| 1 | Effect of extracorporeal bioartificial liver support system on fulminant hepatic failure rabbits显示文摘AIM To evaluate the possibility of usingcultured human hepatocytes as a bridge betweenbioartificial liver and liver transplantation.METHODS In this experiment,the efficacy ofextracorporeal bioartificial liver support system(EBLSS)consisting of spheriodal human livercells and cultured hepatocytes supernatant wasassessed in vivo using galactosamine inducedrabbit model of fulminant hepatic failure.RESULTS There was no difference of survivalbetween the two groups of rabbits,but in thesupported rabbits serum alanineaminotransferase,total bilirubin and creatininewere significantly lower and hepatocyte necrosiswas markedly milder than those in controlanimals.In addition,a good viability of humanliver cells was noted after the experiment.CONCLUSION EBLSS plays a biologic role inmaintaining and compensating the function ofthe liver. | Wang YJ Li MD Wang YM Chen GZ Lu GD Tan ZX | 2000 | World Journal of Gastroenterology2000,6,2: | 17 |
| 2 | Drug resistance mechanisms and novel drug targets for tuberculosis therapy显示文摘Drug-resistant tuberculosis(TB) poses a significant challenge to the successful treatment and control of TB worldwide.Resistance to anti-TB drugs has existed since the beginning of the chemotherapy era.New insights into the resistant mechanisms of anti-TB drugs have been provided.Better understanding of drug resistance mechanisms helps in the development of new tools for the rapid diagnosis of drugresistant TB.There is also a pressing need in the development of new drugs with novel targets to improve the current treatment of TB and to prevent the emergence of drug resistance in Mycobacterium tuberculosis.This review summarizes the anti-TB drug resistance mechanisms,furnishes some possible novel drug targets in the development of new agents for TB therapy and discusses the usefulness using known targets to develop new anti-TB drugs.Whole genome sequencing is currently an advanced technology to uncover drug resistance mechanisms in M.tuberculosis.However,further research is required to unravel the significance of some newly discovered gene mutations in their contribution to drug resistance. | Md Mahmudul Islam H.M. | 3 | Unexpected complication of arteriovenous fistula of the left common carotid to internal jugular vein following central venous catheterization显示文摘Incidence of inadvertent arterial puncture secondary to central venous catheter insertion is not common with an arterial puncture rate of<1%.This is due to the advancements and wide availability of ultrasound to guide its insertion.Formation of arteriovenous fistula after arterial puncture is an unexpected complication.Till date,only five cases(including this case)of acquired arteriovenous fistula formation has been described due to inadvertent common carotid puncture.The present case is a 26-year-old man sustained traumatic brain injuries,chest injuries and multiple bony fractures.During resuscitative phase,attempts at left central venous catheter via left internal jugular vein under ultrasound guidance resulted in inadvertent puncture into the left common carotid artery.Surgical neck exploration revealed that the catheter had punctured through the left internal jugular vein into the common carotid artery with formation of arteriovenous fistula.The catheter was removed successfully and common carotid artery was repaired.Postoperatively,the patient recovered and clinic visits revealed no neurological deficits.From our literature review,the safest method for removal is via endovascular and open surgical removal.The pull/push technique(direct removal with compression)is not recommended due to the high risk for stroke,bleeding and hematoma formation. | Tan Chor Lip Henry Tan Jih Huei Mohamad Yuzaidi Lenny Suryani Safri K.Krishna Imran Alwi Rizal Md Idris Mohamad Azim Hanafiah Harunarashid | 2020 | Chinese Journal of Traumatology2020,23,1: | 5 |

Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions. | 7 | Bioengineered pseudovirus nanoparticles displaying the HA1 antigens of influenza viruses for enhanced immunogenicity显示文摘Even with implementation of current influenza vaccines,influenza still claims up to 500,000 lives worldwide annually,indicating a need for a better vaccine strategy.We have developed a technology to generate unique S_(60)-HA1 pseudovirus nanoparticles(PVNPs)that display the receptor-binding HA1 domains of influenza viruses.Each self-assembled S_(60)-HA1 PVNP consists of a T=1 icosahedral S_(60) nanoparticle that resembles the inner shell of norovirus capsid and 60 surface-displayed HA1 antigens that are excellent vaccine targets.Soluble S_(60)-HA1 PVNPs presenting HA1 antigens of H7N9 influenza virus subtypes have been produced efficiently in large amount.Their three-dimensional(3D)structures have been solved by cryogenic electron microscopy.The PVNP-displayed HA1 antigens react with HA-specific antibody,and retain authentic sialic acid binding specificity and hemagglutinate human erythrocytes.The PVNPs are highly immunogenic,eliciting high titers of HA1-specific antibodies in mice and the mouse sera strongly inhibited hemagglutinations of homologous and heterologous influenza virus HA proteins.Therefore,the S_(60)-HA1 PVNPs may provide useful reagents to study influenza viruses and offer a potential new vaccine tactic to fight the deadly influenza disease. | Ming Xia Md Rejaul Hoq Pengwei Huang Wen Jiang Xi Jiang Ming Tan | 2022 | Nano Research2022,15,5: | 2 |
