| 3 | Recombinant human fibroblast growth factor-2 promotes nerve regeneration and functional recovery after mental nerve crush injury显示文摘Several studies have shown that fibroblast growth factor-2(FGF2) can directly affect axon regeneration after peripheral nerve damage. In this study, we performed sensory tests and histological analyses to study the effect of recombinant human FGF-2(rh FGF2) treatment on damaged mental nerves. The mental nerves of 6-week-old male Sprague-Dawley rats were crush-injured for 1 minute and then treated with 10 or 50 μg/m L rh FGF2 or PBS in crush injury area with a mini Osmotic pump. Sensory test using von Frey filaments at 1 week revealed the presence of sensory degeneration based on decreased gap score and increased difference score. However, at 2 weeks, the gap score and difference score were significantly rebounded in the mental nerve crush group treated with 10 μg/m L rh FGF2. Interestingly, treatment with 10 μg/m L rh FGF had a more obviously positive effect on the gap score than treatment with 50 μg/m L rh FGF2. In addition, retrograde neuronal tracing with Dil revealed a significant increase in nerve regeneration in the trigeminal ganglion at 2 and 4 weeks in the rh FGF2 groups(10 μg/m L and 50 μg/m L) than in the PBS group. The 10 μg/m L rh FGF2 group also showed an obviously robust regeneration in axon density in the mental nerve at 4 weeks. | 4 | Downregulated miR-18b-5p triggers apoptosis by inhibition of calcium signaling and neuronal cell differentiation in transgenic SOD1(G93A)mice and SOD1(G17S and G86S)ALS patients显示文摘Background:MicroRNAs(miRNAs)are endogenous non-coding RNAS that regulate gene expression at the post-transcriptional level and are key modulators in neurodegenerative diseases.Overexpressed miRNAs play an important role in amyotrophic lateral sclerosis(ALS);however,the pathogenic mechanisms of deregulated miRNAS are still unclear.Methods:We aimed to assess the dysfunction of RNAS or miRNAs in fALS(SOD1 mutations).We compared the RNA-seq of subcellular fractions in NSC-34 WT(hSOD1)and MT(hSOD1(G93A))cells to find altered RNAs or miRNAs.We identified that Hif1a and Mef2c were upregulated,and Mctp1 and Rarb were downregulated in the cytoplasm of NSC-34 MT cells.Results:SOD1 mutations decreased the level of miR-18b-5p.Induced Hif1a which is the target for miR-18b increased Mef2c expression as a transcription factor.Mef2c upregulated miR-206 as a transcription factor.Inhibition of Mctp1 and Rarb,which are targets of miR-206,induced intracellular Ca^2+ levels and reduced cell differentiation,respectively.The miR-18b-5p pathway was also observed in G93A Tg mice,fALS(G86S)patient,and iPSC-derived motor neurons from fALS(G17S)patient.Conclusions:Our data indicate that SOD1 mutation decreases miR-18b-5p,which sequentially regulates Hif1a,Mef2c,miR-206,Mctp1 and Rarb in fALS-linked SOD1 mutation.These results provide new insights into the downregulation of miR-18b-5p-dependent pathogenic mechanisms of ALS. | Ki Yoon Kim Yu Ri Kim Kyung Won Choi Mijung Lee Somyung Lee Wooseok Im Je-Young Shin Jin Young Kim Yoon Ho Hong Manho Kim Jong-ll Kim Jung-Joon Sung | 2020 | Translational Neurodegeneration2020,9,3: | 2 |

| 13 | Entacapone promotes hippocampal neurogenesis in mice显示文摘Entacapone,a catechol-O-methyltransferase inhibitor,can strengthen the therapeutic effects of levodopa on the treatment of Parkinson’s disease.However,few studies are reported on whether entacapone can affect hippocampal neurogenesis in mice.To investigate the effects of entacapone,a modulator of dopamine,on proliferating cells and immature neurons in the mouse hippocampal dentate gyrus,60 mice(7 weeks old)were randomly divided into a vehicle-treated group and the groups treated with 10,50,or 200 mg/kg entacapone.The results showed that 50 and 200 mg/kg entacapone increased the exploration time for novel object recognition.Immunohistochemical staining results revealed that after entacapone treatment,the numbers of Ki67-positive proliferating cells,doublecortin-positive immature neurons,and phosphorylated cAMP response element-binding protein(pCREB)-positive cells were significantly increased.Western blot analysis results revealed that treatment with tyrosine kinase receptor B(TrkB)receptor antagonist significantly decreased the exploration time for novel object recognition and inhibited the expression of phosphorylated TrkB and brain-derived neurotrophic factor(BDNF).Entacapone treatment antagonized the effects of TrkB receptor antagonist.These results suggest that entacapone treatment promoted hippocampal neurogenesis and improved memory function through activating the BDNF-TrkB-pCREB pathway.This study was approved by the Institutional Animal Care and Use Committee of Seoul National University(approval No.SNU-130730-1)on February 24,2014. | Dae Young Yoo Hyo Young Jung Woosuk Kim Kyu Ri Hahn Hyun Jung Kwon Sung Min Nam Jin Young Chung Yeo Sung Yoon Dae Won Kim In Koo Hwang | 2021 | Neural Regeneration Research2021,16,6: | 1 |
| 14 | Lichen Planus Pigmentosus Inversus:Two Case Reports显示文摘Introduction:Lichen planus pigmentosus inversus(LPPI)is a rare a rare variant of lichen planus characterized by hyperpigmented patches with predominating localization in intertriginous areas.Due to its rarity,only a few LPPI cases are reported.We herein describe two rare cases of LPPI.Case presentation:The two patients were all with a brownish macular lesion on the intertriginous area.A diagnosis of LPPI was made based on their clinical manifestations,dermoscopic features,and histopathologic features,which revealed an interface change,lichenoid infiltration,and pigmentary incontinence.Discussion:LPPI is pruritic or asymptomatic,hyperpigmented macules and patches on the flexural folds.The axillae and flanks were the most commonly affected areas,followed by the groin and genitalia.About half of the female patients had inframammary fold lesions.LPPI shows higher female predominance than Lichen planus pigmentosus.There were fewer cases that lasted more than 3 years compared to LPP.Conclusion:LPPI is a rare variant of lichen planus,with a locational characteristic and female predominance.Therefore,in the case of a pigmented disease occurring in the flexural folds,it should be placed in the differential diagnosis. | Se Hoon Lee Minah Cho Yu Jin Han Yu Ri Woo Sang Hyun Cho Jeong Deuk Lee Hei Sung Kim | 2022 | International Journal of Dermatology and Venereology2022,5,2: | 0 |
