| 2 | 合并于肝细胞癌的门静脉血栓形成增强超声与螺旋CT的检出及定性显示文摘本研究的目的是对比增强超声(CEUS)与螺旋CT对肝细胞癌(HCC)并发门静脉血栓形成的检出与定性。50例US和多普勒US发现门静脉内血栓形成并经穿刺活检证实的HCC病人被纳入研究。其中13个血栓累及门静脉主干,37个血栓累及门静脉肝段分支。血栓活检后1周内行CEUS和CT检查。由有经验的放射医生以盲法根据每种影像技术分别做出血栓(有/无)与血栓性质(恶性/良性)的诊断,并同病理结果对照,确定血栓检出与定性的准确性。 | S. | 3 | Using the polymerase chain reaction coupled with denaturing gradient gel electrophoresis to investigate the association between bacterial translocation and systemic inflammatory response syndrome in predicted acute severe pancreatitis显示文摘AIM: To investigate the use of PCR and DGGE to investigate the association between bacterial translocation and systemic inflammatory response syndrome in predicted severe AP.METHODS: Patients with biochemical and clinical evidence of acute pancreatitis and an APACHE Ⅱ score ≥8 were enrolled. PCR and DGGE were employed to detect bacterial translocation in blood samples collected on d1,3, and 8 after the admission. Standard microbial blood cultures were taken when there was clinical evidence of sepsis or when felt to be clinically indicated by the supervising team.RESULTS: Six patients were included. Of all the patients investigated, only one developed septic complications;the others had uneventful illness. Bacteria were detected using PCR in 4 of the 17 collected blood samples. The patient with sepsis was PCR-positive in two samples (taken on d 1 and 3), despite three negative blood cultures. Using DGGE and specific primers, the bacteria in all blood specimens which tested positive for the presence of bacterial DNA were identified as E coli.CONCLUSION: Our study confirmed thatunlike traditional microbiological techniques, PCR can detect the presence of bacteria in the blood of patients with severe AP. Therefore, this latter method in conjunction with DGGE is potentially an extremely useful tool in predicting septic morbidity and evaluating patients with the disease. | 4 | Indole phytoalexin derivatives induce mitochondrialmediated apoptosis in human colorectal carcinoma cells显示文摘AIM To investigate the mechanism of the antiproliferative effect of synthetic indole phytoalexin derivatives on human colorectal cancer cell lines.METHODS Changes in cell proliferation and the cytotoxic effect of the tested compounds on human colorectal cancer cell lines and human fibroblasts were evaluated using MTS and Brd U assay,allowing us to choose the most potent substance.Cell cycle alterations were analyzed using flow cytometric analysis.The apoptosis-inducing effect of compound K-453 on the HCT116 cell line was examined with annexin V/PI double staining using flow cytometry,as well as acridine orange/propidium iodide(AO/PI)staining.The flow cytometry method also allowed us to measure changes in levels or activation states of other factors associated with apoptosis,such as poly(ADP-ribose)polymerase(PARP),caspase-3 and-9,cytochrome c,Bcl-2 family proteins,and also the integrity of the mitochondrial membrane.To evaluate activity of the transcription factors and proteins involved in signaling pathways we used Western blot analysis together with flow cytometry.RESULTS Among the ten tested compounds,compound K-453{(±)-trans-1,2-dimethoxy-2’-(3,5-bis-trifluoromethylphenylamino)spiro{indoline-3,5’[4’,5’]dihydrothiazol}exhibited the most potent activity with IC50=32.22±1.14μmol/L in human colorectal HCT116 cells and was thus selected for further studies.Flow cytometric analysis revealed a K-453-induced increase in the population of cells with sub-G1 DNA content,which is considered as a marker of apoptotic cell death.The apoptosis-inducing effect of compound K453 was also confirmed by annexin V/PI double staining and AO/PI staining.The apoptosis was associated with the loss of mitochondrial membrane potential,PARP cleavage,caspase-3 and caspase-9 activation,release of cytochrome c,as well as changes in the levels of Bcl-2family members.Moreover,flow cytometry showed that compound K-453 stimulates phosphorylation of p38MAPK but decreases phosphorylation of Akt and Erk1/2.Activation of p38 MAPK was also confirmed using Western blot analysis.This analysis also revealed downregulation of NF-κB1(p50)and Rel A(p65)proteins and the loss of their anti-apoptotic activity.CONCLUSION In our study compound K-453 exhibited an antiproliferative effect by induction of intrinsic apoptosis as well as modulation of several signaling pathways. | Viera Tischlerova Martin Kello Mariana Budovska Jan Mojzis | 2017 | World Journal of Gastroenterology2017，23，24： | 4 |
| 5 | Biosorption of mercury by Macrocystis pyrifera and Undaria pinnatifida:Influence of zinc,cadmium and nickel显示文摘This study investigated the adsorption of Hg(Ⅱ) on Macrocystis pyrifera and Undaria pinnatifida in monometallic system in the presence of Zn(II),Cd(Ⅱ) and Ni(II).The two biosorbents reached the same maximum sorption capacity (q m = 0.8 mmol/g) for mercury.U.pinnatifida showed a greater affinity (given by the coefficient b of the Langmuir equation) for mercury compared to M.pyrifera (4.4 versus 2.7 L/mmol).Mercury uptake was significantly reduced (by more than 50%) in the presence of competitor heavy metals such as Zn(II),Cd(II) and Ni(Ⅱ).Samples analysis using an environmental scanning electron microscopy equipped with an energy dispersive X-ray microanalysis showed that mercury was heterogeneously adsorbed on the surface of both biomaterials,while the other heavy metals were homogeneous distributed.The analysis of biosorbents by Fourier transform infrared spectrometry indicated that Hg(Ⅱ) binding occurred on S=O (sulfonate) and N-H (amine) functional groups. | Josefina Plaza Marisa Viera Edgardo Donati Eric Guibal | 2011 | Journal of Environmental Sciences2011，23，11： | 3 |

Most of the neuronal damage is caused by complex of biochemical reactions and substantial processes, such as protein agregation, reactions of free radicals, insufficient blood supply, glutamate excitotoxicity, and oxidative stress. The result of these processes can be apoptotic or necrotic cell death and it can lead to an irreversible damage. Therefore, neuroprotection and prevention of the neurodegeneration are highly important topics to study. There are several approaches to prevent the ischemic damage.
