| 3 | Hypocrellin A-based photodynamic action induces apoptosis in A549 cells through ROS-mediated mitochondrial signaling pathway显示文摘Over recent decades, many studies have reported that hypocrellin A(HA) can eliminate cancer cells with proper irradiation in several cancer cell lines. However, the precise molecular mechanism underlying its anticancer effect has not been fully defined. HA-mediated cytotoxicity and apoptosis in human lung adenocarcinoma A549 cells were evaluated after photodynamic therapy(PDT). A temporal quantitative proteomics approach by isobaric tag for relative and absolute quantitation(iTRAQ) 2 D liquid chromatography with tandem mass spectrometric(LC–MS/MS) was introduced to help clarify molecular cytotoxic mechanisms and identify candidate targets of HA-induced apoptotic cell death. Specific caspaseinhibitors were used to further elucidate the molecular pathway underlying apoptosis in PDT-treated A549 cells. Finally, down-stream apoptosis-related protein was evaluated. Apoptosis induced by HA was associated with cell shrinkage, externalization of cell membrane phosphatidylserine, DNA fragmentation,and mitochondrial disruption, which were preceded by increased intracellular reactive oxygen species(ROS) generations. | 7 | Thermodynamic Modeling of Fluid-Bearing Natural Gas Inclusions for Geothermometer and Geobarometer of Overpressured Environments in Qiongdongnan Basin, South China Sea显示文摘It is a very difficult problem to directly determine fluid pressure during hydrocarbon migration and accumulation in sedimentary basins. pVt modeling of coupling hydrocarbon fluid inclusion of its coeval aqueous fluid inclusion provides a powerful tool for establishing the relationship of formation pressure evolution with time. Homogenization temperature of fluid inclusion can routinely be measured under microthermometric microscopy. Crushing technique has been employed to obtain the composition of fluid inclusions, and the commercial software VTFLINC easily and rapidly completes the construction of p t phase diagram. The minimum trapping pressure of hydrocarbon fluid inclusion would be then determined in the p t space. In this paper, three samples of YC21 1 1 and YC21 1 4 wells at YC21 1 structural closure, Qiongdongnan basin, South China Sea, were selected for the pVt modeling practice, and the formation pressure coefficient (equals to fluid pressure/hydrostatic pressure) changing trend with time has primarily been established. | 8 | Effects of Sini San used alone and in combination with fluoxetine on central and peripheral 5-HT levels in a rat model of depression显示文摘OBJECTIVE: To investigate the effects of Sini San and fluoxetine on the levels of central and peripheral 5-HT in a rat model of depression, and provide new insight into the treatment of depression with integrated Chinese-Western Medicine.METHODS: A rat model of depression was established by chronic mild stress(CMS). Model rats received either Sini San, fluoxetine, a combination of the two drugs, or no drug treatment. Healthy naive rats were used as controls. Open field and sucrose preference tests were used to assess depression-like behavior. ELISA and immunohistochemistry were used to determine central and peripheral levels of 5-HT.RESULTS: In the group with no drug treatment,central 5-HT expression decreased while peripheral5-HT concentrations increased as CMS continued.Four weeks after CMS, Sini San alone was less effective in reducing depression-like behavior than fluoxetine alone or in combination with Sini San,but combined use was more effective than fluoxetine alone. Eight weeks after CMS, Sini San alone or in combination with fluoxetine was more effective in reducing depression-like behavior than fluoxetine alone. Furthermore Sini San and fluoxetine used alone or in combination notably increased central 5-HT expression and decreased peripheral 5-HT levels in the rat model.CONCLUSION:The results of the present study indicate that there is a synergistic action between the two medicines in the treatment of depression. Sini San exhibited a relatively long lag before its effects were observed; however, by eight weeks the Traditional Chinese Medicine appeared at least as effective as fluoxetine. | 9 | Spatial and temporal variability of heat content above the thermocline in the tropical Pacific Ocean显示文摘Heat content of the upper layer above the 20℃ isotherm in the tropical Pacific Ocean is estimated by using the sea temperature data set with a resolution 2°latitude × 5°longitude (1980~1993) for the water depths (every 10 m) from 0 m to 400 m, and its temporal and spatial variabilities are analyzed. ( 1 ) The temporal variability indicates that the total heat in the upper layer of the equatorial Pacific Ocean is characterized by the interannual variability. The time series of the equatorial heat anomaly 5 months lead that of the El Nino index at the best positive lag correlation between the two, and the former 13 months lag behind the latter at their best negative lag correlation. Therefore the equatorial heat content can be used as a better predictor than the El Nino index for a warm or cold event. In addition, it is also found that less heat anomaly in the equator corresponds to the stronger warm events in the period (1980~1993) and much more heat was accumulated in the 4 years including 1992/1993 ENSO (1989~1993) than the 4 years including 1982/1983 ENSO (1980~1983); (2) The spatial variability indicates that the area with the highest lag correlation among the grids moves in an anti-clockwise circle in the northern tropical Pacific Ocean within 4 years period and in a clockwise circle in the southern tropical Pacific Ocean. | 10 | Effects of purified zearalenone on selected immunological and histopathologic measurements of spleen in post-weanling gilts显示文摘The present study was aimed at investigating the adverse effects of dietary zearalenone(ZEA) on the lymphocyte proliferation rate(LPR), interleukin-2(IL-2), mRNA expressions of pro-inflammatory cytokines, and histopathologic changes of spleen in post-weanling gilts. A total of 20 crossbred piglets(Yorkshire × Landrace × Duroc) with an initial BW of 10.36 ± 1.21 kg(21 d of age) were used in the study.Piglets were fed a basal diet with an addition of 0.1.1,2.0, or 3.2 mg/kg purified ZEA for 18 d ad libitum.The results showed that LPR and IL-2 production of spleen decreased linearly(P < 0.05) as dietary ZEA increased. Splenic mRNA expressions of interleukin-1β(IL-1β) and interleukin-6(IL-6) were linearly upregulated(P < 0.05) as dietary ZEA increased. On the contrary, linear down-regulation(P<0.05) of mRNA expression of interferon-γ(IFN-γ) was observed as dietary ZEA increased. Swelling splenocyte in1.1 mg/kg ZEA treatments, atrophy of white pulp and swelling of red pulp in 2.0 and 3.2 mg/kg ZEA treatments were observed. | 13 | Prognostic significance of combining high mobility group Box-1 and OV-6 expression in hepatocellular carcinoma显示文摘The inflammatory environment and existence of cancer stem cells are critical for progression and intrahepatic recurrence of hepatocellular carcinoma(HCC) after curative resections. Here, we investigated the prognostic significance of combining high mobility group box 1(HMGB1) expression and hepatic progenitor marker OV6 in hepatocellular carcinoma. Expression of HMGB1 and OV6 was evaluated using immunohistochemistry profiling in tissue microarrays containing samples from 208 HCC patients. Invasive clinical or pathological factors were found in patients with high expression of HMGB1 or OV6. Higher HMGB1 was associated with poorer clinical outcomes, and independently related to elevated 5-year recurrence incidence(85.5% vs. 62.4%, P<0.001). We also found that more OV6 positive staining was correlated with poor prognosis of HCC patients(P<0.001). Notably, expression of HMGB1 was positively correlated with OV6 in density(R2=0.032, P<0.001) and reversely related to HCC outcomes. Abnormal expression of HMGB1 in combination with positive staining of OV6 displayed poorer prognostic performance than single biomarker alone(area under curve(AUC) survival=0.696). | 16 | Magnetostratigraphy and provenance of the Qingzhou loess in Shandong province显示文摘Loess deposits with varying thickness are widely distributed on the intermontane valleys and piedmont zones on the northern side of the central Shandong mountainous regions. However, the basal ages and material resources of the loess deposits are not clear. The paper studied the Qingzhou loess profile in Shandong with magnetostratigraphic and optical stimulated luminescence (OSL) methods and further investigated its main provenances with the mineralogical methods. The magnetostratigraphic results showed that the Brunhes/Matuyama (B/M) reversal boundary was not recognized, suggesting a basal age younger than 0.78 Ma. Extrapolations by sedimentation rates, based on the upper part depositional rate from the OSL age, the basal age of the Qingzhou loess is about 0.5 Ma. Until now, older loess deposits have not been reported on the northern side of the central Shandong mountainous regions. The results of the paper indicate that the loess deposits in this area might have strated from the Middle Pleistocene. The basal age of Qingzhou loess is approximately synchronous with the Xiashu loess in the middle-lower reaches of Yangtze River. Major components of clay minerals in the Qingzhou profile are dominated by illite. Other clay mineral compositions are mainly smectite, chlorite and kaolinite, which are similar with the Xifeng loess in the Loess Plateau. However, the contents of smectite and the ratios of illite and kaolinte in the Qingzhou loess samples are higher than those in the Xifeng loess samples of the Loess Plateau, indicating that the loess in the northern side of the central Shandong mountainous regions has different sources from that of the loess deposits in the Loess Plateau. The clay mineral analysis further reinforces the earlier conclu- sion that the marine strata exposed in the Laizhou Bay and the fluvial plain of the lower reaches of Yellow River during the glacier periods are the main material sources for the Qingzhou loess deposits, which is an indicator to the local aridification of the lower reaches of the Yellow River. Loess deposition in the central Shandong mountainous regions started at around 0.5 Ma. The age of Qingzhou loess is approximately synchronous with the ongoing high-latitude cold since the Middle Pleistocene, which indicates that strengthened East Asian winter monsoon was sufficiently energetic to bring substantial quantities of material from the marine strata exposed in the Laizhou Bay and the fluvial plain of the lower reaches of the Yellow River to the central Shandong mountainous regions. The porous silicon layers (PSL) were created by anodizing the wafers with a denuded zone of 20-40 μm formed by neutron transmutation doping (NTD) and thermal treatment at 940℃ for 4 h and then 700℃ for 2 h, two-step heating of the floating-zone silicon (FZ Si) grown in a hydrogen (H2) ambience. By surface modification with stannic chloride or amine immersion and rapid thermal oxidation (RTO), the PL peak position from the PS can be qualitatively controlled factitiously. The as-prepared PS shows an orange-yellow luminescence, while the modified samples emit red, green and blue luminescence. Mechanisms for the different colors of the PL are discussed. | 18 | Computer Simulation of Mechanical Properties of Nano-scale Cu/FeS Composite显示文摘The mechanical properties of nano-scale Cu/FeS composite were simulated by molecular dynamics (MD) simulation in this paper. Through the analysis on the stress-strain curves, the results of MD simulation were in good agreement with mechanisms of macroscopic deformation. When the size of particles was smaller than a certain value, the relationship between yield strength and size, which was different from the large size crystals abided by the contrary Hall-Petch relationship. Based on the discussion of nano-scale Cu/FeS composite, some interesting conclusions were obtained. For example, the 'S' type curves were discovered in stress-strain curves and the anisotropy of FeS was very evident when the exposures of reinforcing phase (FeS) were different and so on. The basic theories and calculations of the composite that contains nano-scale particles were discussed. | 19 | IAP's Solar-Powered Unmanned Surface Vehicle Actively Passes through the Center of typhoon Sinlaku(2020)显示文摘The solar-powered marine unmanned surface vehicle(USV) developed by the USV team of the Institute of Atmospheric Physics is a rugged, long-duration, and autonomous navigation vessel designed for the collection of longrange, continuous, real-time, meteorological and oceanographic measurements, especially under extreme sea conditions(sea state 6–7). These solar-powered USVs completed a long-term continuous navigation observation test over 26 days.During this time, they coordinated double-USV observations and actively navigated into the path of Typhoon Sinlaku(2020) before collecting data very close to its center during the 2020 USV South China Sea Typhoon Observation Experiment. Detailed high temporal resolution(1 min) real-time observations collected by the USV on the typhoon were used for operational typhoon forecasting and warning for the first time.
