1The Ghd7 transcription factor represses ARE1 expression to enhance nitrogen utilization and grain yield in rice显示文摘The genetic improvement of nitrogen use efficiency(NUE)of crops is vital for grain productivity and sustainable agriculture.However,the regulatory mechanism of NUE remains largely elusive.Here,we report that the rice Grain number,plant height,and heading date7(Ghd7)gene genetically acts upstream of ABC1 REPRESSOR1(ARE1),a negative regulator of NUE,to positively regulate nitrogen utilization.As a transcriptional repressor,Ghd7 directly binds to two Evening Element-like motifs in the promoter and intron 1 of ARE1,likely in a cooperative manner,to repress its expression.Ghd7 and ARE1 display diurnal expression patterns in an inverse oscillation manner,mirroring a regulatory scheme based on these two loci.Analysis of a panel of 2656 rice varieties suggests that the elite alleles of Ghd7 and ARE1 have undergone diversifying selection during breeding.Moreover,the allelic distribution of Ghd7 and ARE1 is associated with the soil nitrogen deposition rate in East Asia and South Asia.Remarkably,the combination of the Ghd7 and ARE1 elite alleles substantially improves NUE and yield performance under nitrogen-limiting conditions.Collectively,these results define a Ghd7–ARE1-based regulatory mechanism of nitrogen utilization,providing useful targets for genetic improvement of rice NUE.Qing Wang Qingmei Su Jinqiang Nian Jian Zhang Meng Guo Guojun Dong Jiang Hu Rongsheng Wang Changshuo Wei Guanwen Li Wan Wang Hui-Shan Guo Shaoyang Lin Wenfeng Qian XianzhiXie Qian Qian Fan Chen Jianru Zuo 2021Molecular Plant2021,14,6:10
2Expression and function of Delta-like ligand 4 in a rat model of retinopathy of prematurity显示文摘The Delta-like ligand 4/Notch signaling pathway was shown to participate in the process of retinal development and angiogenesis.However,the function of the Delta-like ligand 4/Notch signaling pathway in retinopathy of prematurity requires further study.Retinopathy of prematurity was induced in 5-day-old Sprague-Dawley rats exposed to hyperoxia for 7 days,and then returned to room air.Reverse transcription-PCR and western blot revealed that Delta-like ligand 4 levels decreased at postnatal day 12 and increased at postnatal day 17 in retinopathy of prematurity rats.Flat-mounted adenosine diphosphatase stained retina and hematoxylin-eosin stained retinal tissue slices showed that the clock hour scores and the nuclei counts in retinopathy of prematurity rats were significantly different compared to normal control rats.After retinopathy of prematurity rats were intravitreally injected with Delta-like ligand 4 monoclonal antibody to inhibit the Delta-like ligand 4/Notch signaling pathway,there was a significant increase in the severity of retinal neovascularization (clock hours) in the intravitreally injected eyes.The nuclei count was highly correlated with the clock hour score.These results suggest that Delta-like ligand 4/Notch signaling plays an essential role in the process of physiological and pathological angiogenesis in the retina.Shaoyang Shi Xun Li You Li Cunwen Pei Hongwei Yang Xiaolong Chen 2013Neural Regeneration Research2013,8,8:4
3Enhancing rice grain production by manipulating the naturally evolved cis-regulatory element-containing inverted repeat sequence of OsREM20显示文摘Grain number per panicle(GNP)is an important agronomic trait that contributes to rice grain yield.Despite its importance in rice breeding,the molecular mechanism underlying GNP regulation remains largely unknown.In this study,we identified a previously unrecognized regulatory gene that controls GNP in rice,Oryza sativa REPRODUCTIVE MERISTEM 20(OsREM20),which encodes a B3 domain transcription factor.Through genetic analysis and transgenic validation we found that genetic variation in the CArG box-containing inverted repeat(IR)sequence of the OsREM20 promoter alters its expression level and contributes to GNP variation among rice varieties.Furthermore,we revealed that the IR sequence regulates OsREM20 expression by affecting the direct binding of OsMADS34 to the CArG box within the IR sequence.Interestingly,the divergent pOsREM20IR and pOsREM20ΔIR alleles were found to originate from different Oryza rufipogon accessions,and were independently inherited into the japonica and indica subspecies,respectively,during domestication.Importantly,we demonstrated that IR sequence variations in the OsREM20 promoter can be utilized for germplasm improvement through either genome editing or traditional breeding.Taken together,our study characterizes novel genetic variations responsible for GNP diversity in rice,reveals the underlying molecular mechanism in the regulation of agronomically important gene expression,and provides a promising strategy for improving rice production by manipulating the cis-regulatory element-containing IR sequence.Xiaowei Wu Yan Liang Hengbin Gao Jiyao Wang Yan Zhao Lekai Hua Yundong Yuan Ahong Wang Xiaohui Zhang Jiafan Liu Jie Zhou Xiangbing Meng Dahan Zhang Shaoyang Lin Xuehui Huang Bin Han Jiayang Li Yonghong Wang 2021Molecular Plant2021,14,6:3
4Schisandrol A protects AGEs-induced neuronal cells death by allosterically targeting ATP6V0d1 subunit of V-ATPase显示文摘Diabetes have been shown to cause progressive neuronal injury with pain and numbness via advanced glycation end-products(AGEs)-induced neuronal cell apoptosis;however, the valuable drug targets for diabetic neuropathy have been poorly reported so far. In this study, we discovered a natural small-molecule schisandrol A(SolA) with significant protective effect against AGEs-induced neuronal cell apoptosis. ATP6V0D1, a major subunit of vacuolar-type ATPase(V-ATPase) in lysosome was identified as a crucial cellular target of SolA. Moreover, SolA allosterically mediated ATP6V0D1 conformation via targeting a unique cysteine 335 residue to activate V-ATPase-dependent lysosomal acidification.Interestingly, SolA-induced lysosome pH downregulation resulted in a mitochondrial-lysosomal crosstalk by selectively promoting mitochondrial BH3-only protein BIM degradation, thereby preserving mitochondrial homeostasis and neuronal cells survival. 5Biosynthesis of clinically used antibiotic fusidic acid and identification of two short-chain dehydrogenase/reductases with converse stereoselectivity显示文摘Fusidic acid is the only fusidane-type antibiotic that has been clinically used. However,biosynthesis of this important molecule in fungi is poorly understood. We have recently elucidated the biosynthesis of fusidane-type antibiotic helvolic acid, which provides us with clues to identify a possible gene cluster for fusidic acid(fus cluster). This gene cluster consists of eight genes, among which six are conserved in the helvolic acid gene cluster except fusC1 and fusB1. Introduction of the two genes into the Aspergillus oryzae NSAR1 expressing the conserved six genes led to the production of fusidic acid. A stepwise introduction of fusC1 and fusB1 revealed that the two genes worked independently without a strict reaction order. Notably, we identified two short-chain dehydrogenase/reductase genes fusC1 and fusC2 in the fus cluster, which showed converse stereoselectivity in 3-ketoreduction. 6Improving Low Bitrate Video Coding via Computation Incorporating A Priori Information显示文摘The growing number of mobile users, as well as the diversification in types of services have resulted in increasing demands for wireless network bandwidth in recent years. Although evolving transmission techniques are able to enlarge the network capacity to some degree, they still cannot satisfy the requirements of mobile users. Meanwhile, following Moore's Law, the data processing capabilities of mobile user terminals are continuously improving. In this paper, we explore possible methods of trading strong computational power at wireless terminals for transmission efficiency of communications. Taking the specific scenario of wireless video conversation, we propose a model-based video coding scheme by learning the structures in multimedia contents. Benefiting from both strong computing capability and pre-learned model priors, only low-dimensional parameters need to be transmitted; and the intact multimedia contents can also be reconstructed at the receivers in real-time. 7Comprehensive analysis of polarimetric radar cross-section parameters for insect body width and length estimation显示文摘The length and width of insect body are critical parameters for entomological radar species identification. However, the body width is not measurable in entomological radar currently. In this study,the scattering matrices(SM) of 159 insect specimens were measured using an X-band fully polarimetric laboratory rig in a microwave anechoic chamber. The relationships between the polarimetric radar crosssection(RCS) parameters extracted from the SM and the insect body width were studied. It was found that all these parameters have good correlations with the body width and can be used to estimate it. The mopping relationships between the polarimetric RCS parameters and the body width are built, and can be used as empirical formulas for insect body width estimation. In addition, based on these parameters, two new body length estimation methods were proposed. The performance of the proposed body width and length estimation methods when the echo signals were noisy was analyzed. 8Numerical reconstruction of microstructure of graphite anode of lithium-ion battery显示文摘Due to the presence of graphite flake cascades,the real graphite anode of Li-ion battery shows non-isotropic characteristic. The present work developed an ellipsoid-based simulated annealing method and numerically reconstructed the three-dimensional microstructure of a graphite anode. The reconstructed anode is a composite of three clearly distinguished phases: pore(or electrolyte),graphite, and solid additives, well representing the nonisotropic heterogeneous characteristic of real graphite anode. Characterization analysis of the reconstructed electrode gives information such as the connectivity of individual phase, the specific interfacial area between solid and pore phase, and the pore size distribution. The effects of the ellipsoid size on the structural characteristics of graphite anode were particularly studied.
