1Alzheimer’s disease and type 2 diabetes mellitus:Pathophysiologic and pharmacotherapeutics links显示文摘At present,Alzheimer’s disease(AD)and type 2 diabetes mellitus(T2DM)are two highly prevalent disorders worldwide,especially among elderly individuals.T2DM appears to be associated with cognitive dysfunction,with a higher risk of developing neurocognitive disorders,including AD.These diseases have been observed to share various pathophysiological mechanisms,including alterations in insulin signaling,defects in glucose transporters(GLUTs),and mitochondrial dysfunctions in the brain.Therefore,the aim of this review is to summarize the current knowledge regarding the molecular mechanisms implicated in the association of these pathologies as well as recent therapeutic alternatives.In this context,the hyperphosphorylation of tau and the formation of neurofibrillary tangles have been associated with the dysfunction of the phosphatidylinositol 3-kinase and mitogen-activated protein kinase pathways in the nervous tissues as well as the decrease in the expression of GLUT-1 and GLUT-3 in the different areas of the brain,increase in reactive oxygen species,and production of mitochondrial alterations that occur in T2DM.These findings have contributed to the implementation of overlapping pharmacological interventions based on the use of insulin and antidiabetic drugs,or,more recently,azeliragon,amylin,among others,which have shown possible beneficial effects in diabetic patients diagnosed with AD.Milagros Rojas Mervin Chávez-Castillo Jordan Bautista Ángel Ortega Manuel Nava Juan Salazar Edgar Díaz-Camargo Oscar Medina Joselyn Rojas-Quintero Valmore Bermúdez 2021World Journal of Diabetes2021,12,6:5

3Sepsis-induced mitochondrial dysfunction:A narrative review显示文摘Sepsis represents a deranged and exaggerated systemic inflammatory response to infection and is associated with vascular and metabolic abnormalities that trigger systemic organic dysfunction.Mitochondrial function has been shown to be severely impaired during the early phase of critical illness,with a reduction in biogenesis,increased generation of reactive oxygen species and a decrease in adenosine triphosphate synthesis of up to 50%.Mitochondrial dysfunction can be assessed using mitochondrial DNA concentration and respirometry assays,particularly in peripheral mononuclear cells.Isolation of monocytes and lymphocytes seems to be the most promising strategy for measuring mitochondrial activity in clinical settings because of the ease of collection,sample processing,and clinical relevance of the association between metabolic alterations and deficient immune responses in mononuclear cells.Studies have reported alterations in these variables in patients with sepsis compared with healthy controls and non-septic patients.However,few studies have explored the association between mitochondrial dysfunction in immune mononuclear cells and unfavorable clinical outcomes.An improvement in mitochondrial parameters in sepsis could theoretically serve as a biomarker of clinical recovery and response to oxygen and vasopressor therapies as well as reveal unexplored pathophysiological mechanistic targets.These features highlight the need for further studies on mitochondrial metabolism in immune cells as a feasible tool to evaluate patients in intensive care settings.The evaluation of mitochondrial metabolism is a promising tool for the evaluation and management of critically ill patients,especially those with sepsis.In this article,we explore the pathophysiological aspects,main methods of measurement,and the main studies in this field.Wagner Nedel Caroline Deutschendorf Luis Valmor Cruz Portela 2023World Journal of Critical Care Medicine2023,12,3:1

15Hybrid Surface Mesh Adaptation for Climate Modeling显示文摘Solution-driven mesh adaptation is becoming quite popular for spatial error control in the numerical simulation of complex computational physics applications,such as climate modeling.Typically,spatial adaptation is achieved by element subdivision (h adaptation) with a primary goal of resolving the local length scales of interest.A sec- ond,less-popular method of spatial adaptivity is called'mesh motion'(r adaptation); the smooth repositioning of mesh node points aimed at resizing existing elements to capture the local length scales.This paper proposes an adaptation method based on a combination of both element subdivision and node point repositioning (rh adaptation). 16Evolution of the Maturation Point of Wine Grapes Intended for Juice Processing显示文摘Rainy years especially in the ripening period end up weakening grape quality, thus making it unsatisfactory for winemaking. The possibility of using wine grape varieties in the preparation of juices presents an alternative to the grapes that do not reach ideal harvest for winemaking. Assuming that the preparation of juice from wine grapes can be an alternative to these grapes which in harvests are considered poor for winemaking and can be harvested in advance, thus preventing losses from late cycle disease and excessive spending on phytosanitary treatments, the present study aimed to evaluate the juices made from “Cabernet Sauvignon”,“Merlot” and “Chardonnay” grapes harvested at different points of maturation, and subsequently compare with current legislation. Physicochemical analysis of titratable acidity (TA), soluble solids (SS), relative density, total sugar, alcohol, volatile acidity and SS/TA ratio was carried out on the juices produced at three sampling points (15, 17 and 19 °Brix). The results showed variability between treatment factors (cultivars and points of maturation). Analyses of titratable acidity, soluble solids, SS/TA ratio, relative density, total sugars and volatile acidity showed significance for the interaction between the tested treatment factors. For the variable alcohol, there was no significance for the effect of variety, maturation stage or the interaction between the factors of treatment. 17Bacteria in cancer therapy: beyond immunostimulation显示文摘Currently,conventional therapies in cancer are improving;chemotherapy,radiotherapy and surgery have increased survival significantly.New therapies have arisen with the same goal;immunotherapy has appeared as a promising option in the fight against cancer stimulating the immune system by inducing innate and adaptive responses.These responses include release of pro-inflammatory cytokines,making the immune system capable to eliminate or protect against multiple tumors.Nowadays,many of these therapies are being used in clinical settings,such as checkpoint inhibitors,monoclonal anti cytotoxic T-Lymphocyte associated protein 4(CTL-4)and programmed death protein 1(PD1),with inspiring results;however,they may decrease immunotolerance,limiting their use.At the same time,chemotherapy works by passive transport across the cell membrane,limiting its capacity to penetrate in tumor cells.For these reasons,bacteria employment represents one of the best candidates for cancer treatment.They can surpass these barriers with their selective colonization which also has an oncolytic effect by increasing proliferation and immunostimulation in the tumor environment.Attenuated strains,such as Mycobacterium bovis,Clostridium,Salmonella typhimirium and Listeria monocytogenes have been studied showing promising results in experimental models.However,their application in clinical trials has shown the need to maximize their therapeutic effect.Genetic engineering and synthetic biology are necessary to prove the scope that this novel approach has against cancer due to implications of cancer therapy and public health.Wheeler Torres Victor Lameda Luis Carlos Olivar Carla Navarro Jorge Fuenmayor Adrian Perez Andres Mindiola Milagros Rojas Maria Sofia Martinez Manuel Velasco Joselyn Rojas Valmore Bermudez 2018Journal of Cancer Metastasis and Treatment2018,4,1:0
