| 2 | Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD. | Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 3 |

| 5 | Effect of AAPH oxidation on digestion characteristics of seed watermelon(Citrullus lanatus var)kernels protein isolates显示文摘Seed watermelon kernel is a typical complex food with high fat and protein contents.During storage and processing,it is often affected by various factors to undergo interactions between components,which lead to its quality change.In this experiment,seed watermelon kernels were used as research objects,and the effects of 2-Azobis(2-amidinopropane)dihydrochloride(AAPH)on seed watermelon kernel protein isolates(WMP)were investigated.The structure and digestion characteristics of WMP after oxidation were studied.The results showed that with the increase of AAPH concentration(0.05−5 mol/L),WMP showed obvious aggregation,and its solubility decreased from 6.76 mg/mL to 9.59 mg/mL.The free sulfhydryl content of WMP was 18.24 mmol/g decreased to 11.25 mmol/g,α-helix decreased andβ-sheet decreased in secondary structure,and its disulfide bond increased by 43.06 mmol/g from 39.57 mmol/g,enthalpy(H)and denaturation temperature increased(Td)(P<0.05).By mass spectrometry results of simulated gastric digestion products,it was found that oxidation adversely affected the digestion characteristics of WMP.It can be seen that the lipid oxidation product APPH of seed watermelon kernel can significantly affect the structure and function of the protein extracted from the seed kernel. | Shugang Li Zhihao Li Xiuting Li Ping Wang Xiongwei Yu Qinli Fu Sihai Gao | 2020 | Food Science and Human Wellness2020,9,4: | 3 |

| 11 | A near-complete genome assembly of the allotetrapolyploid Cenchrus fungigraminus(JUJUNCAO)provides insights into its evolution and C4 photosynthesis显示文摘JUJUNCAO(Cenchrus fungigraminus;2n=4x=28)is a Cenchrus grass with the highest biomass production among cultivated plants,and it can be used for mushroom cultivation,animal feed,and biofuel production.Here,we report a nearly complete genome assembly of JUJUNCAO and reveal that JUJUNCAO is an allopolyploid that originated2.7 million years ago(mya).Its genome consists of two subgenomes,and subgenome A shares high collinear synteny with pearl millet.We also investigated the genome evolution of JUJUNCAO and suggest that the ancestral karyotype of Cenchrus split into the A and B ancestral karyotypes of JUJUNCAO.Comparative transcriptome and DNA methylome analyses revealed functional divergence of homeologous gene pairs between the two subgenomes,which was a further indication of asymmetric DNA methylation.The three types of centromeric repeat in the JUJUNCAO genome(CEN137,CEN148,and CEN156)may have evolved independently within each subgenome,with some introgressions of CEN156 from the B to the A subgenome.We investigated the photosynthetic characteristics of JUJUNCAO,revealing its typical C4 Kranz anatomy and high photosynthetic efficiency.NADP-ME and PEPCK appear to cooperate in the major C4 decarboxylation reaction of JUJUNCAO,which is different from other C4 photosynthetic subtypes and may contribute to its high photosynthetic efficiency and biomass yield.Taken together,our results provide insights into the highly efficient photosynthetic mechanism of JUJUNCAO and provide a valuable reference genome for future genetic and evolutionary studies,as well as genetic improvement of Cenchrus grasses. | Huakun Zheng Baiyu Wang Xiuting Hua Ruiting Gao Yuhao Wang Zixin Zhang Yixing Zhang Jing Mei Yongji Huang Yumin Huang Hui Lin Xingtan Zhang Dongmei Lin Siren Lan Zhongjian Liu Guodong Lu Zonghua Wang Ray Ming Jisen Zhang Zhanxi Lin | 2023 | Plant Communications2023,4,5: | 1 |
| 12 | CNTs/CNF-supported multi-active components as highly efficient bifunctional oxygen electrocatalysts and their applications in zincair batteries显示文摘Rational construction of active components has been the biggest challenge in preparing efficient bifunctional oxygen electrocatalysts.Herein,electrospinning and chemical vapor deposition(CVD)were employed to embed active species including FeCo nanoparticles,MNx(M=Fe,Co),and FePx in porous and graphitized carbon nanotubes(CNTs)/carbon nanofiber(CNF).The as-prepared FeCo@CoNx@FePx/C exhibited a half-wave potential as high as 0.86 V in oxygen reduction reaction(ORR)and low oxygen evolution reaction(OER)overpotential of 368 mV at 10 mA·cm^(−2),which are superior to Pt/C(0.83 V)and IrO_(2)(375 mV)respectively.The assembled Zn-air battery(ZAB)showed a high energy efficiency(Edischarge/Echarge)of 65%at 20 mA·cm^(−2)and stabilized for 700 charge-discharge cycles.The spectroscopic and microscopic characterizations evidenced that the outstanding bifunctionality of the electrocatalyst can be ascribed to three main reasons:First,FeCo nanoparticles are rich in MOH/MOOH active sites for OER,and FePx/CNTs constructed with CVD also modulate electronic structure to improve electron transfer;second,both MNx in carbon matrix and FePx/CNTs are highly active towards ORR;third,the CNTs/CNF are highly porous and graphitized,which promotes mass transport and improves electrical conductivity and stability of the electrocatalysts.This work gives important implications on the design of bifunctional electrocatalysts. | Weiyuan Ding Ali Saad Yuchen Wu Zhiwei Wang Xiuting Li | 2023 | Nano Research2023,16,4: | 0 |

| 14 | Effects of time-delayed vibration absorber on bandwidth of beam for low broadband vibration suppression显示文摘The effects of time-delayed vibration absorber(TDVA) on the dynamic characteristics of a flexible beam are investigated. First, the vibration suppression effect of a single TDVA on a continuous beam is studied. The first optimization criterion is given,and the results show that the introduction of time-delayed feedback control(TDFC) is beneficial to improving the vibration suppression at the anti-resonance band. When a single TDVA is used, the anti-resonance is located at a specific frequency by the optimum design of TDFC parameters. Then, in order to obtain low-frequency and broad bands for vibration suppression, multiple TDVAs are uniformly distributed on a continuous beam,and the relationship between the dynamic responses and the TDFC parameters is investigated. The obtained relationship shows that the TDVA has a significant regulatory effect on the vibration behavior of the continuous beam. The effects of the number of TDVAs and the nonlinearity on the bandgap variation are discussed.
