1Roles of the MEK1/2 and AKT pathways in CXCL12/CXCR4 induced cholangiocarcinoma cell invasion显示文摘AIM:To evaluate the expression of C-X-C motif chemokine receptor 4(CXCR4)and its signaling cascades,which were previously identified as a key factor for cancer cell progression and metastasis,in cholangiocarcinoma cell lines.METHODS:The expression of CXCR4 and its signaling cascades were determined in the cholangiocarcinoma cell lines(RMCCA1 and KKU100)by Western blotting.The invasion assays and the detection of actin polymerization were tested in these cholangiocarcinoma cells treated with CXC chemokine ligand-12(CXCL12).RESULTS:Expression of CXCR4 was detected in both cholangiocarcinoma cell lines and activation of CXCR4 with CXCL12 triggered the signaling via the extracellular signal-regulated kinase-1/2(ERK1/2)and phosphoinositide 3-kinase(PI3K)and induction of cholangiocarcinoma cell invasion,and displayed high levels of actin polymerization.Addition of CXCR4 inhibitor(AMD3100)abrogated CXCL12-induced phosphorylation of MEK1/2 and Akt in these cells.Moreover,treatment with MEK1/2 inhibitor(U0126)or PI3K inhibitor(LY294 002)also attenuated the effect of CXCL12-induced cholangiocarcinoma cell invasion.CONCLUSION:These results indicated that the activation of CXCR4 and its signaling pathways(MEK1/2 and Akt)are essential for CXCL12-induced cholangiocarcinoma cell invasion.This rises Implications on a potential role for the inhibition of CXCR4 or its signal cascades in the treatment of cholangiocarcinoma.Kawin Leelawat Surang Leelawat Siriluck Narong Suradej Hongeng 2007World Journal of Gastroenterology2007,13,10:27
2Prospective study of MMP7 serum levels in the diagnosis of cholangiocarcinoma显示文摘AIM: To determine whether the serum level of matrix metalloproteinase-7 (MMP7) has the potential to diagnosis cholangiocarcinoma from benign biliary tract diseases. METHODS: This study was performed according to the PRoBE (a prospective-specimen-collection, retrospectiveblinded-evaluation) design. A total of 187 patients with obstructive jaundice were consecutively enrolled. After the diagnostic status of these patients was ascertained, their levels of serum MMP7 were assayed and compared with serum carbohydrate antigen 19-9 (CA19-9). This was conducted in a blinded case (cholangiocarcinoma)control (benign biliary tract disease) setup. RESULTS: MMP7 and CA19-9 serum levels were significantly elevated in cholangiocarcinoma patients (P < 0.001). The area under the curve (AUC) from a receiver operating characteristic (ROC) curve analysis for thediagnosis of cholangiocarcinoma, using MMP7 was more accurate than CA19-9 (AUC = 0.84, 95% CI: 0.778-0.903 for MMP7 and AUC = 0.79, 95% CI: 0.708-0.868 for CA19-9). The sensitivity and specificity of serum MMP7 (cut-off value of 5.5 ng/mL) was 75% and 78%, respectively, while the sensitivity and specificity of serum CA19-9 (cut-off value of 100 U/mL) was 68% and 87%, respectively. 3Prognostic relevance of circulating CK19 mRNA in advanced malignant biliary tract diseases显示文摘AIM: To determine the role of circulating tumor cells (CTCs) in prediction of the overall survival of patients with advanced malignant biliary tract obstruction. METHODS: We investigated the prognostic value of CTCs by examining two markers, cytokeratin (CK) 19 and human telomerase reverse transcriptase (hTERT) mRNA, in 40 patients diagnosed with advanced malig- nant biliary tract diseases. Quantitative real-time re- verse transcription polymerase chain reaction was used to detect CK19 and hTERT mRNA in the peripheral blood of these patients. Overall survival was analyzed using the Kaplan-Meier method and Cox regression modeling.RESULTS: Positive CK19 and hTERT mRNA expression was detected in 45% and 60%, respectively, of the 40 patients. Univariable analysis indicated that positive CK19 mRNA expression was significantly associated with worse overall survival (P = 0.009). Multivariable analysis determined that positive CK19 mRNA expres- sion, patient's age and serum bilirubin were each inde- pendently associated with overall survival. 4Conversion enhancement of tubular fixed-bed reactor for Fischer-Tropsch synthesis using static mixer显示文摘Recently,Fischer-Tropsch synthesis(FTS) has become an interesting technology because of its potential role in producing biofuels via Biomassto-Liquids(BTL) processes.In Fischer-Tropsch(FT) section,biomass-derived syngas,mainly composed of a mixture of carbon monoxide(CO) and hydrogen(H2),is converted into various forms of hydrocarbon products over a catalyst at specified temperature and pressure.Fixed-bed reactors are typically used for these processes as conventional FT reactors.The fixed-bed or packed-bed type reactor has its drawbacks,which are heat transfer limitation,i.e.a hot spot problem involved highly exothermic characteristics of FT reaction,and mass transfer limitation due to the condensation of liquid hydrocarbon products occurred on catalyst surface.This work is initiated to develop a new chemical reactor design in which a better distribution of gaseous reactants and hydrocarbon products could be achieved,and led to higher throughput and conversion.The main goal of the research is the enhancement of a fixed-bed reactor,focusing on the application of KenicsTM static mixer insertion in the tubular packed-bed reactor.Two FTS experiments were carried out using two reactors i.e.,with and without static mixer insertion within catalytic beds.The modeled syngas used was a mixed gas composed of H2/CO in 2:1 molar ratio that was fed at the rate of 30mL(STP)min1(GHSV≈136mL g1 cat h1) into the fixed Ru supported aluminum catalyst bed of weight 13.3g.The reaction was carried out at 180℃ and atmospheric pressure continuously for 36h for both experiments.Both transient and steady-state conversions(in terms of time on stream) were reported.The results revealed that the steady-state CO conversion for the case using the static mixer was approximately 3.5 times higher than that of the case without static mixer.In both cases,the values of chain growth probability of hydrocarbon products(α) for Fischer-Tropsch synthesis were 0.92 and 0.89 for the case with and without static mixer,respectively.Phavanee Narataruksa Sabaithip Tungkamani Karn Pana-Suppamassadu Phongsak Keeratiwintakorn Siriluck Nivitchanyong Piyapong Hunpinyo Hussanai Sukkathanyawat Prayut Jiamrittiwong Visarut Nopparat 2012Journal of Natural Gas Chemistry2012,21,4:2

15The influence of environmental factors on species composition and distribution in a community forest in Northern Thailand显示文摘Understanding the environmental factors that influence tree species composition is essential for successful management of biodiversity and sustainable use of community forest resources.This study aims to assess tree species composition and distribution in the deciduous Ban Mae Chiang Rai Lum Community Forest in Northern Thailand and to analyze the influence of environmental factors on tree biodiversity in the forest.We conducted a stratified systematic sampling of the forest’s total area of 3925 ha,and twenty-five 0.16 ha survey plots were established in three different stands of the deciduous forests to estimate and characterize the difference in biological diversity among the stands.Canonical correspondence analysis(CCA)was used to investigate the environment factors affecting such differences in biodiversity of the stands.The results showed a high diversity of trees in the forest as 197 species,144 genera,and 62 plant families were recorded.The CCA ordination identified the environmental factors—the most important of which were elevation,distance to streams,soil moisture,organic matter,and distance to communities—that signifi-cantly influenced the diversity and distribution of tree species(p<0.05)in the community forest.Our findings indicate that the implementation of drought reduction measures such as building check dams,fire protection,and monitoring community forest-product usage would be recommended to further biodiversity conservation and the sustainable use of community forest resources.Siriluck Thammanu Dokrak Marod Hee Han Narayan Bhusal Lamthai Asanok Pipat Ketdee Noppakoon Gaewsingha Seunghyun Lee Joosang Chung 2021Journal of Forestry Research2021,32,2:1

17Strong expression of CD133 is associated with increased cholangiocarcinoma progression显示文摘AIM:To determine the role of CD133 in cholangiocarcinoma progression. METHODS:CD133 protein expression was evaluated by immunohistochemistry in 34 cholangiocarcinoma specimens.In addition,proliferation,chemoresistance and invasive properties of CD133-enriched(CD133 + ) and CD133-depleted(CD133 )RMCCA1 cholangiocarcinoma cells were studied and compared. RESULTS:Strong CD133 expression was observed in 67.6%(23/34)of the cholangiocarcinoma specimens. Strong expression of CD133 was significantly associated with nodal metastasis(P=0.009)and positive surgical margin status(P=0.011).In the in vitro study, both the CD133 + and CD133 cells had similar proliferation abilities and resistance to chemotherapeutic drugs.However,the CD133 + cells had a higher invasive ability compared with CD133 cells. 20rePROBE:Workflow for Revised Probe Assignment and Updated Probe-set Annotation in Microarrays显示文摘Commercial and customized microarrays are valuable tools for the analysis of holistic expression patterns,but require the integration of the latest genomic information.This study provides a comprehensive workflow implemented in an R package(rePROBE)to assign the entire probes and to annotate the probe sets based on up-to-date genomic and transcriptomic information.The rePROBE package can be applied to available gene expression microarray platforms and addresses both public and custom databases.The revised probe assignment and updated probe-set annotation are applied to commercial microarrays available for different livestock species,i.e.,chicken(Gallus gallus;ChiGene-1_0-st:443,579 probes and 18,530 probe sets),pig(Sus scrofa;PorGene-1_1-st:592,005 probes and 25,779 probe sets),and cattle(Bos Taurus;BovGene-1_0-st:530,717 probes and 24,759 probe sets),as well as available for human(Homo sapiens;HuGene-1_0-st)and mouse(Mus musculus HT_MG-430_PM).Using current species-specific transcriptomic information(RefSeq,Ensembl,and partially non-redundant nucleotide sequences)and genomic information,the applied workflow reveals 297,574 probes(15,689 probe sets)for chicken,384,715 probes(21,673 probe sets)for pig,363,077 probes(21,238 probe sets)for cattle,481,168 probes(23,495 probe sets)for human,and 324,942 probes(32,494 probe sets)for mouse.These are representative of 12,641,15,758,18,046,20,167,and 16,335 unique genes that are both annotated and positioned for chicken,pig,cattle,human,and mouse,respectively.Additionally,the workflow collects information on the number of single nucleotide polymorphisms(SNPs)within respective targeted genomic regions and thus provides a detailed basis for comprehensive analyses such as expression quantitative trait locus(eQTL)studies to identify quantitative and functional traits.The rePROBE R package is freely available at http://gffzz188fe103f8f1460aspxocnq0fwq0o6c9w.ffgz.tsg.suse.edu.cn/friederhadlich/rePROBE.Frieder Hadlich Henry Reyer Michael Oster Nares Trakooljul Eduard Muráni Siriluck Ponsuksili Klaus Wimmers 2021Genomics, Proteomics & Bioinformatics2021,19,6:0
