9高脂饮食大鼠饲粮中添加西番莲皮可提高胰岛素的敏感性,增加肠促胰素和下丘脑致饱食感显示文摘试验旨在探索斯普拉-道来(SD)大鼠高脂饲粮(HF)中添加西番莲果皮粉(PEPF)对大鼠下丘脑m RNA表达、胰岛素敏感性和其他代谢参数的影响。SD大鼠随机分为3组:对照组饲喂脂肪含量正常的饲粮;HF组饲喂高脂饲粮(脂肪含量35%);HFPF组饲喂添加PEPF的高脂饲粮(脂肪含量35%)。HFPF组和HF组大鼠在试验开始前进行为期4周的高脂饲喂,诱导肥胖引起代谢异常之后,HFPF组开始饲喂添加PEPF的高脂饲粮,为期6周。对照组和HF组分别保持正常的饲粮和高脂饲粮直至试验结束。试验结束后,通过糖耐量测试(GTT)和胰岛素耐量测试(ITT)评估糖耐量和胰岛素敏感性,免疫测定法检测胃肠激素和脂肪因子,实时荧光定量聚合酶链反应评估下丘脑神经肽的表达。ITT测试中,PEPF可显著增加大鼠下丘脑可卡因和安非他明调节转录肽(CART)的表达(P<0.05),极显著降低体增重(P<0.01),显著减轻肥胖症状(P<0.05)和血清瘦素水平(P<0.05);极显著增加脂联素含量(P<0.01)和葡萄糖依赖性促胰岛素多肽(P<0.01);随着葡萄糖利用率极显著增加(P<0.01),胰高血糖素样肽-1(GLP-1)极显著的改善胰岛素敏感性(P<0.01)。其他胃肠激素(酪酪肽、胰多肽和胰淀素)和白介素(IL-6、肿瘤坏死因子-α、IL-1β和单核细胞趋化蛋白-1)均无显著变化。试验结果为探究PEPF在改善血糖稳态、增强饱腹感的作用和机制方面提供了进一步的试验依据。Lima G C Vuolo M M Batista A G Dragano N R V Solon C Junior M R M 2016饲料博览2016,0,9:1
