haoming Lin Da Wang Qiang Li Hong Luo Guoxiong Li Xiaohui Chen Yongqiang Li Peng Chen Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Mingming Zhang Xuemeng Han Qiujie Li Liuxi Chen Ying Liu Xiaying Chen Guohua Li Yu Xiang Ting Duan Jiao Feng Jianshu Lou Xingxing Huang Qin Zhang Ting Pan Lili Yan Ting Jin Wenzheng Zhang Lvjia Zhuo Yitian Sun Tian Xie Xinbing Sui | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 4
TBM penetration rate prediction based on the long short-term memory neural network显示文摘Tunnel boring machines(TBMs)are widely used in tunnel engineering because of their safety and efficiency. The TBM penetration rate(PR)is crucial,as its real-time prediction can reflect the adaptation of a TBM under current geological conditions and assist the adjustment of operating parameters. In this study, deep learning technology is applied to TBM performance prediction, and a PR prediction model based on a long short-term memory(LSTM)neuron network is proposed. To verify the performance of the proposed model, the machine parameters, rock mass parameters, and geological survey data from the water conveyance tunnel of the Hangzhou Second Water Source project were collected to form a dataset.Furthermore,2313 excavation cycles were randomly composed of training datasets to train the LSTM-based model, and 257 excavation cycles were used as a testing dataset to test the performance. The root mean square error and the mean absolute error of the proposed model are 4.733 and 3.204,respectively.Compared with Recurrent neuron network(RNN)based model and traditional time-series prediction model autoregressive integrated moving average with explanation variables(ARIMAX),the overall performance on proposed model is better. Moreover,in the rapidly increasing period of the PR,the error of the LSTM-based model prediction curve is significantly smaller than those of the other two models. The prediction results indicate that the LSTM-based model proposed herein is relatively accurate, thereby providing guidance for the excavation process of TBMs and offering practical application value. | Boyang Gao RuiRui Wang Chunjin Lin Xu Guo Bin Liu Wengang Zhang | 2021 | Underground Space2021,6,6: | 4
Slope stability prediction using ensemble learning techniques: A case study in Yunyang County, Chongqing, China显示文摘Slope stability prediction plays a significant role in landslide disaster prevention and mitigation. This study develops an ensemble learning-based method to predict the slope stability by introducing the random forest(RF)and extreme gradient boosting(XGBoost). As an illustration, the proposed approach is applied to the stability prediction of 786 landslide cases in Yunyang County,Chongqing,China. For comparison, the predictive performance of RF,XGBoost, support vector machine(SVM),and logistic regression(LR)is systematically investigated based on the well-established confusion matrix, which contains the known indices of recall rate, precision, and accuracy.Furthermore, the feature importance of the 12 influencing variables is also explored. Results show that the accuracy of the XGBoost and RF for both the training and testing data is superior to that of SVM and LR,revealing the superiority of the ensemble learning models(i.e.XGBoost and RF)in the slope stability prediction of Yunyang County.Among the 12 influencing factors, the profile shape is the most important one.The proposed ensemble learning-based method offers a promising way to rationally capture the slope status. It can be extended to the prediction of slope stability of other landslide-prone areas of interest. | Wengang Zhang Hongrui Li Liang Han Longlong Chen Lin Wang | 2022 | Journal of Rock Mechanics and Geotechnical Engineering2022,14,4: | 4
Seismic behaviors of utility tunnel-soil system: With and without joint connections显示文摘Seismic responses of utility tunnel-soil system were studied via shaking table model tests with considerations of two kinds of double box utility tunnels:with and without joint connections. These two testing utility tunnel models were made of galvanized iron wire and micro-concrete,and the ground was simulated by the dry standard sand through layered tamping treatment. The utility tunnel-soil system was subjected to horizontal vibration in uniaxial direction perpendicular to the longitudinal direction of tunnel model.Via instrumentations of earth pressure gauges, accelerometers and strain gauges, the earth pressure response, acceleration response and bending moment response were measured. The testing results show that the joint connections in the utility tunnel along the longitudinal direction play an important role in determining the characteristic of earth pressure response and bending moment response, whereas the effect of joint connections on acceleration response is less significant. In addition, the partition wall exhibits the consistent acceleration response with the side-wall of double box utility tunnel model under seismic condition.Based on the testing results,it is suggested that the joint connection should be taken reasonably into consideration during design and construction for engineering practice. | Liang Han Hanlong Liu Wengang Zhang Xuanming Ding Zhixiong Chen Li Feng Zhenyu Wang | 2022 | Underground Space2022,7,5: | 3
Extraction mechanism of rare earths from chloride acidic solution with ammonium-bifunctionalized ionic liquid extractants显示文摘A series of ammonium-bifunctionalized ionic liquid extractants(ammonium-Bif-ILEs) combined with a number of anions, including carboxylic acids and 1,3-diketonates, have been prepared and studied in this report. Their extraction behavior and properties toward rare earth ions(REs(Ⅲ)) are systematically investigated in chloride media as a function of important parameters such as aqueous phase p H, salting-out agent concentrations, and extraction temperature. The separation performance of ammonium-Bif-ILEs toward REs(Ⅲ) are systematically discussed. The results demonstrate that ammonium-Bif-ILEs have a synergistic effect between the cation and anions in separation of REs(Ⅲ). The influences of different anions on separation factor(b) values are further studied. By comparison, ammonium-Bif-ILEs containing 1,3-diketonates have more potential applications in La(Ⅲ)/RE(Ⅲ) separation than those containing carboxylic acids. | Hualing Yang Ji Chen Wei Wang Hongmin Cui Wengang Liu Yu Liu | 2016 | Science China Chemistry2016,59,5: | 2
