| 1 | Extracellular vesicles in the diagnosis and treatment of central nervous system diseases显示文摘Extracellular vesicles,including exosomes and microvesicles,play a fundamental role in the activity of the nervous system,participating in signal transmission between neurons and providing the interaction of central nervous system with all body systems.In many neurodegenerative diseases,neurons pack toxic substances into vesicles and release them into the extracellular space,which leads to the spread of misfolded neurotoxic proteins.The contents of neuron-derived extracellular vesicles may indicate pathological changes in the central nervous system,and the analysis of extracellular vesicle molecular content contributes to the development of non-invasive methods for the diagnosis of many central nervous system diseases.Extracellular vesicles of neuronal origin can be isolated from various biological fluids due to their ability to cross the blood-brain barrier.Today,the diagnostic potential of almost all toxic proteins involved in nervous system disease pathogenesis,specificallyα-synuclein,tau protein,superoxide dismutase 1,FUS,leucine-rich repeat kinase 2,as well as some synaptic proteins,has been well evidenced.Special attention is paid to extracellular RNAs mostly associated with extracellular vesicles,which are important in the onset and development of many neurodegenerative diseases.Depending on parental cell type,extracellular vesicles may have different therapeutic properties,including neuroprotective,regenerative,and anti-inflammatory.Due to nano size,biosafety,ability to cross the blood-brain barrier,possibility of targeted delivery and the lack of an immune response,extracellular vesicles are a promising vehicle for the delivery of therapeutic substances for the treatment of neurodegenerative diseases and drug delivery to the brain.This review describes modern approaches of diagnosis and treatment of central nervous system diseases using extracellular vesicles. | Alisa A.Shaimardanova Valeriya V.Solovyeva Daria S.Chulpanova Victoria James Kristina V.Kitaeva Albert A.Rizvanov | 2020 | Neural Regeneration Research2020,15,4: | 7 |

| 3 | Functionality of a bicistronic construction containing HEXA and HEXB genes encoding β-hexosaminidase A for cell-mediated therapy of GM2 gangliosidoses显示文摘Tay-Sachs disease and Sandhoff disease are severe hereditary neurodegenerative disorders caused by a deficiency ofβ-hexosaminidase A(HexA)enzyme,which results in the accumulation of GM2 gangliosides in the nervous system cells.In this work,we analyzed the efficacy and safety of cell-mediated gene therapy for Sandhoff disease and Sandhoff disease using a bicistronic lentiviral vector encoding cDNA of HexAα-andβ-subunit genes separated by the nucleotide sequence of a P2A peptide(HEXA-HEXB).The functionality of the bicistronic construct containing the HEXA-HEXB genetic cassette was analyzed in a culture of HEK293T cells and human umbilical cord blood mononuclear cells(hUCBMCs).Our results showed that the enzymatic activity of HexA in the conditioned medium harvested from genetically modified HEK293T-HEXA-HEXB and hUCBMCs-HEXA-HEXB was increased by 23 and 8 times,respectively,compared with the conditioned medium of native cells.Western blot analysis showed that hUCBMCs-HEXA-HEXB secreted both completely separated HEXA and HEXB proteins,and an uncleaved protein containing HEXA+HEXB linked by the P2A peptide.Intravenous injection of genetically modified hUCBMCs-HEXA-HEXB to laboratory Wistar rats was carried out,and the HexA enzymatic activity in the blood plasma of experimental animals,as well as the number of live cells of immune system organs(spleen,thymus,bone marrow,lymph nodes)were determined.A significant increase in the enzymatic activity of HexA in the blood plasma of laboratory rats on days 6 and 9(by 2.5 and 3 times,respectively)after the administration of hUCBMCsHEXA-HEXB was shown.At the same time,the number of live cells in the studied organs remained unchanged.Thus,the functionality of the bicistronic genetic construct encoding cDNA of the HEXA and HEXB genes separated by the nucleotide sequence of the P2A peptide was shown in vitro and in vivo.We hypothesize that due to the natural ability of hUCBMCs to overcome biological barriers,such a strategy can restore the activity of the missing enzyme in the central nervous system of patients with GM2 gangliosidoses.Based on the obtained data,it can be concluded that intravenous administration of hUCBMCs with HexA overexpression is a promising method of the therapy for GM2 gangliosidoses.The animal protocol was approved by the Animal Ethics Committee of the Kazan Federal University(No.23)on June 30,2020. | Alisa A.Shaimardanova Daria S.Chulpanova Valeriya V.Solovyeva Aleksandr M.Aimaletdinov Albert A.Rizvanov | 2022 | Neural Regeneration Research2022,17,1: | 3 |
| 4 | Expression of auxin synthesis gene tms1 under control of tuber-specific promoter enhances potato tuberization in vitro显示文摘Phytohormones, auxins in particular, play an important role in plant development and productivity.Earlier data showed positive impact of exogenous auxin on potato(Solanum tuberosum L.) tuberization. The aim of this study was to generate potato plants with increased auxin level predominantly in tubers. To this end, a p Bin B33-tms1 vector was constructed harboring the Agrobacterium auxin biosynthesis gene tms1 fused to tuber-speci fi c promoter of the class I patatin gene(B33-promoter) of potato. Among numerous independently generated B33:tms1 lines, those without visible differences from control were selected for detailed studies. In the majority of transgenic lines, tms1 gene transcription was detected, mostly in tubers rather than in shoots. Indoleacetic acid(IAA) content in tubers and the auxin tuber-to-shoot ratio were increased in tms1-expressing transformants. The organ-speci fi c increase in auxin synthesis in B33:tms1-transformants accelerated and intensi fi ed the process of tuber formation, reduced the dose of carbohydrate supply required for in vitro tuberRization, and decreased the photoperiodic dependence of tuber initiation. Overall, a positive correlation was observed between tms1 expression, IAA content in tubers,and stimulation of tuber formation. | 5 | Monitoring dioxins and PCBs in eggs as sensitive indicators for environmental pollution and global contaminated sites and recommendations for reducing and controlling releases and exposure显示文摘This review compiles information on PCDD/F-and PCB-contaminated eggs from 20 years of global egg monitoring around emission sources in four continents conducted by the International Pollutants Elimination Network(IPEN)and Arnika as well as a compilation of data from scientific literature.IPEN monitored 127 pooled egg samples including samples from 113 chicken flocks at potential PCDD/F-and PCB-contaminated sites around priority sources listed in the Stockholm Convention(e.g.waste incinerators,metal industries,cement plants,and open burning).99(88%)of pooled egg samples were above the EU maximum limits for PCDD/Fs(2.5 pg PCDD/F-TEQ/g fat)or the sum of PCDD/Fs and dioxinlike PCBs(5 pg PCDD/F-PCB-TEQ/g fat).Children consuming such eggs exceed the tolerable weekly intake(TWI).This demonstrates that close to 90%of these areas were not safe for the production of free-range eggs.Sixteen out of the 113 egg samples(14%)were contaminated above 50 pg TEQ/g fat and exceeded the EU maximum limit more than 10 times.From the 26 pooled egg samples around incinerators 24(92%)exceeded the limit with a mean of 43.1 pg TEQ/g fat(2.6e234 pg TEQ/g).All 21 egg samples around metal industries(4.4e112.6 pg TEQ/g fat)were above limits with mean concentration of 26.0 pg TEQ/g fat.Also all 7 egg samples measured at e-waste recycling sites were above limits(mean 308 pg TEQ/g fat).In 58(51%)pooled egg samples the PCB-TEQ was above 5 pg TEQ/g fat exceeding the EU maximum limit with dioxin-like PCBs alone.This highlights the role of commercial PCBs for global contamination with dioxin-like compounds.It was discovered that around metal industries,shredder plants,open burning sites of e-waste and dump sites,a high share of contamination was caused by dl-PCBs.This clearly shows severe PCB release from the end-of-life management of PCB-containing equipment in developing countries.Also highly contaminated eggs were found at many sites where plastic was incinerated.The highest contaminated egg sample ever measured came from an e-waste site in Ghana and had 856 pg TEQ/g fat plus 300 pg TEQ from brominated dioxins(PBDD/Fs).Other extreme PCDD/F contaminations of eggs were found at a chlor-alkali site(514 pg TEQ/g fat),Agent Orange contaminated areas in Vietnam(490,249 and 246 pg TEQ/g fat)and e-waste sites(568 and 520 pg TEQ/g fat).Where DR CALUX®bioassay revealed higher TEQ compared to measured PCDD/F-PCB-TEQ in IPEN studies,polybrominated PBDD/F were also measured and detected up to 300 pg TEQ/g fat at e-waste sites.One positive outcome from the IPEN studies is that all 10 pooled supermarket eggs in developing countries were below regulatory limit.Policy recommendations are made including:a systematic assessment of areas around PCDD/Fs and PCBs sources;measures for reduction of exposures of populations;urgent control of emission sources including PCB equipment,the open burning of plastic,and the use of plastic as fuel in boilers/incinerators in developing countries without air pollution control.Furthermore,soil limits need to be re-assessed and lowered for free-range poultry. | Jindrich Petrlik Lee Bell Joe DiGangi Serge Molly Allo'o Allo'o Gilbert Kuepouo Griffins Ochieng Ochola Valeriya Grechko Nikola Jelinek Jitka Strakova Martin Skalsky Yuyun Ismawati Drwiega Jonathan N.Hogarh Eric Akortia Sam Adu-Kumi Akarapon Teebthaisong Maria Carcamo Bjorn Beeler Peter Behnisch Claudia Baitinger Christine Herold Roland Weber | 2022 | Emerging Contaminants2022,8,1: | 2 |
