1Lifetime prediction based on Gamma processes from accelerated degradation data显示文摘Accelerated degradation test is a useful approach to predict the product lifetime at the normal use stress level, especially for highly reliable products. Two kinds of the lifetime prediction based on Gamma processes were studied. One was to predict the lifetime of the population from accelerated degradation data, and the other was to predict the lifetime of an individual by taking the accelerated degradation data as prior information. For an extensive application, the Gamma process with a time transformation and random effects was considered. A novel contribution is that a deducing method for determining the relationships between the shape and scale parameters of Gamma processes and accelerated stresses was presented. When predicting the lifetime of an individual, Bayesian inference methods were adopted to improve the prediction accuracy, in which the conjugate prior distribution and the non-conjugate prior distribution of random parameters were studied. The conjugate prior distribution only considers the random effect of the scale parameter while the non-conjugate prior distribution considers the random effects of both the scale and shape parameter. 2Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5
3Diagnostic value of negative enrichment and immune fluorescence in situ hybridization for intraperitoneal free cancer cells of gastric cancer显示文摘Objective:To explore the intraperitoneal free cancer cell(IFCC)detection value of negative enrichment and immune fluorescence in situ hybridization(NEimFISH)on chromosomes(CEN)8/17.Methods:To verify the reliability of NEimFISH,29 gastric cancer tumors,their adjacent tissues and greater omental tissues were tested.Our study then included 105 gastric cancer patients for IFCC.We defined patients as IFCC-positive if a signal was detected,regardless of the detailed cancer cell numbers.A comparison of clinicopathological features was conducted among IFCC groups.We also compared the diagnosis value and peritoneal recurrence predictive value among different detection methods.The comparison of IFCC number was also conducted among different groups.Results:A cutoff of 2.5 positive cells could distinguish all benign tissue samples and 97%of malignant tissue samples in our study.Compared to intestinal gastric cancer,patients with diffuse gastric cancer tended to have more IFCCs(6 vs.4,P=0.002).The IFCC counts were often higher in the lymphovascular invasion positive group than negative group(3 vs.1,P=0.022).All IFCC samples that were considered positive using conventional cytology were also found to be positive using NEimFISH.When compared to conventional cytology and paraffin pathology,NEimFISH had a higher IFCC positive rate(68.9%)and higher one-year peritoneal recurrence predictive value with area under the curve(AUC)of 0.922.Conclusions:Gastric cancer could be effectively diagnosed by NEimFISH.The IFCC number found using NEimFISH on CEN8/17 is closely associated with Lauren type and vascular invasion of cancer.NEimFISH is a reliable detection modality with a higher positive detection rate,higher one-year peritoneal recurrence predictive value and quantitative features for IFCC of gastric cancer.Anqiang Wang Zhongwu Li Qian Wang Yali Bai Xin Ji Tao Fu Ke Ji Yanwen Xue Tingxu Han Xiaojiang Wu Ji Zhang Yingjie Yang Guobin Xu Zhaode Bu Jiafu Ji 2019Chinese Journal of Cancer Research2019,31,6:3
4QTL-Seq Identified a Major QTL for Grain Length and Weight in Rice Using Near Isogenic F_2 Population显示文摘Mapping and isolation of quantitative trait loci(QTLs)or genes controlling grain size or weight is very important to uncover the molecular mechanisms of seed development and crop breeding.To identify the QTLs controlling grain size and weight,we developed a near isogenic line F_2(NIL-F_2)population,which was derived from a residual heterozygous plant in an F_7 generation of recombinant inbred line(RIL).With the completion of more than 30×whole genome re-sequencing of the parents,two DNA bulks for large and small grains,a total of 58.94 Gb clean nucleotide data were generated.A total of455 262 single nucleotide polymorphisms(SNPs)between the parents were identified to perform bulked QTL-seq.A candidate genomic region containing SNPs strongly associated with grain length and weight was identified from 15 to 20 Mb on chromosome 5.We designated the major QTL in the candidate region as q TGW5.3.Then,q TGW5.3 was further validated with PCR-based conventional QTL mapping method through developing simple sequence repeat and Insertion/Deletion markers in the F_2 population.Furthermore,recombinants and the progeny tests delimited the candidate region of q TGW5.3 to 1.13 Mb,flanked by HX5009(15.15 Mb)and HX5003(16.28 Mb).A set of NILs,selected from the F_2 population,was developed to evaluate the genetic effect of q TGW5.3.Significant QTL effects were detected on grain length,grain width and 1000-grain weight of H12-29 allele with 1.14 mm,-0.11 mm and 3.11 g,which explained 99.64%,95.51%and 97.32%of the phenotypic variations,respectively.QIN Yaobin CHENG Peng CHENG Yichen FENG Yue HUANG Derun HUANG Tingxu SONG Xianjun YING Jiezheng 2018Rice science2018,25,3:2

11Modeling Tracer Flow Characteristics in Different Types of Pores: Visualization and Mathematical Modeling显示文摘Structure of porous media and fluid distribution in rocks can significantly affect the transport characteristics during the process of microscale tracer flow.To clarify the effect of micro heterogeneity on aqueous tracer transport,this paper demonstrates microscopic experiments at pore level and proposes an improved mathematical model for tracer transport.The visualization results show a faster tracer movement into movable water than it into bound water,and quicker occupancy in flowing pores than in storage pores caused by the difference of tracer velocity.Moreover,the proposed mathematical model includes the effects of bound water and flowing porosity by applying interstitial flow velocity expression.The new model also distinguishes flowing and storage pores,accounting for different tracer transport mechanisms(dispersion,diffusion and adsorption)in different types of pores.The resulting analytical solution better matches with tracer production data than the standard model.The residual sum of squares(RSS)from the new model is 0.0005,which is 100 times smaller than the RSS from the standard model.The sensitivity analysis indicates that the dispersion coefficient and flowing porosity shows a negative correlation with the tracer breakthrough time and the increasing slope,whereas the superficial velocity and bound water saturation show a positive correlation.Tongjing Liu Weixia Liu Pengxiang Diwu Gaixing Hu TingXu Yuqi Li Zhenjiang You Runwei Qiao Jia Wang 2020Computer Modeling in Engineering & Sciences2020,,6:1

14Spinosin protects Neuro-2a/APP695 cells from oxidative stress damage by inactivating p38显示文摘OBJECTIVE:To explore the protective mechanism of spinosin(SPI)on Alzheimer's disease(AD)model cells,Neuro-2a/APP695(N2a/APP695),against H_(2)O_(2)-induced oxidative stress damage,to reflect the influence of oxidative stress on the development of AD,and to provide a valuable basis for the research and development of therapeutic drug for AD.METHODS:N2a/APP695 cells were exposed to H2O2 and then treated with spinosin.Firstly,the secretion level of amyloidβ(Aβ)1-42 and the production of malondialdehyde(MDA)and lactate dehydrogenase(LDH)were detected by enzyme linked immunosorbent assay kits.Secondly,the oligomerization degree of Aβ1-42 was performed by Thioflavin T staining.Thirdly,the expression levels of p-Tau(Ser199/202/396),synaptophysin(SYP),postsynaptic density protein 95(PSD95),and mitogen-activated protein kinase(MAPK)family-related proteins were detected by Western blot analysis.In addition,FITC-labeled phalloidin was used in cytoskeleton staining to reflect synaptic function.RESULTS:This study showed that H2O2 stimulated N2a/APP695 cells to produce excessive MDA and LDH and secrete a large amount of Aβ,promoted the aggregation of Aβ,induced Tau protein hyperphosphorylation,and led to synaptic dysfunction.Spinosin reversed these changes caused by H2O2 by inactivating p38,which was verified by treatment with the p38 inhibitor BIRB796.CONCLUSION:Spinosin protects N2a/APP695 cells from oxidative stress damage caused by H2O2 through inactivating p38.ZHANG Xiaoying WANG Ruixuan WANG Yiqing XU Fanxing YAN Tingxu WU Bo ZHANG Ming JIA Ying 2023Journal of Traditional Chinese Medicine2023,43,5:0

16Noradrenergic, glutamatergic and dopaminergic systems are implicated in antidepressant-like effect of flavonoids extracted from okra (Abelmoschus esculentus [L.] Moench.) fruit in the forced swimming test显示文摘Okra(Abelmoschus esculentus[L.]Moench.)is one of the most frequently used herbals in East or West Africa,and its various biological activities have been widely studied.Flavonoids extracted from many plants are reported to have neurological properties,e.g antidepressant and antifatigue.However,its neurological protect in antidepressant-like effect of flavonoids extracted from okra have not yet been demonstrated.The present study was aimed at investigating the antidepressant-like eff ect of the flavonoids extracted from okra fruit(FOF)using the forced swimming test(FST)pattern and preliminary exploration its potential mechanism.We also used the open fi eld test(OFT)to estimate the spontaneous locomotor activity.We found that oral administration(p.o.)of FOF(300 mg/kg)alone signifi cantly reduced the immobility time in the FST without changes in locomotor activity in the OPT.The experimental data indicated the antidepressant-like eff ect of FOF involved in noradrenergic,glutamatergic and dopaminergic systems.Fuyuan Li Xiaozhuo Zhang Qianqian Mao Bo Wu Ying Jia Tingxu Yan 2022Journal of Polyphenols2022,4,1:0
